#1 - Reproductive biology, morphology, and morphometry of ovaries and oviducts of Trachemys scripta elegans in Brazilian Cerrado

Abstract in English:

Although Trachemys scripta elegans is an exotic species popular as a pet in Brazil, studies on reproductive biology and capacity are non-existent in the Brazilian Cerrado. This study analyzed ovarian and oviduct characteristics and the egg production capacity of T. scripta elegans grown in this biome. The findings will associate with the size of the specimens and the sexual maturity, aiming at comparisons with native and exotic populations, as well as interspecific and contributing to the understanding of its impact on the invaded ecosystems and the establishment of eradication programs. Thus, 39 females had evaluated the body biometry and the morphology and morphometry of the ovaries and oviducts. G2 (N=20): with Class I (>5-10mm) follicles, with Class I and Class II (>10-fold) follicles, 25mm) and G3 (N=9) with Class I, Class II and Class III (>25mm) follicles. Analysis of variance, Scott-Knott’s test, and Pearson’s correlation analysis showed that there was no significant difference between the groups in body biometry; in the mean gonadosomatic index and gonadal morphometry, only the width of the oviducts in the right antimer and the mass and width in the left antimer were higher in G3, the only one that presented eggs. There was positive and harmonic development between body mass, carapace, and plastron, and gonadal growth occurred concomitantly with body growth, indicating a higher reproductive potential and a positive relationship between the size of the litter and the female litter. The gonadosomatic index proved to be an excellent reproductive indicator, and the ovarian evaluation was a better indicator of sexual maturity than the maximum carapace length. Ovaries were irregular structures, without delimitation between the cortical and medullary regions and filled with vitelogenic follicles of different diameters, atresic follicles, and corpora lutea, which reflected the ovarian complexity of the species and the presence of follicular hierarchy. In the scarce stroma, two germinative beds were observed per ovary and the presence of gaps very close to the follicles and associated with the blood vessels. Analysis of gonadal tissue revealed three types of oocytes according to cytoplasmic characteristics: homogeneous, vesicular or vesicular in the cortex with apparent granules. Oviducts were functional and separated, joining only in the final portion to form the cloaca and subdivided into infundibulum, tuba, isthmus, uterus, and vagina. The structure of the uterine tube was composed of serosa, muscular and mucous, which was full of glands. The presence of eggs in the oviducts indicated that the specimens can reproduce in the Brazilian Cerrado. This study provides necessary and relevant information on the reproductive biology and capacity of T. scripta elegans in the Brazilian Cerrado and can contribute to the understanding of its impact on the invaded ecosystems and the establishment of eradication programs. The extraction of females with capacity can reduce the annual reproductive yield of the species and decrease its effect on local biodiversity.

Embora Trachemys scripta elegans seja uma espécie exótica popular como animal de estimação no Brasil, estudos sobre biologia e capacidade reprodutivas são inexistentes no Cerrado brasileiro. Este estudo analisou características ovarianas e do oviduto e a capacidade de produção de ovos em T. scripta elegans criadas neste bioma, correlacionando estes achados ao tamanho dos espécimes e a maturidade sexual, visando comparações com populações nativas e exóticas, bem como interespecíficas e contribuir para a compreensão de seu impacto nos ecossistemas invadidos e com o estabelecimento de programas de erradicação. Assim, 39 fêmeas tiveram avaliadas a biometria corporal e a morfologia e morfometria dos ovários e ovidutos. De acordo com o tamanho dos folículos ovarianos as fêmeas foram separadas em G1 (N= 10): com folículos Classe I (>5-10 mm), G2 (N= 20): com folículos Classe I e Classe II (>10-25 mm) e G3 (N= 9) com folículos Classe I, Classe II e Classe III (>25 mm). À análise de variância, teste de Scott-Knott e à análise de correlação de Pearson verificou-se que não houve diferença significativa entre os grupos na biometria corporal; no índice gonadossomático médio e na morfometria gonadal, apenas a largura dos ovidutos no antímero direito e a massa e a largura no antímero esquerdo foram maiores no G3, o único que apresentou ovos. Houve desenvolvimento positivo e harmônico entre massa corporal, carapaça e plastrão e o crescimento gonadal ocorreu concomitante ao crescimento corporal, indicando maior potencial reprodutivo e relação positiva entre o tamanho da ninhada de ovos e o da fêmea. O índice gonadossomático mostrou-se um bom indicador reprodutivo e a avaliação ovariana um melhor indicador da maturidade sexual que o comprimento máximo da carapaça. Ovários foram estruturas irregulares, sem delimitação entre a região cortical e medular e repletos de folículos vitelogênicos de diferentes diâmetros, folículos atrésicos e corpos lúteos, que refletiram a complexidade ovariana da espécie e a presença de hierarquia folicular. No estroma escasso foram observados dois leitos germinativos por ovário e a presença de lacunas muito próximas aos folículos e associadas aos vasos sanguíneos. A análise do tecido gonadal revelou três tipos de oócitos de acordo com as características do citoplasma: homogêneo, vesicular ou vesicular no córtex com grânulos aparentes. Ovidutos eram funcionais e separados, unindo-se apenas na porção final para formar a cloaca e subdividiam-se em infundíbulo, tuba uterina, istmo, útero e vagina. A estrutura da tuba uterina era constituída de serosa, muscular e mucosa, a qual era repleta de glândulas. A presença de ovos nos ovidutos indicou que os espécimes podem se reproduzir no cerrado brasileiro. Este estudo fornece informações básicas e relevantes da biologia e capacidade reprodutivas de T. scripta elegans no Cerrado brasileiro e pode contribuir com a compreensão de seu impacto nos ecossistemas invadidos e com o estabelecimento de programas de erradicação, uma vez que a extração de fêmeas com capacidade reprodutiva pode contribuir com a diminuição do rendimento reprodutivo anual da espécie e diminuir seu efeito sobre a biodiversidade local.


