#1 - Clinical, histopathological, and hematological changes due to isoimmune thrombocytopenic purpura in piglets

Abstract in English:

Isoimmune thrombocytopenic purpura (ITP) is an immune-mediated disease that causes severe hemorrhagic lesions and high mortality in piglets. The disease can occur early in newborn piglets (EITP) or late in 2- to 3-week old piglets (LITP). In this study, we analysed the clinical, pathological, and hematological aspects of 391 ITP cases (312 with EITP and 79 with LITP). In LIPT cases, morbidity and mortality rates were higher, with rates of 60% (morbidity) and 53% (mortality). The main clinicopathological findings in ITP cases were different patterns of hemorrhages organs and tissues. In EITP, clinical signs were characterized by extensive subcutaneous hemorrhages and death occurred within a few days; however, in LITP, often sudden death occurred. In macroscopic analysis, hemorrhagic diathesis was observed in all affected animals. In EITP, the most severe hemorrhagic lesions were integumentary, mainly in the dermis and epidermis. In LITP, visceral lesions were predominant, mainly in the epicardium and intestines. Microscopic bone marrow analysis revealed mild cellular hyperplasia in EITP and bone marrow aplasia in LITP. hematological analyses revealed leucopenia, thrombocytopenia, and anemia in all ITP-affected animals. However, fostering by a different sow was only efficient in controlling EITP and had little effect in LITP-symptomatic piglets, due to more severe lesions. Further studies on the etiopathogenesis of LITP are required to improve our understanding of this disease form.

Púrpura trombocitopênica isoimune (PTI) é uma doença imunomediada que causa lesões hemorrágicas graves e alta mortalidade em leitões, que pode se apresentar através de uma forma precoce em leitões neonatos (PTIP) ou uma forma tardia em leitões com duas a três semanas de idade (PTIT). Neste trabalho analisamos aspectos clínicos, hematológicos e histopatológicos de 391 casos de PTI, sendo 312 de PTIP e 79 de PTIT. Observou-se maiores morbidade (60%) e mortalidade (53%) na PTIT. Os principais achados clínico-patológicos observado na PTI são hemorragias em diferentes graus de intensidade e nos diferentes órgãos e tecidos. Na PTIP observou-se predominantemente hemorragias subcutâneas extensas e morte em alguns dias, já na PTIT, observou-se além de grave hemorragia, morte súbita. Na análise macroscópica, observou-se diátese hemorrágica em todos os animais afetados. Na PTIP as lesões hemorrágicas mais graves foram tegumentares, principalmente em derme e epiderme, enquanto, na forma tardia, observou-se lesões predominantemente viscerais, em epicárdico e intestino. A análise microscópica de medula óssea revelou discreta hiperplasia celular na forma PTIP, enquanto, na PTIT observou-se aplasia medular. Na análise hematológica observou-se leucopenia, trombocitopenia e anemia em todos os animais com PTI. Os achados clínicos, histopatológicos e hematológicos para PTIP e PTIT da doença permitiram o diagnóstico de PTI. Entretanto, a troca de mãe se mostrou eficiente apenas para controle PTIP, uma vez que, esta estratégia apresenta pouco resultado para leitões sintomáticos com a PTIT, devido lesões mais severas. Estudos sobre a etiopatogênese da PTIT ainda são necessários para melhor entendimento desta forma da doença.


#2 - Searching bovine papillomavirus presence in lesions seen on teats of cows

Abstract in English:

The samples were taken from 106 cows with various-looking lesions on their teats and ranged in age from 2 to 8 years. Enzyme-linked immunosorbent assay (ELISA) antigen (Ag) positive for the bovine papillomavirus (BPV) was found in 59 (55.7%) blood serum samples. PCR using FAP59/64 primers was positive for 24 (22.6%) samples. BPV-2 (40, 37.7%), BPV-6 (28, 26.4%), BPV-8 (30, 28.3%), BPV-9 (36, 34%), BPV-10 (32, 30.3%), and BPV-12 (22, 20.8%) were found in a PCR type-specific analysis of single and mixed type teat warts. The highest positivity was observed in BPV-2, BPV-9 and BPV-10 in flat and round forms, BPV-6, BPV-10, BPV-12, and mixed types in rice grain-cauliflower forms, BPV-9 and mixed types in filiform in the distribution of types based on the macroscopic appearance of teat lesions. As for the distribution of BPV types according to age, the most BPV-2 types were found in the age group of two years, the most BPV-10 types in the age group of three years, the most BPV-9 types in the age group of four years, the most BPV-8+BPV-12 types in the age group of five years, and the most mixed types between the ages of six and eight years. The existence of the virus was then checked using electron microscopy on the chosen samples (at least one investigation was conducted), and it was positively identified using BPV type-specific primers. The authors concluded that BPV detection using an ELISA (Ag) test from blood serum samples was shown to be less sensitive than BPV type-specific PCR from wart samples.

