#221 - Estudo comparativo de éguas repetidoras ou não de cio através da avaliação histológica do endométrio e das concentrações plasmáticas de progesterona, p.506-512

Abstract in English:

ABSTRACT.- Eigenheer-Moreira J.F., Fernandes F.T., Queiroz F.J.R, Pinho T.G.& Ferreira A.M.R. 2007. [Comparative study of repeat breeds and healthy mares through endometrial histology and plasmatic progesterone concentrations.] E-mail: joana.vet@gmail.com.br The study aimed to compare endometrial histology and plasmatic progesterone (P4) concentration of repeat breeds and healthy mares. The hypothesis was that there is a correlation between infertility and endometrial histology and P4 concentration in both groups. A total of 36 Campolina and Mangalarga Marchador mares in reproductive age (3-23 years) were used, 11 of them were healthy mares (Control group, 7 embryo recipient and 4 embryo donors), and 25 repeat breeders (10 embryo recipient and 15 embryo donors), classified as based on their reproductive history. Endometrial and blood samples were collected for respectively histological and plasma progesterone concentration evaluation. The endometrial samples obtained after biopsy were fixed in Bouin’s fluid, processed, included in paraffin, and stained with Hematoxylin-Eosin (HE) for histopathological examination. Plasmatic progesterone concentrations were evaluated by enzyme immunoessay (ELISA). There was no correlation between progesterone concentration and fertility. But there was a positive correlation between age and fertility, as older mares had major tendency of subfertility than younger ones. There was also a correlation between biopsy categories and fertility, as more histological alterations were found, higher were the chances for the mares to be subfertile. However not all mares classified as Category I and II maintained pregnancy until parturition. Other factors could influence pregnancy maintenance. In the same way, not all mares in Category III were infertile. The endometrial biopsy was shown to be an easy and cheap diagnostic technique with minimal discomfort to the animals and, together with other data, to be a very important component in the investigation of mare fertility.

ABSTRACT.- Squarzoni R., Morales M., Safatle A.M.V. & Barros P.S.M. 2007. [Ultrasonographic evaluation of the eyes’ posterior segment of diabetic and non-diabetic dogs with cataracts.] E-mail: renatasq@usp.br Cataracts jeopardize the light penetration into the eye and may restrain direct exam of eye structures. It is necessary to identify very early any eye disease that could cause the failure of cataract surgery. The ocular ultrasonography or echography is a safe, non invasive, inexpensive and easy procedure that complements ocular examination. Two hundred and twenty five eyes of 123 dogs were evaluated using a 10 Mhz linear transducer ultrasound. The dogs were divided into three groups, as there is Group 1: 36 healthy control dogs; Group 2: 52 non diabetic dogs with cataracts; and Group 3: 35 diabetic dogs with cataracts; all of them were patients of the Veterinary Teaching Hospital, University of Sao Paulo. The ultrasonographic findings were in Group 1: vitreous degeneration in 43%, posterior vitreous detachment in 7.7%, asteroid hyalosis in 4.6% of the eyes; in Group 2: vitreous degeneration in 58.9%, posterior vitreous detachment in 8.4%, asteroid hyalosis in 2.1%, inflammatory or haemorrhagic process in 2.1%, retinal detachment in 4.3% of the eyes; and in Group 3: vitreous degeneration in 50.7%, posterior vitreous detachment in 13.8%, asteroid hyalosis in 12.3%, inflammatory or haemorrhagic process in 3.1%, retinal detachment in 3.1% of the eyes. It could be concluded that (1) diabetic dogs, mainly the ones with diabetes for more than 2 years, have greater chances to develop asteroid hyalosis; (2) there were no significant differences in retinal detachment frequencies for the diabetic and non diabetic dogs; and (3) the ocular ultrasonography is a safe, effective and important exam for the pre-operatory evaluation of dogs with cataracts.

Prof. Orlando Marques de Paiva 87, São Paulo, SP 05508-900, Brazil. E-mail: edas@uol.com.br In dogs, the evidence of viability of the right pneumonectomy, as well as the study of complications resulting from surgical procedure, are very important regarding the few specific studies made on the pneumonectomy in this animal species. The objective of this project was to carry out an experimental study to evaluate the viability of the right pneumonectomy in dogs through parametric, hemogasometric and radiographic evaluation. Ten adult healthy mongrel dogs, males and females, weighing 13-32 kg, were used. The dogs were submitted to selective intubation and right thoracotomy at the 5th intercostal space, where the pneumonectomy was performed. A radiografic evaluation was made on days 7, 30 and 60 after the surgery, as well as parametric and hemogasometric evaluations were done (before anesthetic induction, 1 hour after extubation, 48 hours and on days 7, 30 and 60 after the surgical procedure), considered important to evaluate possible complications due to the anesthetic and surgical techniques, as well as complications resulting from these procedures. The results were analyzed statistically. Besides the parametric and hemogasometric alterations, all dogs showed compensation for the gaseous exchanges after removing 57% of the pulmonary volume. Regarding the radiographic evaluation, it could be observed that the expansion of the remaining lung caused heart and lung displacement into the right hemithorax. It can be concluded that the right pneumonectomy is completely feasible in dogs, allowing a parametric, hemogasometric and radiographic satisfactory evaluation in all the animals.