#2 - Morphological description of ovary and uterus of the nurse shark (Ginglymostoma cirratum) caught off at the Fortaleza coast, Northeast Brazil

Abstract in English:

The nurse shark, Ginglymostoma cirratum (Bonnaterre, 1778) is one of the most studied species of elasmobranchs. However, the knowledge of their reproductive biology is still relatively rare, particularly in the western South Atlantic. This study aimed to describe the morphology of the uterus and the ovary of G. cirratum, based on specimens caught off at the Fortaleza/CE coast, northeast Brazil. Samples were collected from September 2012 to June 2013, from regular landings of artisanal fishing, which commercialize this species freely. A total of ten females were collected. The methodologies followed for analyzing the ovaries and uterus of those females included both macroscopic and histological analysis. G. cirratum has internal type ovary morphology, with invaginations of connective tissue, which defines compartments and separate oocyte groups in ovigerous lots. The epithelium lining the ovary changes from simple columnar ciliated in the area without ovigerous lots, which turns into a simple cubic epithelium in the coating portion of the epigonal organ where ovarian tissue is absent. The uterine mucosa has secretory cells denoted by Alcian Blue staining, indicating the production of mucopolysaccharides, even in immature individuals. This lecithotrophic shark has a uterine vascularized mucosa that is one characteristic of viviparous elasmobranch species.

O tubarão-lixa, Ginglymostoma cirratum (Bonnaterre, 1778) é uma das espécies mais estudadas de elasmobrânquios. O conhecimento de sua biologia reprodutiva, no entanto, ainda é relativamente raro, particularmente no Oeste do Atlântico Sul. Este estudo objetivou a descrição morfológica do útero e ovário de G. cirratum, baseado em espécimes capturados na costa de Fortaleza/CE, no Nordeste do Brasil. Um total de dez fêmeas foi coletado de setembro de 2012 a junho de 2013, nos desembarques regulares da pesca artesanal, que comercializam essa espécie livremente. A metodologia foi de acordo com as analises macroscópica e histológica dos ovários e útero. G. cirratum tem o tipo de morfologia de ovário interno, com invaginações de tecido conjuntivo, que define compartimentos e separa grupos de oócitos em lotes ovígeros. O epitélio que reveste o ovário varia de epitélio simples colunar ciliado na área que não apresenta lotes ovígeros para epitélio simples cúbico na porção de revestimento do órgão epigonal, onde o tecido ovariano é ausente. A mucosa uterina possui células secretoras, evidenciadas pela coloração em Alcian Blue, indicando a produção de mucopolissacarídeos, mesmo em indivíduos imaturos. Este tubarão lecitotrófico apresenta a mucosa uterina vascularizada, sendo esta uma característica de espécies de elasmobrânquios vivíparos.


