#31 - Assessing the histopathology to depict the different stages of bovine tuberculosis infection in a naturally infected herd, 32(2):135-139

E-mail: lusmedeiros@yahoo.com.br The standard method for detection of bovine tuberculosis (TB) is the single intradermal tuberculin test (SITT). Nevertheless, current studies suggest that a single test is not enough to detect all cattle infected by TB, particularly when animals present different stages of infection. A dairy herd comprised of 270 cows was studied and 15 were reactive to SITT plus nine inconclusive animals. Blood samples (for IFN and ELISA) were collected from these 24 cows. At 30 days after injection of PPD, all the cows that were reactive to any of the employed tests were slaughtered, and tissues were processed by Bacteriology, Histopathology (HP) and PCR. According to HP 33.4% of the animals were positive, 45.8% inconclusive and 20.8% were negative. The inconclusive samples came from IFN positive animals, signalizing recent infection. Regarding the animals that were negative to HP, all of them were identified by IFN while ELISA was negative. Immune responses are different in recent and advanced infections, what supports the identification between chronically or recently infected animals. This multidisciplinary approach is mandatory for the interpretation of the various tools that are frequently employed for the diagnosis of TB and mainly to identify all infected animals.

No entanto, estudos atuais sugerem que um único teste não é suficiente para detectar todos os bovinos infectados por TB, particularmente quando os animais de uma rebanho apresentam diferentes estágios de infecção. Um rebanho leiteiro composto de 270 vacas foi estudado e no TCS 15 animais foram reagentes e nove animais inconclusivos. Amostras de sangue (para IFN e ELISA) foram coletadas destas 24 vacas. Trinta dias após a injeção do PPD, todas as vacas que foram reativas a qualquer um dos testes utilizados foram abatidas e os tecidos foram processados ​​por bacteriologia, histopatologia (HP) e PCR. De acordo com a HP 33,4% dos animais foram positivos, 45,8% inconclusivos e 20,8% foram negativos. As amostras classificadas como inconclusivas foram provenientes de animais IFN positivo, sinalizando infecção recente. Em relação aos animais negativos na HP, todos eles foram identificados por IFN enquanto no ELISA apresentaram resultados negativos. Respostas imunes são diferentes em infecções recentes e avançadas, o que suporta a identificação entre os animais cronicamente ou recentemente infectados. Esta abordagem multidisciplinar é obrigatória para a interpretação das várias ferramentas que são freqüentemente empregadas para o diagnóstico da TB e, principalmente, para identificar todos os animais infectados em um rebanho.

E-mail: fassisle@gmail.com The presence of anti leptospiral agglutinins (microscopic agglutination test - MAT) and DNA of leptospires was investigated in the kidney and urine (Polymerase Chain Reaction - PCR) in samples collected at the time of slaughter of cattle originating from the dairy basin of Parnaíba, Piauí, Brazil, as also the lesions in kidney, lung, liver, uterus, ovary and placenta (histopathology and immunohistochemistry). In the MAT, Hardjo was the predominant serovar with the highest number of reagent animals for the strain Hardjobovis/Sponselee. Anti-leptospiral antigens were scored in epithelial cells, interstitial vascular endothelium, endothelium of glomerular capillaries and Bowman’s capsule of 20 positive animals. Inflammatory cells were more common in the kidney. PCR was positive in urine and kidney tissue.

E-mail: fassisle@gmail.com Foi investigada a presença de aglutininas anti leptospiras (reação de soroaglutinação microscópica - SAM), de DNA de leptospiras no rim e na urina (reação de cadeia pela polimerase - PCR), bem como de lesões no rim, pulmão, fígado, útero, ovário e placenta (histopatologia e imunohistoquímica), em materiais colhidos, por ocasião do abate, de bovinos originários da bacia leiteira da região de Parnaíba, Piauí, Brasil. Na SAM o sorovar predominante foi o Hardjo com maior número de animais reagentes para a estirpe Hardjobovis/Sponselee. Antígenos anti leptospira foram marcados em 20 animais positivos nas células epiteliais e do endotélio vascular, endotélio dos capilares glomerulares e na cápsula de Bowman, somente nos animais infectados. O infiltrado inflamatório foi maior no rim do que nos demais órgãos. A PCR foi positiva em amostras de urina e tecido renal.