#3 - Cutaneous pythiosis in equines in the Amazon Biome

Abstract in English:

The study aimed to describe the clinicopathological aspects of 37 cases of pythiosis, 34 in horses and three in mules, from properties located in the Amazon biome of Pará, Brazil. The clinical signs observed in the animals were weakness, poor-to-regular nutritional status, pale mucous membranes, itching at the lesion site, and lameness when the limbs were affected. The lesions were located on the lips, nostrils, rib region, thoracic and abdominal walls, scapular, distal limbs, foreskin, perineum and udder. Macroscopically, ulcerative and granulation-tissue-like masses were observed, with fistulous tracts filled with yellowish and foul-smelling serosanguinous discharges. In the biopsy, it was possible to visualize white and firm areas with foci of yellowish necrotic material and hardened masses, called kunkers. The histopathological examination showed a pyogranulomatous inflammatory reaction with the presence of Pythium insidiosum hyphae, which were impregnated with black, confirming the diagnosis of pythiosis in equids in the Amazon biome, being the first report of the disease in mules in the region.

O trabalho objetivou descrever os aspectos clínico-patológicos de 37 casos suspeitos de pitiose, 34 em equinos e três em muares, provenientes de propriedades localizadas no Pará, bioma amazônico brasileiro. Os sinais clínicos observados nos animais eram caracterizados por debilidade, estado nutricional de ruim a regular, mucosas pálidas, prurido no local da lesão, além de claudicação quando os membros foram acometidos. As lesões eram localizadas nos lábios, narinas, região das costelas, parede torácica e abdominal, escapular, distais dos membros, prepúcio, períneo e úbere. Macroscopicamente observavam-se extensas lesões ulceradas com intensa proliferação de tecido de granulação, de bordos irregulares, com tratos fistulosos, de consistência firme, denominados de “kunkers”, preenchidos com material amarelado e friável, possuindo exsudação serossanguinolenta de odor fétido. Ao exame histopatológico observou-se reação inflamatória piogranulomatosa com presença de hifas de Pythium insidiosum, as quais se impregnaram de negro, confirmando o diagnóstico de pitiose em equídeos no Bioma Amazônico, sendo o primeiro relato da doença em muares na região.


#4 - Detection of paratuberculosis in dairy cows from southern Brazil

Abstract in English:

Bovine paratuberculosis causes chronic, incurable diarrhea and weight loss, resulting in decreased cattle production. The disease is caused by Mycobacterium avium subsp. paratuberculosis (MAP), an obligate intracellular mycobactin-dependent mycobacterium that replicates slowly in the host and has heightened environmental resistance. In countries where the disease is found and the damage has been quantified, direct and indirect economic losses are extremely high. Local epidemiological data is of paramount importance for the implementation of control programs. Our objective was to verify whether paratuberculosis is present in commercial dairy herds in different mesoregions of RS. Therefore, a prospective, cross-sectional and observational study was performed on dairy cattle from five mesoregions of the RS state, Brazil. Milk samples taken from individual cows on commercial farms were tested using indirect ELISA tests and classified as negative, suspicious, or positive. In herds containing at least one positive cow, we conducted convenience sampling of feces directly from the rectal ampulla to identify MAP through PCR. Of the 362 cows tested, 20 were seroreactive for paratuberculosis from two mesoregions. The PCR tests were all positive; cows with a negative ELISA and positive PCR results probably indicate that the MAP was ingested and eliminated without causing infection. We found that paratuberculosis is likely endemic in the northwest and northeast mesoregions.

