E-mail: isabellecpires@hotmail.com Bacteria of the genus Aeromonas are highly disseminated pathogens in the aquatic environment, responsible for great economic losses in the pisciculture of several countries. They are opportunistic microorganisms and their pathogenicity is linked to some virulence factors, such as biofilm formation. Saline stress is one of the factors that favor the formation of these colonies and, consequently, the increase of infections. These infections, when associated with biofilm, are even more resistant to antimicrobials. In this context, polypyrrole stands out as an antimicrobial alternative because it has several therapeutic attributes and does not present toxicity to organisms. Thus, the objective of this study was to evaluate the susceptibility profile and the biofilm formation capacity of Aeromonas spp. associated with different levels of salinity and polypyrrole. The antibacterial activity of the isolates was determined and motility assays were performed with bacteria bearing the fla gene. The ability of sodium chloride and polypyrrole to interfere with biofilm formation has also been demonstrated. The results were evidenced with scanning electron microscopy. Concentrations of 2 and 3% of NaCl inhibited bacterial motility. In the biofilm formation, 83% of the bacterial isolates induced production at the concentration of 0.25%. Polypyrrole caused the death of all the isolates tested at the concentration of 125μg/mL. In addition, this compound decreased bacterial motility at concentrations of 0.25 to 3%, and no biofilm was produced. These results show that the different levels of NaCl influence in the formation of the biofilm favoring the persistence of the infection. This study also highlighted the potential of polypyrrole as a bactericidal agent, being an effective alternative to antimicrobial drugs for the treatment of infections caused by Aeromonas spp.

São micro-organismos oportunistas e sua patogenicidade está ligada a alguns fatores de virulência, como a formação de biofilme. O estresse salino é um dos fatores que favorecem a formação dessas colônias e, consequentemente, o aumento de infecções. Essas infecções quando estão associadas ao biofilme são ainda mais resistentes aos antimicrobianos. Nesse contexto, o polipirrol destaca-se como uma alternativa antimicrobiana por possuir vários atributos terapêuticos e não apresentar toxicidade aos organismos. Dessa forma, o objetivo desse estudo foi avaliar o perfil de susceptibilidade e a capacidade de formação de biofilme dos isolados de Aeromonas spp. associados aos diferentes níveis de salinidade e polipirrol. Determinou-se a atividade antibacteriana dos isolados e ensaios de motilidade foram realizados com bactérias que carreavam o gene fla. Também verificou-se a capacidade do cloreto de sódio e polipirrol em interferir na formação do biofilme. Os resultados foram evidenciados com a microscopia eletrônica de varredura. As concentrações de 2 e 3% de NaCl inibiram a motilidade bacteriana. Na formação do biofilme, 83% dos isolados bacterianos induziram a produção na concentração de 0,25%. O polipirrol causou a morte de todos os isolados testados na concentração de 125µg/mL. Além disso, esse composto diminuiu a motilidade bacteriana nas concentrações de 0,25 a 3%, sendo que em relação à produção de biofilme, não houve interferência. Esses resultados evidenciam que os diferentes níveis de NaCl influenciam na formação do biofilme favorecendo a persistência da infecção. Este estudo também realçou a potencialidade do polipirrol como agente bactericida, sendo uma alternativa eficaz às drogas antimicrobianas para o tratamento das infecções causadas por Aeromonas spp.


#2 - Cholesterol-loaded cyclodextrin on the cryopreservation of stallion sperm of the Northeastern breed

Abstract in English:

In the study effect of cholesterol was evaluated on the sperm quality of Nordestina stallion breed. Twenty semen samples were used from two stallions, diluted with BotuSemen extender and cholesterol add as follows: control, 0.75mg of cholesterol-loaded cyclodextrin (CLC) and 1.0mg of CLC/120x106 sperm/mL, and incubated for 15 min at 22°C. The samples were diluted 1:5 with lactose-yolk egg extender and cooled to 5°C over two hours, loaded into 0.5mL straws and frozen in static liquid nitrogen vapor before being plunged into nitrogen. Samples were thawed (37°C/30s) and analyzed. The variables were analyzed by ANOVA and means compared by Tukey test (P<0.05). Higher percentages of total and progressive motile was higher for sperm treated with CLC compared to control, and CLC promoted higher percentages (P<0.05) of total and progressive motility for the 3 hours of incubation. The percentage of viability and plasma membrane integrity of spermatozoa were higher (P<0.05) in sperm treated with CLC compared to the control group. The number of spermatozoa reacted to hypoosmotic test was higher (P<0.05) in sperm treated with CLC than control. Addition of CLC in the semen of Nordestina stallion breed improve the sperm quality after cryopreservation.

Nesta pesquisa avaliou-se o efeito do colesterol sobre o sêmen de garanhões da raça Nordestina sobre a qualidade espermática. Vinte ejaculados de dois garanhões foram diluídos com BotuSemen e colesterol carreado pela ciclodextrina (CCC) adicionado no sêmen: controle, 0,75mg de CCC e 1,0mg de CCC/120x106 sptz/mL, e incubado a 26°C/15min. O sêmen foi diluído 1:5 (v/v) com diluente Lactose-gema de ovo e resfriado a 5°C/2h, envasado em palhetas de 0,5mL, e acondicionado sob vapor de nitrogênio líquido, e depois imersos. As amostras foram descongeladas (37 °C/30s) e avaliadas. As variáveis foram avaliadas com ANOVA e teste de Tukey (P<0,05). A motilidade total e progressiva foi maior (P<0,05) no sêmen tratado com CCC comparado as amostras do grupo controle, e CCC promoveu maior percentual (P<0,05) de motilidade total e progressiva durante as 3 horas de incubação. A percentagem de espermatozoides com viabilidade e integridade foi maior (P<0,05) no sêmen tratado com CCC (81,47 e 86,07%) comparado ao controle (72,12 e 70,19%). O número de espermatozoides reativos ao teste hiposmótico foi maior (P<0,05) nas amostras de sêmen tratadas com CCC comparado ao controle. Adição de colesterol no sêmen de garanhões Nordestino melhora a qualidade espermática apos a criopreservação.

There are currently two BBSE systems, those of the Society for Theriogenology (SFT) and the Western Canadian Association of Bovine Practitioners (WCABP). Scrotal circumference (SC), sperm motility (SM) and normal sperm (NS) of 454 bulls aged between 12 and 15 months of a Spanish beef breed were used to compare both systems, and since there is no agreement on that BBSE system must be applied in Spain, a single one was proposed for its consideration. SC was adjusted to 15 months (SC15) and the mean of the BBSE traits was: SC15 (34.2±2.4cm), SM (76.6±14.6%) and NS (76.8±12.3%). In the PROPOSED system, the SM and NS thresholds were those defined by the WCABP system, while the SC15 thresholds were set by combining the SFT threshold and SC15±1SD in order to establish four classification categories, the three proposed by the WCABP system: unsatisfactory, questionable and satisfactory, and other category, called superior, for bulls with SM&#8805;60%, NS&#8805;70% and SC15&#8805;Mean+1SD. The PROPOSED system scored fewer bulls as unsatisfactory than the SFT and the WCABP systems (8.6%, 23.6% and 22.5%, respectively; P<0.01), while the percentage of bulls from worst to best in the other three categories under the PROPOSED system was: 26.0%, 54.2% and 11.2%, respectively. In conclusion, the PROPOSED system gives more emphasis to SC, sets differences between bulls classified as satisfactory by the other systems and can be considered a good system for Spain and for other countries that have no defined their own system.