E-mail: luciele.sm@gmail.com Cytopathologic and histopathologic tests are important to obtain a definitive diagnosis of osteosarcoma. The sample for cytopathological exam is collected through fine-needle aspiration cytology (FNA). On the other hand, histopathological exams need a larger sample that is usually obtained by incisional biopsy. The objective of this article is to develop a FNA technique to biopsy and evaluate histopatologically samples of dogs with suspected osteosarcoma. Two FNS samples were collected from 12 such dogs. Samples obtained in the first procedure were examined cytologically. The material sampled at the second biopsy was fixed in 10% formalin and submitted to histopathological analysis. Four out of the 12 samples (33.3%) examined by the herein proposed method were diagnosed as osteosarcoma. These results indicate a possible adaptation of FNA for histopathological examination.

E-mail: luciele.sm@gmail.com Para a obtenção do diagnóstico definitivo do osteossarcoma realizam-se exames citopatológico e histopatológico. O material para exame citopatológico é coletado através de punção aspirativa por agulha fina (PAAF), já para a realização do exame histopatológico é necessário uma amostra de tamanho maior, geralmente conseguida através de biópsia incisional. Este trabalho tem como objetivo desenvolver uma técnica de coleta de material em cães com suspeita de osteossarcoma através de PAAF para a realização de exame histopatológico. Foram coletadas duas amostras de 12 cães suspeitos de osteossarcoma por PAAF. O material obtido pela primeira coleta foi utilizado para confirmar o diagnóstico através do exame citopatológico, enquanto que o material oriundo da segunda coleta foi fixado em formol a 10% para a análise histopatológica. Quatro das 12 amostras (33,3%) avaliadas histopatologicamente pela metodologia proposta obtiveram também o diagnóstico de osteossarcoma. Esses resultados apontam para uma possível adequação da técnica de coleta de material por PAAF para exame histopatológico.

The 96 h lethal concentration 50 was 97.47mg.L-1 (P<0.05). In the gill, necrotic and proliferative lesions were detected. In the liver, congestion, degenerative foci, hyaline droplets and lipidic vacuolization of the hepatocytes were observed. In the stomach mild hyperplasia of mucous cells was detected, which was also observed in the skin. In this latter tissue, a large increase in the thickness of the epidermis with necrotic lesions, infiltration of leukocytes and melanin pigment were observed. In the brain, degenerative foci of neuronal bodies in the telencephalon associated with gliosis and infiltration of eosinophilic granule cells/mast cells were shown. In conclusion, gills, liver, skin and brain are susceptible to Roundup®. Moreover, effects on the central nervous system could affect olfaction as well as individual and group behavior, the reproductive performance of the fish and hence have repercussions at the population level.

A total of 36 Campolina and Mangalarga Marchador mares in reproductive age (3-23 years) were used, 11 of them were healthy mares (Control group, 7 embryo recipient and 4 embryo donors), and 25 repeat breeders (10 embryo recipient and 15 embryo donors), classified as based on their reproductive history. Endometrial and blood samples were collected for respectively histological and plasma progesterone concentration evaluation. The endometrial samples obtained after biopsy were fixed in Bouin’s fluid, processed, included in paraffin, and stained with Hematoxylin-Eosin (HE) for histopathological examination. Plasmatic progesterone concentrations were evaluated by enzyme immunoessay (ELISA). There was no correlation between progesterone concentration and fertility. But there was a positive correlation between age and fertility, as older mares had major tendency of subfertility than younger ones. There was also a correlation between biopsy categories and fertility, as more histological alterations were found, higher were the chances for the mares to be subfertile. However not all mares classified as Category I and II maintained pregnancy until parturition. Other factors could influence pregnancy maintenance. In the same way, not all mares in Category III were infertile. The endometrial biopsy was shown to be an easy and cheap diagnostic technique with minimal discomfort to the animals and, together with other data, to be a very important component in the investigation of mare fertility.

#36 - Histopathological aspects of Bovine Enzootic Hematuria ln Brazil, 23(2): 65-81