A paratuberculose bovina causa diarreia crônica e incurável, perda de peso e resulta em diminuição da produção. A doença é causada pelo Mycobacterium avium subsp. paratuberculosis (MAP), micobactéria intracelular obrigatória, dependente de micobactina, que se replica lentamente no hospedeiro e possui elevada resistência ambiental. Nos países onde a doença é encontrada e os danos foram quantificados, as perdas econômicas diretas e indiretas são extremamente altas. Os dados epidemiológicos locais são de suma importância para a implementação de programas de controle. Nosso objetivo foi verificar se a paratuberculose está presente em rebanhos leiteiros comerciais em diferentes mesorregiões do RS. Para tanto, foi realizado um estudo prospectivo, transversal e observacional em bovinos leiteiros de cinco mesorregiões do estado do RS, Brasil. Amostras de leite individuais, provenientes de vacas leiteiras de fazendas comerciais foram testadas com ELISA indireto e classificadas como negativas, suspeitas ou positivas. Em rebanhos contendo pelo menos uma vaca positiva, realizamos amostragem por conveniência, em que foram coletadas fezes diretamente da ampola retal, para identificar MAP por meio da PCR. Das 362 vacas testadas, 20 foram sororreativas para paratuberculose, oriundas de duas mesorregiões. Os testes de PCR foram todos positivos. Vacas com resultado negativo no teste ELISA e PCR positivo provavelmente indicam que o MAP foi ingerido e eliminado sem causar infecção. Sugere-se que a paratuberculose é provavelmente endêmica nas mesorregiões noroeste e nordeste.


#5 - Retrospective evaluation of canine anal sac carcinoma (260 cases) in Brazil: late detection and failures in lymph node evaluation

Abstract in English:

Anal sac neoplasms are common in companion animals, and the epidemiological profile has been extensively described in international studies; however, national data are still lacking. Data on the Brazilian reality of anal sac carcinoma cases’ diagnosis and treatment are also scarce. The purpose of this study was to retrospectively evaluate cases of canine anal sac carcinoma and assess the profile of involvement, tumor size, and lymphatic invasion at the time of diagnosis. Information was obtained from Vetpat laboratory database, from 260 cases spanning a 12-year period (2010-2021). In histopathological evaluation at the time of diagnosis, data on sex, age, and race were described, as well as tumor size and lymphatic invasion. The presence of metastasis was also assessed in cases where lymph nodes were sampled. Simple descriptive statistical analysis was used to evaluate the data. Adult and elderly, female, and mixed-breed animals were more involved, indicating differences from international studies that can be attributed to sociocultural factors. In terms of tumor size, it was observed that 93% of the cases had the largest diameter above 2.5cm. Only 7% of the cases had the largest diameter below 2.5cm, demonstrating the often late diagnosis and the importance of rectal palpation examination during the general physical assessment of canine patients, particularly at an advanced age. Regarding lymphatic invasion and affected lymph nodes, 50% of the cases had lymphatic invasion described in the histopathological examination. However, only 5% of the lymph nodes were sent along with the primary tumor, indicating the disease’s aggressive behavior but with possible metastases underdiagnosed.

As neoplasias de saco anal possuem incidência importante nos animais de companhia, tendo o seu perfil epidemiológico de acometimento amplamente descrito em estudos internacionais, entretanto dados nacionais ainda são escassos. De modo semelhante, dados acerca da realidade brasileira, associados ao diagnóstico e conduta desses casos, são inexistentes. O objetivo do presente trabalho foi avaliar, de forma retrospectiva, os casos de carcinoma de saco anal canino, avaliando o perfil de acometimento, o tamanho tumoral e a invasão linfática no momento do diagnóstico. Foi obtido informações de 260 casos, respectivo a um período de 12 anos (2010-2021), proveniente do banco de dados do laboratório Vetpat. Foram descritos os dados acerca do sexo, idade e raça, bem como o tamanho tumoral e presença de invasão linfática em avaliação histopatológica no momento do diagnóstico. Os casos que cursavam com envio dos linfonodos também foram avaliados quanto a presença de metástase. Os dados foram avaliados mediante análise estatística descritiva simples. Foi encontrado um maior acometimento em animais adultos a idosos, do sexo feminino e sem raça definida, evidenciando diferenças em relação a estudos internacionais, que podem ser atribuídas a fatores socioculturais. Em relação ao tamanho tumoral, observou-se que 93% dos casos apresentavam o maior diâmetro acima de 2,5cm e apenas 7% dos casos apresentavam o maior diâmetro abaixo de 2,5cm, evidenciando o diagnostico frequentemente tardio, bem como a importância do exame de palpação retal durante a avaliação física geral de pacientes caninos, principalmente em idade avançada. Quanto a avaliação da invasão linfática e linfonodos acometidos, 50% dos casos cursavam com invasão linfática descrita em exame histopatológico, entretanto em apenas 5% dos casos os linfonodos foram enviados junto ao tumor primário, evidenciando o comportamento agressivo da doença, porém com metástases possivelmente subdiagnosticadas.