#3 - Interval in the replacement of in vitro culture medium affects the integrity and development of equine preantral follicles

Abstract in English:

The efficiency of a culture system is related to the elaboration and replacement of a medium with conditions suitable for follicular development. Recent investigations suggested that in vitro culture medium should be replaced after specific time periods in various species. However, the suitable interval for the exchange of in vitro culture medium has not yet been established in equine species. The objective of this investigation was to evaluate the effect of medium exchange intervals of 24 hours (T24) or 48 hours (T48) for in vitro culture of preantral follicles at 2 or 6 days. At the end of the culture period, the fragments were processed using classical histology. Equine preantral follicles were classified according to morphological integrity and developmental stage. Data analysis was performed using Fisher’s test with a significance level of p<0.05. Out of a total of 399 follicles evaluated, 174 (43.6%) were primordial follicles, 225 (56.4%) were in development, and 63.76% were morphologically intact. In the in vitro culture performed over two days, there was no significant difference in relation to follicular integrity after medium replacement (p>0.05). Compared to the medium replacement at six days of culture, there was a statistically significant difference for T24 (68.9%, p<0.05). Therefore, we suggest changing the medium for equine species at 48 hours after the start of culture followed by subsequent daily replacements.

A eficiência de um sistema de cultivo está relacionada à elaboração e substituição do meio de cultivo com condições adequadas ao desenvolvimento folicular. Pesquisas recentes sugerem que o meio de cultivo in vitro deve ser substituído após períodos de tempo específicos para várias espécies. No entanto, o intervalo adequado para a troca de meio de cultivo in vitro ainda não foi estabelecido na espécie equina. O objetivo desta investigação foi avaliar o efeito de intervalos de troca média de 24 horas (T24) ou 48 horas (T48) para cultivo de folículos pré-antrais aos 2 ou 6 dias. No final do período de cultivo, os fragmentos foram processados usando histologia clássica. Os folículos pré-antrais equinos foram classificados de acordo com a integridade morfológica e o estágio de desenvolvimento. A análise dos dados foi realizada utilizando o teste de Fisher com um nível de significância de p<0,05. De um total de 399 folículos avaliados, 174 (43,6%) foram folículos primordiais, 225 (56,4%) estavam em desenvolvimento e 63,76% estavam morfologicamente intactos. No cultivo in vitro realizado ao longo de dois dias, não houve diferença significativa em relação à integridade folicular após a substituição do meio (p>0,05). Comparado com a substituição média aos seis dias de cultivo, houve diferença estatisticamente significativa para T24 (68,9%, p<0,05). Portanto, sugerimos alterar o meio para as espécies equinas às 48 horas após o início da cultura, seguindo as subsequentes substituições diárias.


#4 - Immunolocalization of leptin receptor of Spix’s Yellow-toothed Cavy (Galea spixii Wagler, 1831) ovary

Abstract in English:

Leptin, a cytokine produced by adipose cells, is the target of the scientific community for believing that it has an impact on the reproduction of the animals promoting puberty, folliculogenesis and oogenesis, estrous cycle and aiding in fertilization. The understanding of the mechanisms controlling the reproductive activity of Spix’s Yellow-toothed Cavy (Galea spixii) plays a relevant role in the preservation of the species. Thus, the present study proposed to analyze the immunolocalization of leptin receptors (Ob-R) in the ovary of cavies. Ovaries from 20 adult, non-pregnant, healthy females were collected. The samples were fixed in 4% phosphate buffered paraformaldehyde, embedded in paraffin and sectioned for immunohistochemistry. The sections were photomicrographs and intensity of the reaction was measured. Strong immunoreaction was observed in oocyte and theca cells, moderate in ovarian stromal cells and large luteal cells and weak stained in granulosa, endothelial, perivascular and small luteal cells. When compared to receptor expression along follicular development it was observed that the oocyte and the theca cells remained with expression at the same intensity. However, the granulosa cells presented strong stained in the preantral stages, whereas in the antral follicles it presented low intensity. We conclude that in the ovaries of Galea spixii there is the presence of Ob-R in the main structures of the ovary sugesting that this hormone plays a fundamental role in the reproduction of this species