E-mail: peixotop@ufrrj.br The bladder lesions of 59 cattle, from the States of Rio de Janeiro, São Paulo, Minas Gerais, Espírito Santo, Rio Grande do Sul, Santa Catarina, Paraná and Amazon, affected by Bovine Enzootic Haematuria (BEH), were studied histologically. The objective of this study was to describe and redassify neoplastic and non-neoplastic alterations not yet reported, according to the more complete current nomendature used in human medicine. There was an almost complete identity with alterations observed in the bladder of man. Due to the occurrence of two or more neoplasms in the sarne animal, differences in the methodology and in the concept of dassification, a more precise comparison was not possible. Coexistence of different types of epithelial and/or mesenchymal tumour growth was frequently seen. Rare neoplasms or differentiations not previously described were found in the bladder of some animais affected by BEH. These were trabecular carcinoma with Paneth cells differentiation, mesonephroid adenoma, mesonephroid adenocarcinoma, "signet ring" cell carcinoma, plasmocytoid carcinoma, chromophobe cell carcinoma and nested type of transitional cell carcinoma. Haemangiosarcomas originating from haemangiomas were also observed. This study also revealed the occurrence of many tumors with anaplasia and pronounced infiltrative features, but which did not metastasize. The elucidation of the cause of this "barrier against metastases" and its relationship with chemical carcinogenesis induced by the ptaquiloside, the active principie of bracken fern (Pteridium aquilinum), could be of interest to future research on the control of neoplasia in man and animais.

[Aspectos histopatológicos da Hematúria Enzoótica Bovina no Brasil.] Depto Nutrição Animal e Pastagem, Instituto de Zootecnia, UFRRJ, Km 47, Seropédica, Rio de Janeiro 23835-000, Brazil. E-mail: peixotop@ufrrj.br Com o objetivo de descrever alterações neoplásicas e não-neoplásicas ainda não relatadas e, paralelamente, reclassificá-las de acordo com nomenclatura mais completa e atual utilizada em medicina humana, foram estudadas, histologicamente, lesões da bexiga de 59 bovinos com Hematúria Enzoótica (HEB), oriundos dos Estados do Rio de Janeiro, São Paulo, Minas Gerais, Espírito Santo, Rio Grande do Sul, Santa Catarina, Paraná e Amazonas. Verificou-se, em termos qualitativos, quase uma perfeita identidade com as lesões de bexiga observadas em seres humanos. Comparações mais exatas com relação à freqüência dessas alterações ficaram prejudicadas, dadas a ocorrência de duas ou mais neoplasias em um mesmo animal e as diferenças da metodologia empregada ou do conceito de classificação. Coexistência entre neoplasias diversas, epiteliais e/ou mesenquimais, foi vista com freqüência. Neoplasias ou diferenciações raras, ainda não descritas na bexiga de bovinos, como carcinoma trabecular com diferenciação em células de Paneth, adenoma e adenocarcinoma mesonefróides, carcino ma "signet ring" (anel de sinete), carcinoma plasmocitóide, carcinoma de células cromófobas e carcinoma transicional tipo ninhado foram observadas na bexiga de alguns animais com HEB. Foram verificados hemangiossarcomas proliferando a partir de hemangiomas. O estudo revelou, ainda, a· ocorrência de. diversos tumores com anaplasia e carácter infiltrativo acentuados, incapazes, porém, de metastizarem. O esclarecimento da(s) causa(s) dessa "barreira à metástase" e suas relações com a carcinogênese química induzida pelo ptaquilosídeo, o princípio ativo de Pteridium aquilinum, talvez possa ser de interesse em futuros estudos que visem combater o câncer no homem e nos animais.


#37 - A comparative study between cytology and histopathology for the diagnosis of canine neoplasms, 21(1):23-32

[A comparative study between cytology and histopathology for the diagnosis of canine neoplasms] Estudo comparativo entre citopatologia e histopatologia no diagnóstico de neoplasias caninas. Pesquisa Veterinária Brasileira 21(1):23-32. Universidade Federal Rural do Rio de janeiro, Seropédica, RJ 23851-970, Brazil. A comparative study between cytology and histopathology of various neoplasms from 150 dogs was carried out. Severa) staining methods including Wright, May-Gri.inwald-Giemsa, NewMethylene Blue and Papanicolau, and histological staining methods such as Hematoxilin & Eosin, van Gieson, Sudan, Toluidine Blue and PeriodicAcid Schiffwere used. The data obtained indicated an 85.3% accuracy for the cytological diagnosis when the histopathological diagnosis was considered as the correct parameter. In 4.0% of the cases only the embrionary origin of the neoplasrns was determined, and in 1.3% only the prognosis was established. The cytological diagnosis differed from the histopathological diagnosis in 8.1% of the cases. In two cases (1.3%), although the cytological anel histopathological diagnoses differed, a reexamination determined that the former was correct which increased its accuracy up to 86.6%. Fine needle aspiration biopsy proved to be the best technique to obtain samples for the study. Cytology was found not suitable for the diagnosis of mammary neoplasms due to the wide variation of the cytological aspects from different areas. The imprint on slides is a technique not recommended to evaluate mesenchymal tumors and should be replaced by exfoliative cytology. The Wright method proved to be the more efficient staining method. Adapted staining methods from histopathology such as van Gieson for leiomyomas and leiomyosarcomas, Sudan Black B for lipomas anel liposarcomas, and Periodic Acid of Schiff and Toluidine Blue for mastocytomas, were especially useful in visualizing the fine morphological details of various epithelial anel mesenchymal neoplasms examined.