#6 - Evaluation of five parasitological techniques for diagnosing phylum Ciliophora cysts in fecal samples from free-living wild artiodactyls

Abstract in English:

A variety of laboratory techniques are used in parasitological diagnosis. However, studies that analyze their laboratory efficiency are very scarce, especially with regard to biological samples from wild animals that are little known, with little popular attachment, such as artiodactyls. These can be infected by different parasites, including protozoa of the phylum Ciliophora, which includes the parasites Balantioides coli and Buxtonella sulcata. In this light, the aim of this study was to compare the efficiency of five coproparasitological techniques for diagnosing protozoan cysts of the phylum Ciliophora in the feces of free-living artiodactyls. To this end, 101 fecal samples were collected from trails in Pedra Selada State Park, Rio de Janeiro state, from 2020 to 2021. All the samples were analyzed using the qualitative techniques of modified Sheather floatation, modified Ritchie sedimentation and Lutz, as well as the quantitative techniques of Mini-FLOTAC and McMaster. Cyst recovery was best achieved using the modified Ritchie technique, in which 62.5% positivity was detected, followed by Lutz (47.5%), modified Sheather (37.5%) and the quantitative techniques of Mini-FLOTAC (30%) and McMaster (17.5%). In most of the comparisons between the techniques, reasonable agreement regarding the diagnosis was observed (Kappa 0.21 to 0.40), which was statistically significant (p≤0.05). McMaster showed higher mean and standard deviation values for counts of cysts per gram of feces than Mini-FLOTAC. However, there was no significant difference in the estimates for cyst counts (Wilcoxon p>0.05). Sedimentation qualitative techniques were more indicated for diagnosing cysts of protozoa of the phylum Ciliophora in the feces of free-living wild artiodactyls. These techniques can therefore be used as laboratory tools for environmental parasite monitoring. In addition, between the two quantitative techniques, Mini-FLOTAC presented better performance, thus showing its potential as a tool for estimating the abundance of cystic forms of the phylum Ciliophora in environmental samples.

Diversas são as técnicas laboratoriais utilizadas no diagnóstico parasitológico. No entanto, estudos que analisam a eficiência laboratorial das mesmas são muito escassos, principalmente quando se trata de amostras biológicas de animais silvestres, que possuem pouca divulgação e apego popular, como os artiodáctilos. Estes podem se infectar por diferentes parasitos, incluindo os protozoários do Filo Ciliophora, no qual se inclui Balantioides coli, bem como Buxtonella sulcata. Mediante o exposto, este estudo objetivou comparar a eficiência entre cinco técnicas coproparasitológicas para o diagnóstico de cistos de protozoário do Filo Ciliophora em fezes de artiodáctilos em vida livre. Entre 2020 e 2021 foram coletadas 101 amostras fecais em trilhas do Parque Estadual da Pedra Selada localizado no estado do Rio de Janeiro. Todas as amostras foram analisadas pelas técnicas qualitativas de flutuação de Sheather modificada, sedimentação de Ritchie modificada e Lutz, bem como por meio das técnicas quantitativas de Mini-FLOTAC e McMaster. Pode-se verificar uma superioridade na recuperação dos cistos por meio da técnica de Ritchie modificada, no qual foi detectado 62,5% de positividade, seguida pelo Lutz (47,5%), Sheather modificada (37,5%) e pelas técnicas quantitativas de Mini-FLOTAC (30%) e McMaster (17,5%). Na maioria das comparações entre as técnicas foi possível verificar uma concordância razoável (Kappa 0,21 a 0,40) em relação ao diagnóstico entre as mesmas, sendo este estatisticamente significativo (p≤0,05). McMaster em comparação com o Mini-FLOTAC foi a que apresentou os maiores valores médio e de desvio padrão na contagem dos cistos por grama de fezes. No entanto, não foi evidenciado diferença significativa na estimativa de contagem dos cistos (Wilcoxon=p>0,05). Pode-se verificar que as técnicas qualitativas de sedimentação foram consideradas as mais indicadas para o diagnóstico de cistos do protozoário do Filo Ciliophora em fezes de artiodáctilos silvestres em vida livre, podendo ser utilizadas como ferramentas laboratoriais de monitoramento parasitário ambiental. Além disso, dentre as técnicas quantitativas, o Mini-FLOTAC foi a que apresentou a melhor performance, denotando o seu potencial como uma ferramenta para se estimar a abundância de formas císticas do Filo Ciliophora em amostra ambiental.