A leptina, uma citocina produzida pelas células adiposas, é alvo da comunidade científica por acreditarem que ela apresente impacto sobre a reprodução dos animais promovendo a puberdade, foliculogênese e oogênese, ciclo estral e auxiliando na fecundação. A compreensão dos mecanismos que controlam a atividade reprodutiva de preás (Galea spixii) possui papel relevante para a preservação da espécie. Desta forma, o presente trabalho propôs analisar a imunolocalização dos receptores de leptina (Ob-R) no ovário de preás. Coletaram-se os ovários de 20 fêmeas adultas, não prenhes e saudáveis. As amostras foram fixadas em paraformaldeído a 4% em tampão fosfato, incluídas em parafina e seccionadas para a realização de imunohistoquímica (IHC). As secções foram fotomicrografadas e avaliadas quanto à intensidade da reação. Observou-se forte imunorreação no oócito e nas células da teca, moderada nas células do estroma ovariano e nas células luteínicas grandes e fracamente coradas nas células da granulosa, endoteliais, perivasculares e células luteínicas pequenas. Quando comparado a expressão de receptores ao longo do desenvolvimento folicular foi observado que o oócito e as células da teca se mantiveram com expressão na mesma intensidade. Entretanto, as células da granulosa apresentaram forte marcação nos estádios pré-antrais enquanto que nos folículos antrais apresentou fraca intensidade. Concluímos que em ovários de Galea spixii existe a presença de Ob-R nas principais estruturas do ovário sugerindo que este hormônio desempenhe papel fundamental na reprodução desta espécie.


#5 - Analysis of the reproductive stage and ovarian histomorphometry of Dermanura cinerea (Chiroptera: Phyllostomidae) in an Atlantic Forest fragment of Pernambuco, northeastern Brazil

Abstract in English:

This study aimed to analyze the reproductive stage, histology and morphometry of the ovary of Dermanura cinerea in an Atlantic Forest fragments in the Biological Reserve of Saltinho, Pernambuco, Brazil. Adult females were captured monthly by mist net, during two consecutive nights from June/2014 to November/2015. The meteorological data were provided by the National Institute of Meteorology and grouped together with the reproductive data, in six periods: period I (June to August/2014), period II (September to November/2014), period III (December/2014 to February/2015), period IV (March to May/2015), period V (June to August/2015) and period VI (September to November/2015). The females were classified according to their reproductive stages in: Inactive, pregnant, lactating and postlactating. Among the 25 females caught, 10 inactive females were selected for the histomorphometric analysis of the ovaries and then they were euthanized. After that, the specimens had their ovaries removed, fixed, processed and included in Paraplast. The organs were cut and stained by Hematoxylin – Eosin and the histological slides produced were submitted to the appropriate optical microscope analysis. Each type of ovarian follicle was estimated according to the histological characteristics evidenced for other mammals. In the morphometric analysis, it was considered: The area of the ovary (measured in square millimeter), Quantification of different types of ovarian follicles (unilaminar and multilaminar primary follicles, antral and matures) and corpus luteum, as well as the area of each by those different follicles (measured in square micrometer) The results indicated that 16 females were inactive, 5 were lactating, 4 were pregnant and no post-lactating. The histological analysis revealed that the ovaries of D. cinerea exhibit characteristics, structures and cellular types similar to other mammals. The morphometric data of the study area were statistically evaluated using the Student’s t-test. Therefore, ovarian histomorphometric analysis are very important to infer with greater certainty about the reproduction of a species. Thus, it was observed that females of D. cinerea exhibit two reproductive peaks, that is, they can give birth either at the end of dry periods or from the beginning to the middle of the rainy season. The main feature observed in the ovary of this species, found in the Atlantic Forest area of the state of Pernambuco, is the preparation of the ovarian cortex to ovulate in months of high rainfall throughout the year. Therefore, this species tries to adjust its reproductive period with the variations of the environmental conditions, so that the reproduction might occur during the period more favorable for the maintenance of their offspring.