Prof. Dr. Orlando Marques de Paiva 87, São Paulo, SP 05508-270, Brazil. E-mail: flaviovet@usp.br The Brown Capuchin, Cebus apella, has a wide distribution in the northern and southern Brazilian Amazon region and in the Cerrado (savanna). These monkeys are usually submitted to predatory chase, increasing the need for preservation of this wild animal species. An ultrasonographic examination of 10 Brown Capuchins was made in order to describe the normal ultrasonographic anatomy of their abdominal cavity. The urinary bladder revealed its wall thickness with an average of 0.2cm, the topographic situation of which allowed close relation with the wall of uterus and descendent colon. Using caudal abdominal scan, images of aorta, caudal vena cava and right iliac vein were obtained. Liver was accessible for examination by sagittal and cross-section ultrasound, allowing visualization of gallbladder and hepatic vessels. Renal scan allowed accuracy to evidence the echogenicity differences between pelvis, renal sinus, as well as the cortical-medullary relationship. The mean length of the kidneys was 6.24±0.31cm, and no significant differences were observed between left and right kidney length (Student’s t-test and ANOVA). The renal volume obtained was 2.37±0.18cm3. Correlation Coefficients of Pearson between right and left renal length and between right and left renal volume were r = 0.74 and 0.51. Mean thickness for cortical and medullar regions was 0.75±0.11 and 0.39±0.06cm, respectively. Correlation Coefficient of corticomedullar relation between right and left renal was r = 0.19. Examination by ultrasound was considered an efficient, non-invasive, fast and repeatable technique which provides useful data for clinicians and surgeons engaged in wild animal medicine.

ABSTRACT.- Faleiros R.R., Macoris D.G., Alves G.E.S., Saquetti C.H.C. & Alessi A.C. 2007. [Histomorphometric and ultrastructural evaluation of the mucosa of the equine small colon subjected to distention.] Avaliação histomorfométrica e ultra-estrutural da mucosa do cólon menor eqüino submetido à distensão. Departamento de Clínica e Cirurgia Veterinária, Faculdade de Ciências Agrárias e Veterinárias, Universidade Estadual Paulista, Campus de Jaboticabal, Rodovia Carlos Tonanni Km 5, Jaboticabal, SP 14870-000, Brazil. E-mail: faleiros@ufmg.br Recently it has been shown that experimental distention of the small colon of horses promotes reduction of microvascular circulation and inflammation of the seromuscular layer associated with neutrophil accumulation in the lungs. However this model was not sufficient to induce evident histophatological changes in the mucosal layer. The aim of this study was to evaluate the mucosa subjected to that model of small colon distention by histomorphometry and scan electronic microscopy (SEM). Sixteen horses were used. In the distended group (DG), nine of them were subjected to distention of the small colon by a surgically implanted intraluminal balloon that was inflated with a pressure of 40mm Hg during 4 hours. In the sham-operated group (SG), the balloon was implanted but not inflated. Full-thickness intestinal samples were collected before and after obstruction and after 1.5 and 12 hours of decom-pression. By SEM, it was observed that the mucosa turned flat and smooth after distention and returned to the wrinkled original appearance after decompression. Twelve hours after decompression the mucosa had a more irregular appearance with points of fragmentation. There was a reduction in mucosa thickness after distention, returning to basal values after decompression. Instead of the fact that there were changes in appearance and thickness, it was concluded that the mucosa could borne up the compression caused by distention returning to the original characteristics without major lesions.

#226 - Avaliação hemogasométrica em cães submetidos à pneumonectomia esquerda, p.345-351

Abstract in English:

ABSTRACT.- Binoki D.H., Jatene F.B., Fantoni D.T., Stopiglia A.J., Santos A.L.S., Freitas R.R., Simões E.A., Irino E.T., Monteiro R. & Lofiego R. 2007. [Hemogasometric evaluation in dogs submitted to left pneumonectomy.] Avaliação hemogasométrica em cães submetidos à pneumonectomia esquerda. Pesquisa Veterinária Brasileira 27(8):345-351. Departamento de Cirurgia, Faculdade de Medicina Veterinária e Zootecnia, Universidade de São Paulo, São Paulo, SP 05508-270, Brazil. E-mail: stopa@usp.br It is well known that different diseases of infectious, traumatic or neoplasic origin can occur in the lungs of dogs, and pneumonectomy technique may be an option for the treatment of some of these diseases. The objective was to evaluate hemogasometric parameters, oxygen partial pressure (PaO2), carbon dioxide partial pressure (PaCO2), hydrogen ion concentration (pH) and bicarbonate ion concentration on the pre- and post-operative moments of the left pneumonectomy. Eighteen adult mongrel dogs, males and females, were randomly distributed into two groups with 9 dogs each. In Group A, the left bronchial stump of the dogs was sutured manually with polypropylene 5-0, and, in Group B, the left bronchial stump of the dogs was sutured mechanically with a surgical stapler. The data were collected at 6 moments: Before the pre-anesthetic administration (T0), one hour after the extubation (T1EXT), 48 hours after the surgery (T48h), 7 days after the surgery (T7d), 15 days after the surgery (T15d), and 36 days after the surgery (T36d). The results were statistically analyzed. PaO2 values of Group A on T1EXT (67.00±11.31) were significantly lower in relation to T0 (99.4±18.34), a fact that did not happen in Group B: T1EXT (87.00±8.35) in regard to T0 (87.00±7.55). There was no difference on pH values in dogs of Group A, but in Group B was observed a decrease on T1EXT (7.3644±0.0353) in relation to T0 (7.4189±0.0136), although the animals did not develop acidosis. It was concluded that dogs submitted to left pneumonectomy (sutured manually or sutured mechanically of left bronchial) did not show immediate and mediate acid-basic instability during the study.

Two ewes and six lambs were divided into seven treatments and fed the plant. The adult ewes did not show clinical signs of poisoning. The 4 to 5-month-old lambs presented partial wool loss 6 days after beginning of the administration of the plant and total wool loss after 12 or 20 days. The age of the sheep in this experiment suggests resistance of adult ewes and susceptibility of young lambs to L. leucocaephala poisoning.

#228 - Desenvolvimento e avaliação de novas estratégias de imunização contra colibacilose suína, p.84-90

Abstract in English:

Simionatto S., Vaz E.K., Michelon A., Seixas F.K., Dellagostin O.A. 2005. [Development and evaluation of new strategies for immunization against swine colibacillosis.] Postal 354, Pelotas, RS 96010-900, Brazil. E-mail: ssimionatto@bol.com.br Swine colibacillosis caused by enterotoxigenic Escherichia coli remains one of the main sanitary problems in pig farms. The recombinant DNA technology offers the possibility of developing new immunization strategies. This paper describes the development of a subunit vaccine through the expression and purification of the E. coli K88 FaeC fimbrial protein. The gene that codes for this antigen was amplified by PCR and cloned into an E. coli expression vector fused to a 6X histidine tag. The recombinant protein was purified by affinity chromatography and used for mice immunization. In parallel, the same gene was cloned into an eucariotic expression vector with the addition of the Kozak sequence for improving translation of this gene in muscle cells. The resulting plasmid named pUP310 was purified in large scale and used to immunize mice. The immune response afforded by both forms of immunization was monitored by ELISA. There was an immune response in mice inoculated with pUP310 and purified FaeC. It was possible to detect anti-FaeC antibodies 42 days after the first inoculation. The antibody titer increased with time, being still detectable 7 months after the first inoculation. It is concluded that recombinant FaeC and pUP310 are potential tools for immunization of swine against E. coli K88.

[Efficiency of the disinfection of teatcups and teats during mechanic milking of dairy cows.] E-mail: lamaral@fcav.unesp.br The purpose of this investigation was to evaluate the efficiency of the teatcup disinfection process by immersion into a disinfectant solution, using sodium hypochlorite and organic chlorine, to verify the residual chlorine dynamics in both solutions during milking time, and to evaluate three premilking disinfection methods: immersion, spray and sponge. Total coliforms, fecal coliforms, Staphylococcus sp and mesophilic microorganism counting was done. The results showed that the disinfecting clusters between individual cows, using immersion in sodium hypochlorite or organic chlorine solutions with a concentration of 150ppm, was not efficient for the rubber teatcup microorganism reduction. The organic chlorine solution showed more stability. Teatcup is a potential microorganism vehicle for milk and the mammary gland. Disinfection of the teats reduced the number of the microorganisms studied in all treatments, showing to be a simple method that can be used to reduce the microrganism transmission risks during milking and to increase the microbial quality of the milk.

E-mail: davetpat@vortex.ufrgs.br The study assessed differences of pathogenicity among 19 strains of Brachyspira pilosicoli isolated from cases of diarrhea in swine in the State of Rio Grande do Sul, Brazil, using an one-day-old chick model. The chicken inoculated with live bacterial cultures were submitted to euthanasia 21 days later, and the cecum was examined histologically using hematoxylin-eosin, silver staining and immunohistochemistry with a polyclonal antibody anti-Brachyspira pilosicoli. With silver staining and immunohistochemistry, respectively, 21.59% and 70.96% of the chicken showed colonization of the cecal epithelium with B. pilosicoli. Differences in the type of colonization characterized by continuous adhesion, focal adhesion or free bacteria in the intestinal lumen were observed. Immunohistochemistry was more efficient to assess intestinal colonization, because 49.37% more colonized chicken could be detected with silver staining. With three strains, elongated figures inside the cytoplasm of epithelial cecal cells in the inoculated chicken were observed.