#7 - Occurrence of Salmonella spp. in fecal samples from foals with and without diarrhea in the state of São Paulo: microbiological diagnosis, antimicrobial susceptibility profile, and molecular detection

Abstract in English:

The present study investigated Salmonella spp. in the feces of 200 foals up to one year of age (100 with clinical signs of diarrhea and 100 without clinical signs of diarrhea). Bacteriological culture, serotyping, antimicrobial susceptibility, and real-time PCR (qPCR SYBR® Green or a TaqMan®) for detecting the invA gene (with and without a selective pre-enrichment step in tetrathionate broth) were performed. Bacterial culture revealed 15% (n=30) of positive animals (21 animals with diarrhea and nine without diarrhea). Among the 30 isolates, 13 different serovars were identified: S. Infantis, S. Minnesota, S. I.4,5,12:i:-; S. Anatum, S. Cerro, S. Oranienburg, S. Braenderup, S. Give, S. Newport, S. IIIb 61:c:z35, S. 109:-:1.5, S. I.4.12:d:-, S. I.6.8:-:-. Multidrug resistance was found in 43.33% (n=13) of the isolates, with one isolate obtained from animals without diarrhea and 12 isolates from animals with diarrhea. All qPCR techniques used in the study classified more samples as positive for Salmonella spp. than the bacterial culture of feces. In addition, all qPCR techniques detected more positive animals in the diarrhea group than in the diarrhea-free group. The results confirm the utility of the qPCR method without the pre-enrichment step in tetrathionate as a rapid test for Salmonella spp. in carrier animals. In animals with clinical signs of diarrhea, it can be combined with bacterial culture (antimicrobial susceptibility testing and serotyping). The isolation of Salmonella spp. in nine animals without diarrhea confirms the importance of asymptomatic carrier animals in the epidemiology of the disease. The multidrug resistance observed highlights the importance of rational antimicrobial use in horses and adopting biosecurity protocols that are efficacious in controlling the spread of infections between animals and zoonotic transmission in farms.

O presente estudo investigou a ocorrência de Salmonella spp. em fezes de 200 potros com até um ano de idade (100 com sinais clínicos de diarreia e 100 sem sinais clínicos de diarreia), utilizando as técnicas de cultivo bacteriológico e PCR em tempo real (qPCR) pelos métodos de corante fluorescente (SYBR® Green) e sonda específica (Taqman®) para a detecção do gene invA com e sem etapa de pré-enriquecimento seletivo em caldo de tetrationato. O cultivo bacteriológico revelou 15% (n=30) de animais positivos (21 animais com diarreia e nove animais sem diarreia). Dentre esses 30 isolados, 13 sorovares diferentes foram identificados: S. Infantis, S. Minnesota, S. I.4,5,12:i:-; S. Anatum, S. Cerro, S. Oranienburg, S. Braenderup, S. Give, S. Newport, S. IIIb 61:c:z35, S. 109:-:1.5, S. I.4.12:d:-, S. I.6.8:-:-. Multirresistência foi constatada em 43,33% (n=13) dos isolados, sendo um isolado obtido de animal sem diarreia e 12 isolados de animais com diarreia. Todas as técnicas de qPCR empregadas no estudo apresentaram maior número de amostras classificadas como positivas para Salmonella spp. comparadas ao cultivo bacteriológico de fezes. Os resultados confirmaram a utilidade do método qPCR sem a etapa de pré-enriquecimento em tetrationato, como um teste rápido para detecção de Salmonella spp. em animais portadores ou em animais com sinais clínicos de diarreia. O cultivo bacteriológico deve ser associado para a realização do teste de sensibilidade aos antimicrobianos e sorotipificação. O isolamento de Salmonella spp. em nove animais sem diarreia, confirma a importância dos animais portadores assintomáticos na epidemiologia da doença. A multirresistência observada evidencia a importância do uso racional de antimicrobianos em equinos e a importância da adoção de protocolos de biossegurança que sejam eficazes para controlar a disseminação de infecções entre animais e a transmissão zoonótica nas fazendas.