Esse estudo objetivou analisar o estágio reprodutivo, histologia e morfometria do ovário de Dermanura cinerea em um fragmento de Mata Atlântica da Reserva Biológica de Saltinho (Pernambuco, Brasil). Foram utilizadas fêmeas adultas capturadas por redes de neblina, de modo que as coletas ocorreram mensalmente, durante duas noites consecutivas de Junho/2014 a Novembro/2015. Os dados meteorológicos foram fornecidos pelo Instituto Nacional de Meteorologia e agrupados, juntamente com os dados reprodutivos, em seis períodos, sendo estes: período I (Junho a agosto/2014), período II (Setembro a novembro/2014), período III (Dezembro/2014 a fevereiro/2015) período IV (Março a maio/2015), período V (Junho a agosto/2015) e período VI (Setembro a novembro/2015). As fêmeas foram classificadas, de acordo com o estágio reprodutivo em: inativas, grávidas, lactantes e pós-lactantes. Das 25 fêmeas capturadas, foram selecionadas, para as análises histomorfométricas dos ovários, 10 fêmeas inativas, as quais foram eutanasiadas. Depois disso, os espécimes tiveram os seus ovários removidos, fixados, processados e inclusos em paraplast. Os cortes foram corados por Hematoxilina – Eosina e as lâminas histológicas produzidas foram submetidas às devidas análises em microscópio óptico. Cada tipo de folículo ovariano foi estimado de acordo com as características histológicas evidenciadas para outros mamíferos. Nas análises morfométricas, considerou-se: a área do ovário (mensurada em milímetro quadrado), quantificação dos diferentes tipos de folículos ovarianos (primários unilaminar e multilaminar, antrais e maduros) e corpo lúteo, bem como foi medida a área de cada um desses diferentes tipos de folículos (mensurada em micrômetro quadrado). Os resultados indicaram a presença de 16 fêmeas inativas, 5 lactantes, 4 grávidas e nenhuma pós-lactante. As análises histológicas destacaram que os ovários de D. cinerea possui características, estruturas e tipos celulares padrões semelhantes ao dos demais mamíferos. Os dados morfométricos da área do ovário foram avaliados estatisticamente por meio do teste t de Student. Assim, as análises histomorfométricas ovariana são de suma importância para inferir com maior segurança sobre a reprodução de uma dada espécie. Portanto, ficou constatado que as fêmeas de D. cinerea apresentam dois picos reprodutivos, isto é, podem parir no final de períodos secos ou do início para metade do período chuvoso. A principal característica observada no ovário dessa espécie, encontrada em área de Mata Atlântica do estado de Pernambuco, foi a preparação do córtex ovariano para ovular em meses de elevados índices pluviométricos ao longo do ano. Logo, essa espécie procura ajustar seu período reprodutivo com as variações das condições ambientais, de forma que a reprodução ocorra em períodos mais favoráveis para manutenção da prole.


#6 - Morphology of the female genital organs of freshwater turtle Mesoclemmys vanderhaegei, 37(9):1015-1024

E-mail: walkiria.ferreira@gmail.com Mesoclemmys vanderhaegei (Testudines: Chelidae) is a freshwater turtle with occurrence in Amazon, Tocantins, Paraguai, Paraná and Uruguai rivers basins. Although according to International Union for Conservation of Nature, it has low risk of extinction, there is an uptade necessity of ecological and biological data. Considering that the management and conservation plans are related to a wide knowledge of reproductive biology, a first macroscopic description about the young and adults females of M. vanderhaegei is important. These points were correlated to the specimens size and period of the year. The samples of M. vanderhaegei were collected in Chapada dos Guimarães county, Mato Grosso, Brazil, an area of large natural occurrence of the specimens. Genital organs of seventeen females were fixed in 10% formalin and then dissected to demonstrate the particularities related to external and internal anatomy. The young and adult M. vanderhaegei genitals organs are composed of ovaries and oviducts pairs that dorsolaterally discharge into the cloaca, forming with the ureter, the urogenital papilla. The ovaries are elongated organs with larger cranial and elongated caudal portions. The oviducts, which are in adults very differentiated in its shape and size compared to the young, are long and located laterally to the ovaries. In both age groups, the genital organs are supported by celomatic membrane folds that emerge from the ceiling of the cavity, constituting the mesovary and mesoviduct. In adult females, according to the shape and pattern of the oviduct mucosa, the cranial segment corresponds to the regions of the infundibulum and magnum; the middle segment, the isthmus and the caudal segment identify the uterus and vagina regions. The clitoris is sited on the floor of the urodeum. The carapace linear length and body mass between immature and adult females vary to 134-155.6mm e 134.43-365g respectively. The main part of young females was captured in rainy period and the adults, with and without eggs, at dry period. The macroscopic characteristics of the M. vanderhaegei genital organs are also observed in others Testudines, with the exception of urogenital papilla and by the clitoris presence.