Foi realizado um estudo comparativo entre os diagnósticos citológico e histopatológico em diversas neoplasias de 150 cães, pelas colorações de Wright, May-GrünwaldGiemsa, Novo Azul de Metileno e Papanicolau. Colorações histológicas como Hematoxilina-Eosina, van Gieson, Sudan, Azul de Toluidina e Ácido Periódico de Schiff também foram empregadas. Os dados revelaram uma eficácia de ordem de 85,3% no diagnóstico citopatológico, considerando-se os resultados histopatológicos como corretos. Em 4,0% dos casos somente a origem embrionária das neoplasias foi estabelecida. Em 1,3% das neoplasias apenas o prognóstico foi determinado; o diagnóstico citológico diferiu da histopatologia em 8,1% dos casos. Em dois casos (1,3%) o diagnóstico citológico diferiu do histológico, mas um reexame determinou que o primeiro estava correto, o que elevou a sua eficácia para 86,6%. Entre as técnicas utilizadas, a punção aspirativa por agulha fina foi o melhor método para obter amostras. A citologia não foi adequada para o diagnóstico de neoplasias mamárias, dadas às variações morfológicas em diferentes áreas. A impressão em lâmina não é recomendada para análise de tumores mesenquimais e deve ser substituída pela citologia esfoliativa. O Wright revelou-se o método de coloração mais eficiente. As colorações adaptadas da histopatologia, van Gieson em leiomiomas e leiomiossarcomas, Sudan em lipomas e lipossarcomas, e Ácido Periódico de Schiff e Azul de Toluidina em mastocitomas, foram empregadas com sucesso fornecendo assim maior clareza de detalhes para as diversas neoformações de origem epitelial e mesenquimal.


#38 - ARE ALVEOLAR BONE CHANGES A DETERMINANT FACTOR FOR "CARA INCHADA" IN CATTLE ?

Abstract in English:

In order to study possible alterations of the skeleton which might play a role in the pathogenesis of the periodontitis of "cara inchada" in young cattle, ribs from 20 affected calves, 2 to 10 months old, were examined. Electrolytically decalcified longitudinal sections of the costochondral junction and cross sections through the corpus costae, stained with Haematoxylin-Eosin, were studied. In five calves, longitudinal sections of the proximal humerus were examined as well. The status of mineralization was checked by microradiograms. Systemic alteration of the skeleton due to disturbances of mineral metabolism could not be shown in any of the animals. In seven 2 to 4 months old calves, no bone changes were found. The reduced osteogenesis in six 3 to 5 months old calves and the reduced osteogenesis and diminished chondral growth in seven 5 to 10 months old calves are therefore a consequence of the disease. The results show that the development of the alveolar bone was not defective, so this cannot be a determinant factor for the development of the periodontitis of "cara inchada" in cattle.

Para verificar possíveis alterações do esqueleto, que poderiam ter papel importante na patogênese da periodontite da "cara inchada" em bovinos jovens, costelas de 20 bezerros com 2 a 10 meses de idade e afetados pela doença foram estudadas. Examinaram-se cortes longitudinais eletroliticamante descalcificados das junções cartilaginosas e cortes transversais do corpus costae, corados pela hematoxilina-eosina. Em cinco bezerros examinaram-se também cortes longitudinais do úmero proximal. O estado de mineralização óssea foi avaliado através de microrradiogramas. Em nenhum dos animais alterações do esqueleto por distúrbios do metabolismo mineral podiam ser demonstradas. Nos bezerros com 2 a 4 meses de idade nenhuma alteração patológica do esqueleto foi encontrada. Por isso, a osteogênese reduzida observada nos bezerros com 3 a 5 meses de idade, bem como a osteogênese e o crescimento cartilaginoso reduzidos nos bezerros de 5 a 10 meses de idade, devem ser interpretados como consequências da doença. Os resultados desse estudo demonstram que não existe um desenvolvimento deficiente do osso alveolar como fator predisponente para o desenvolvimento da periodontite da "cara inchada" nos bovinos.