#8 - Evaluation of skin prick test, exclusion diet and dietary challenge in the diagnosis of food allergy in dogs with chronic pruritus

Abstract in English:

Allergic dogs are commonly sensitized to food allergens. We evaluated the use of the prick test to diagnose food allergies in dogs with pruritus and the efficacy of an exclusion diet based on the test. The prick test was performed in 10 healthy dogs and 34 dogs with pruritus, of which 25 received an exclusion diet for 60 days and was challenged with positive food on the test. pVAS and CADESI-4 were assessed on days 0, 30, and 60 after re-exposure. As a result, two control group dogs reacted to a single food allergen, milk and wheat. Of the 25 dogs with pruritus that reacted to food allergens, 24 (96%) reacted to more than one food allergen, and only one (4%) reacted to a single food protein: pork. In the test group (n=25), there was a significant improvement of pVAS and CADESI-4 after 30 and 60 days of dietary exclusion, with significant worsening of the scores with food allergen challenge. In conclusion, the prick test can be used for screening food allergens to make an exclusion diet.

Cães alérgicos são comumente sensibilizados a alérgenos alimentares. Nós avaliamos o uso do teste de puntura para o diagnóstico de alergias alimentares em cães com prurido e a eficácia de uma dieta de exclusão baseada no teste. O teste de puntura foi realizado em 10 cães saudáveis e em 34 cães com prurido, dos quais 25 receberam uma dieta de exclusão por 60 dias e depois desafiados com o alimento que foi positivo no teste. pVAS e CADESI-4 foram realizados nos dias 0, 30, 60 e depois da reexposição. Como resultados, dois cães do grupo controle reagiram a um único alérgeno alimentar, leite e trigo. Dos 25 cães com prurido que reagiram aos alérgenos alimentares, 24 (96%) reagiram a mais de um alérgeno alimentar e apenas um (4%) reagiu a uma única proteína, que foi porco. No grupo teste (n=25), houve uma significativa melhora do pVAS e CADESI-4 depois de 30 e 60 dias de dieta de exclusão, com significante piora dos scores com o desafio alimentar. Como conclusão, o teste de puntura pode ser usado para selecionar alérgenos alimentares para fazer uma dieta de exclusão.


#9 - Detection of feline panleukopenia virus (Carnivore protoparvovirus 1) in free-ranging Panthera onca in Brazil

Abstract in English:

The decline in the jaguar population confirms how much the species is vulnerable to extinction in Brazil. It also indicates the degradation of its natural habitat’s environmental integrity and quality. Studies claim that large felids are susceptible to feline panleukopenia virus (FPV) and are presumptively diagnosed clinically in Brazil. A free-living jaguar (Panthera onca) cub was found unconscious and rescued due to a possible hit-and-run in the savannah of Mato Grosso. During recovery, it exhibited clinical and hematological signs consistent with FPV infection. The PCR was positive for FPV, with 99.61% identity between the FPV sequences available in the GenBank database through the BLAST tool. Due to habitat restrictions, certain diseases threaten wild cats and habitat encroachment by domestic animals can alter the pattern of spread of pathogens. We highlight the importance of the molecular diagnosis and phylogenetic analysis of FPV to elucidate how it has reached wild felids.