Consta na lista vermelha de espécies ameaçadas da International Union for Conservation of Nature, como espécie de baixo risco, mas que poderá se tornar ameaçada, sendo necessário a atualização de seus dados ecológicos e biológicos. Considerando que planos de manejo e conservação de espécies dependem também de vasto conhecimento sobre a biologia reprodutiva, apresentamos a primeira descrição macroscópica dos órgãos genitais de fêmeas jovens e adultas de M. vanderhaegei correlacionando esses achados ao tamanho do espécime e ao período do ano. Exemplares de M. vanderhaegei foram coletados no município de Chapada dos Guimarães, área de ampla ocorrência natural da espécie. A descrição dos órgãos genitais foi realizada a partir de 17 fêmeas fixadas em formol a 10% e dissecadas para evidenciação de particularidades relacionadas à anatomia externa e interna. Os órgãos genitais M. vanderhaegei jovens e adultas são constituídos por pares de ovários e ovidutos que desembocam dorsolateralmente na cloaca, formando junto com o ureter a papila urogenital. Os ovários são órgãos alongados, com a extremidade cranial mais larga e caudal afunilada, já os ovidutos são longos e localizados lateralmente aos ovários, sendo em adultas, muito diferenciado na sua forma e tamanho em relação ao das jovens. Em ambas faixas etárias, os órgãos genitais são sustentados por pregas de membrana celomática que emergem do teto da cavidade, constituindo os mesovário e mesoviduto. De acordo com a forma e o padrão de mucosa do oviduto em fêmeas adultas, o segmento cranial corresponde as regiões do infundíbulo e magnum, o segmento médio, ao istmo e no caudal identificam-se as regiões útero e vagina. No assoalho do urodeum aloja-se o clitóris. O comprimento retilíneo de carapaça e massa corpórea entre fêmeas imaturas e adultas variam, respectivamente, entre 134-155,6mm e 134,43-365g. A maioria das fêmeas jovens foram capturadas no período chuvoso e as adultas sem e com ovos no período seco. As características macroscópicas dos órgãos genitais observadas em M. vanderhaegei são compartilhadas com outros Testudines, exceto pela papila urogenital e presença do clitóris.

E-mail: murielpimentel@yahoo.com.br This study aimed to evaluate the characteristics of two different murine models of hormone-treated renal-encapsulated bovine ovarian tissue xenotransplantation. Two immunodeficient mouse models (BALB/c Nude and C57BL6 SCID) were xenografted with ovarian pieces from heifers and each group was subjected to two hormonal treatments of eCG or a combination of FSH+LH. Donor ovaries and recipients were evaluated by histology and infrared thermography at different times. At the time of xenograft collection, animals were evaluated for alterations in hepatorenal biochemistry. The statistical test used in the study was ANOVA, followed by Tukey’s test. Among the strains, 80% of C57BL6 SCID and 77% of BALB/c Nude mice showed development and vascularization of the transplanted tissue, which acquired cyclicity at 19 and 9 days post-transplant, respectively. Hemorrhagic follicles in xenografts induced with FSH+LH were found in the C57BL6 SCID strain. Infrared thermography was insufficient to distinguish the tissue donor recipient. In conclusion, the C57BL6 SCID strain appears to be the best host for ovarian xenografts, since the transplants in these mice were viable and showed robust follicular development. This work will aid future choices of immunodeficient strains for xenografting procedures.

Dois modelos de camundongos imunodeficientes (BALB/c NUDE e C57BL6 SCID) receberam fragmentos de ovário de novilhas e cada grupo foi submetido a dois tratamentos hormonais de eCG ou uma combinação de FSH+LH. Ovários doadores e receptores foram avaliados por histologia e termografia infravermelha em diferentes momentos. No momento da retirada do xenotranplante, os animais foram avaliados quanto a alterações na bioquímica hepatorrenal. O teste estatístico utilizado no estudo foi ANOVA, seguido do teste de Tukey. Entre as linhagens, 80% de C57BL6 SCID e 77% das BALB/c NUDE mostraram desenvolvimento e vascularização do tecido transplantado, que adquiriu a ciclicidade 19 e 9 dias após o transplante, respectivamente. Corpos hemorrágicos foram encontrados após o xenotransplante induzidos com FSH+LH na linhagem C57BL6 SCID. A termografia infravermelha foi insuficiente para distinguir o tecido doador do receptor. Em conclusão, a linhagem C57BL6 SCID demonstrou ser o melhor hospedeiro para xenotransplante de ovário, uma vez que os transplantes nestes camundongos foram viáveis e mostraram desenvolvimento folicular. Este trabalho ajudará futuras escolhas de linhagens imunodeficientes para procedimentos de xenotranplante.

E-mail: corcinicd@gmail.com The study of embryological development of reproductive organs is an important tool for understanding the particularities of the equine species. Also it is important for increasing knowledge about the development of antral follicles that can be used for in vitro production of embryos. This study aimed to describe the morphological changes of fetal equine ovaries occurring during gestational development. It was used equine fetuses from a slaughterhouse. After slaughter, it was performed the measurement of cefalococcígea distance to calculate the gestational age (days of pregnancy - DP). Afterwards, the ovaries were grossly evaluated, removed and fixed. Then, they were histologically assessed by hematoxylin-eosin and PAS techniques. It was used 19 fetuses aged 50 to 269 days of gestation, distributed in 9 groups divided according to DP. In the grossly evaluation, it was observed differences between the volume of the ovaries according to gestational age, being observed bigger/larger ovarian volume between 210 days to 269 DP. During the histological evaluation, it was observed a simple cuboidal epithelium and a distinction between the cortical and medullary layers in fetal ovaries from 50-89 DP. The main characteristics identified were: the morphological distinction between the cortical and medullary layers in equine fetal ovaries; the ovigerous cords emerged in the interval between 150 and 179 DP; the cortex initiated as a thin layer with an increased amount of connective tissue rich in blood vessels by the 210 DP, structural uniformity acquired in the medular region by the 150 DP, showing similar cells in size and blood vessels of larger caliber. From this stage, the cells in this region had decreased in size and dense connective tissue bundles, which made this zone partially divided.

O presente trabalho teve como objetivo descrever as alterações morfológicas de ovários fetais equinos ocorrentes durante o desenvolvimento gestacional. Foram utilizados fetos equinos provenientes de abatedouro. Imediatamente após o abate foi realizada medição da distância cefalococcígea para o cálculo da idade gestacional em dias (dias de gestação - DG) e dissecação dos fetos para a retirada e fixação dos ovários. Foram realizadas avaliações macroscópicas e histológicas pelas técnicas de Hematoxilina-eosina e PAS. Foram utilizados 19 fetos com idade de 50-269 dias de gestação, distribuídos em 9 grupos de DG. Na avaliação macroscópica foi observada diferença entre o volume dos ovários de acordo com a idade gestacional, sendo observado maior volume ovariano entre os dias 210 a 269 de desenvolvimento gestacional. Na avaliação histológica foi observado epitélio de revestimento cúbico e distinção entre as camadas cortical e medular nos ovários fetais a partir de 50-89 DG. As principais características identificadas foram à distinção morfológica entre as camadas cortical e medular nos ovários fetais equinos: os cordões ovígeros surgiram no intervalo de 150-179 DG; o córtex iniciada como uma camada delgada e por volta do 210º dia de gestação apresentou espessamento pelo aumento da quantidade de tecido conjuntivo rico em vasos sanguíneos, o qual se manteve até os estágios finais da gestação; a região medular adquiriu uniformidade estrutural por volta do 150º dia, mostrando células similares em tamanho e vasos sanguíneos de calibre maior. A partir desta etapa, as células medulares diminuíram em tamanho e feixes de tecido conjuntivo denso tornaram esta zona parcialmente dividida.

E-mail: nataliaocarino@gmail.com Morphological and immunohistochemical characterization of angiogenic and apoptotic factors and the expression of thyroid receptors in the ovary of tilapia Oreochromis niloticus in captivity were studied. The morphological evaluation of the ovaries was performed by histological paraffin embedded and stained with HE. The immunohistochemical expressions of CDC47, VEGF, Flk-1, angiopoietin, Tie-2 and thyroid receptor (TR&#945;) were performed by the technique of streptavidein-biotin-peroxidase. Apoptosis was assessed using the TUNEL kit. The relative expression of thyroid hormone receptors (TR&#945; and TR&#946;) was assessed by RT-PCR real time. The nuclear expression of CDC47 increased with the stage of maturation of the oocyte and was observed in the follicle cells. Apoptotic bodies were observed in the follicular cells of atretic follicles and postovulatory follicles from the ovaries of 150g and 350g fish. Expression of VEGF and its receptor Flk-1 was also observed in the follicular cells, and the expression of both increased with the maturity of the oocyte, with a higher intensity observed in the full-grown follicle. The expression of angiopoietin and of its receptor (Tie 2) was discrete and moderate respectively. TR&#945; expression was independent of follicular development. However, the 350 g tilapia exhibited higher expression of TR&#946; compared with the 50 g tilapia. We conclude that the proliferative activity and the expression of VEGF and its receptor increase with follicular maturation and that the TRs expression increases with ovarian maturity in tilapia (Oreochromis niloticus).