O declínio da população de onça-pintada confirma o quanto a espécie está vulnerável à extinção no Brasil, indicando também a degradação da integridade ambiental e da qualidade de seu habitat natural. Estudos afirmam que felinos de grande porte são suscetíveis ao vírus da panleucopenia felina (FPV) e são diagnosticados clinicamente de forma presuntiva no Brasil. Um filhote de onça-pintada (Panthera onca) de vida livre foi encontrado inconsciente e resgatado devido a um possível atropelamento no cerrado do Mato Grosso. Durante a recuperação, apresentou sinais clínicos e hematológicos compatíveis com infecção por FPV. A PCR foi positiva para FPV, com 99,61% de identidade entre as sequências de FPV disponíveis no banco de dados GenBank por meio da ferramenta BLAST. Devido a restrições de habitat, certas doenças ameaçam felinos selvagens e a invasão de habitat por animais domésticos pode alterar o padrão de propagação de patógenos. Destacamos a importância do diagnóstico molecular e da análise filogenética do FPV para elucidar como ele atinge os felídeos silvestres.


#10 - Comparison of macroscopy, histopathology and PCR for diagnosing Eimeria spp. in broiler chickens

Abstract in English:

Coccidiosis is a disease of great importance in industrial poultry. The correct diagnosis directs the poultry industry to its best treatment and control. Thus, a survey of Eimeria spp. was carried out in intestines of 64 broiler flocks, with an average age of 29 days. Eight broilers from each flock were randomly removed from the slaughter line, in a total of 512 samples. Macroscopic and histopathological lesions in the intestine were classified into Scores 0 to 4. Polymerase chain reaction (PCR) was used to research the oocysts from the seven species of Eimeria spp. in the intestinal content. The macroscopic evaluations showed that 59.4% (38/64) of the flocks were positive for E. acervulina, 32.8% (21/64) for E. maxima, 29.7% (19/64) for E. tenella, and 34.4% (22/64) for E. brunetti. The histopathological evaluation showed that 87.5% (56/64) of the flocks had at least one broiler with parasitic structures compatible with Eimeria spp. in the duodenum, 70.3% (45/64) in the jejunum, 18.8% (12/64) in the ileum, 46.9% (30/64) in the cecum, and 4.7% (3/64) in the colon. In PCR, 21.9% (14/64) of the flocks were positive for E. acervulina, 12.5% (8/64) for E. maxima, 3.1% (2/64) for E. mitis, and 32.8% (21/64) for E. tenella. The Kappa Cohen test between macroscopy, histopathology, and PCR demonstrated concordance ranging from weak to moderate with the exception of histopathology and PCR of the cecum, which was strong. In the comparison between macroscopy and histopathology, there were significative differences between Scores 0 and 1 (apart from the cecum). For Score 3, there were significative differences in duodenum, jejunum and cecum (p<0.05). In conclusion, the macroscopic diagnosis and PCR can generate false-negative results, and the histopathological exam proved to be effective, making it essential to associate different techniques for the correct diagnosis of Eimeria spp. in broiler chickens.

A coccidiose é uma doença de grande importância na avicultura industrial. O diagnóstico correto direciona a indústria avícola ao seu melhor tratamento e controle. Desta forma, realizou-se a pesquisa de Eimeria spp. em intestinos de 64 lotes de frangos de corte, com idade média de 29 dias. Em cada lote foram retirados aleatoriamente oito frangos da linha de abate, totalizando 512. Os intestinos foram classificados na macroscopia e na histopatologia em Grau de 0 a 4. No conteúdo intestinal pesquisou-se por reação em cadeia da polimerase (PCR) oocistos das sete espécies de Eimeria. As avaliações macroscópicas demonstraram que 59,4% (38/64) dos lotes foram positivos para E. acervulina, 32,8% (21/64) para E. maxima, 29,7% (19/64) para E. tenella e 34,4% (22/64) para E. brunetti. Na avaliação histopatológica, 87,5% (56/64) dos lotes apresentaram pelo menos um frango com estruturas parasitárias compatíveis com Eimeria spp. no duodeno, 70,3% (45/64) no jejuno, 18,8% (12/64) no íleo, 46,9% (30/64) no ceco e 4,7% (3/64) no cólon. Na comparação dos graus de macroscopia e histopatologia, foram encontradas diferenças significativas entre o Grau 0 e 1 (exceto no ceco) e no Grau 3 houve diferença para duodeno, jejuno e cecos (p<0,05). Conclui-se que o diagnóstico macroscópico e a PCR podem gerar resultados falsos negativos e que o exame histopatológico se demostrou eficaz, tornando fundamental a associação de diferentes técnicas para o correto diagnóstico de Eimeria spp. em frangos de corte.