A avaliação morfológica dos ovários foi realizada por cortes histológicos incluídos em parafina e corados por HE. As expressões imuno-histoquímicas de CDC47, VEGF e seu receptor Flk-1, angiopoetina e seu receptor Tie-2 e recertor tireoidiano (TR&#945;) foram realizadas pela técnica de estreptavideina-biotina-peroxidade. A apoptose foi avaliada utilizando-se kit de TUNEL. A expressão relativa dos receptores de hormônios tireoidianos (TR&#945; e TR&#946;) foi avaliada pela técnica de RT-PCR tempo real. A expressão nuclear de CDC47 aumentou com a fase de maturação do oócito e foi observada nas células foliculares. Corpos apoptóticos foram observados nas células foliculares de folículos atrésicos e folículos pós-ovulatórios de ovários de peixes com 150g e 350g. A expressão de VEGF e do seu receptor Flk-1 foi também observada nas células foliculares , e a expressão de ambos aumentou com a maturidade do oócito , com uma maior intensidade no folículo maduro. A expressão de angiopoietina e do seu receptor (Tie 2) foi discreta e moderada, respectivamente. A expressão de TR&#945; foi independente do desenvolvimento folicular. No entanto, a tilápia de 350g apresentou maior expressão de TR&#946; em comparação com a tilápia de 50g. Conclui-se que a atividade proliferativa e a expressão de VEGF e de seu receptor aumenta com a maturação folicular e que a expressão dos TRs aumenta com a maturidade do ovário em tilápia (Oreochromis niloticus).

E-mail: sanelylc@hotmail.com The aim of this study was to investigate the effects of the insulin-like growth factor -I (IGF-I) on survival, activation (transition from primordial to primary follicles) and growth of caprine preantral follicles cultured in vitro. Fragments of ovarian cortex were cultured for one and seven days in the absence or presence of IGF-I (0, 50 and 100ng/ml). The non-cultured and cultured tissues were processed and analyzed by histology and transmission electron microscopy. The culture for one day in a medium with 100ng/ml of IGF-I showed 86.7% of morphologically normal follicles. These results were similar (P>0.05) to the percentage of normal follicles found in the control (96.7%). It was also found that this medium increased the percentage of follicular activation (developing follicles) with one day of culture. The oocyte and follicular diameters remained similar to the control by culturing for one day in a medium containing 100ng/ml of IGF-I. The ultrastructural analysis did not confirm the integrity of the follicular fragments in a medium containing IGF-I (100ng/ml) after one and seven days of culture. In conclusion, this study demonstrated that the addition of 100 ng/ml of IGF-I in the culture medium enables the development of preantral follicles of goats with one day of culture. However, it is not sufficient to maintain the follicular integrity and the follicular survival rate after seven days of culture.

Os tecidos não cultivados e cultivados foram processados e analisados por histologia e microscopia eletrônica de transmissão. O cultivo por um dia em meio com 100ng/mL de IGF-I apresentou 86,7% de folículos morfologicamente normais. Estes resultados foram semelhantes (P>0,05) ao percentual de folículos normais encontrados no controle (96,7%). Verificou-se ainda que este meio aumentou o percentual de ativação folicular (folículos em desenvolvimento) com um dia de cultivo. Os diâmetros ovocitário e folicular mantiveram-se semelhantes ao controle ao cultivar por um dia em meio contendo 100ng/mL de IGF-I. As análises ultraestruturais não confirmaram a integridade folicular dos fragmentos em meio contendo IGF-I (100 ng/mL) após um e sete dias de cultivo. Em conclusão, esse estudo demonstrou que a adição de 100 ng/mL de IGF-I no meio de cultivo ativa o desenvolvimento de folículos pré-antrais de caprinos com um dia de cultivo. Entretanto, não é suficiente para manter a integridade folicular e a taxa de sobrevivência folicular após sete dias de cultivo.
