Pesquisa Veterinária Brasileira 31(10):867-874. Projeto Sanidade Animal Embrapa/UFRRJ, Seropédica, RJ 23890-000, Brazil. E-mail: michel_abdallavet@yahoo.com.br This study aimed to evaluate the protective effect of acetamid in experimental poisoning by Pseudocalymma elegans in sheep, goats and rabbits, in order to prove indirectly that monofluoroacetate (MF) is responsible for the clinical signs and death of animals that ingested the plant. Experiments were performed to determine for sheep and goats the lethal dose of P. elegans collected in Rio Bonito, RJ, in different seasons, and to adjust the dose of acetamid to be administered. - In the first experiment, two sheep and two goats received 1.0g/kg of fresh P. elegans, and two (one sheep and one goat) were pretreated with 2.0g/kg of acetamid. None of the animals showed clinical signs or died. Possibly, the plant could be less toxic, since it was collected at the end of the rainy season. - In the second experiment, two sheep and two goats received 0.67 and 1.0g/kg of the dried plant, after pretreatment with 2.0 and 3.0g/kg of acetamid, respectively. All animals died, as the administered doses of P. elegans were very high. - In the third experiment, two sheep and two goats received 0.333g/kg of dried P. elegans after previous administration of 2.0g/kg of acetamid; a week later, the protocol above was repeated, but without the antidote. In experiments with rabbits, doses of 0.5 and 1.0g/kg of dried P. elegans were given after administration of 3.0g/kg of acetamid; seven days later, the same protocol was repeated, except the administration of acetamide. This procedure, when acetamid was administered before, prevented the appearance of clinical signs and death of sheep, goats and rabbits. But the animals not treated with acetamid showed symptoms of poisoning and died. Clinically, the sheep and goats had tachycardia, engorged jugular vein, positive venous pulse, lateral recumbence, and muscle tremors. In the “dramatic phase”, the animals fell into lateral position, stretched the limbs, were paddling and died within minutes. The rabbits showed apathy, muscle tremors, vocalization and lateral decumbence minutes before death. At postmortem examination, the sheep and goats had engorged jugular veins and atria, dilated vena cava cranialis and caudalis, as well as pulmonary edema, hepatic congestion and edema of the gallbladder subserosa. In rabbits, the main macroscopic alterations were dilated atria, engorged vena cava cranialis and caudalis, and congested liver and diaphragm vessels. Histopathology revealed, in two sheep and one goat, vacuolar-hydropic degeneration of the distal convoluted kidney tubules, together with caryopicnosis. In the rabbits, the liver showed severe congestion with numerous shock corpuscles. The experimental results show indirectly that MF is to be held responsible for death of the animals that ingested P. elegans; since “acetate donor” compounds, such as acetamid, are capable to reduce the competitive inhibition of MF for the same active site (Coenzyme A) which prevents the formation of fluorocitrate, its active metabolite, formed in the body through the so-called “lethal synthesis”.

E-mail: teitsouza@yahoo.com.br The aim of this study was to determine the prevalence of the equine skin tumors diagnosed at the Laboratory of Veterinary Pathology (LPV) of the Universidade Federal de Santa Maria (UFSM). Cases of equine skin biopsies diagnosed between January 1999 and December 2009 and filed at the LPV-UFSM were reviewed. In all cases the morphological diagnosis considered was the one written in the filed biopsy report. During the period studied 139 equine skin biopsies were submitted to the LPV-UFSM. Out of those, 108 (77.6%) were skin tumors, neoplastic or otherwise and 31 cases were non-tumoral dermatosis. Fifty three of the 108 horses were females (49.1%), 37 (34.2% %) were males and in 18 cases (16.7%) the sex was not stated in the report. Sixty six affected horse (61.1%) were purebreds, 13 (12%) were crosses and in 29 instances (26.9%) the breed was not stated in the report. The Crioulo was the most prevalent breed (44/108 [40.7%]) and the remaining 22 purebreds horses were distribute among seven different breed. The most prevalent age categories affected in decreasing order were 1-5 year-old (47/108 [43.5%]) and 6-14 year-old (21/108 [19.5%]); 12 horses (11,1%) were 15 year-old or above .Only one horse was less than 1-year-old. In 27 reports the age was not mentioned. More prevalent tumors included equine sarcoid (62/108 [57.4%]); squamous cell carcinomas (11/108 [10.2%]); pythiosis (9/108 [8.3%]); granulation tissue (“proud flesh”) (7/108 [6.5%]) and eosinophilic granuloma (4/108 [3.8%]). The remaining tumors accounted for approximately 14% of the cases.

#93 - Clinic-pathological and laboratory aspects of experimental poisoning by sodium monofluoroacetate in sheep, 30(12):1021-1030

Abstract in English:

ABSTRACT.- Peixoto T.C., Nogueira V.A., Coelho C.D., Veiga C.C.P., Peixoto P.V. & Brito M.F. 2010. [Clinic-pathological and laboratory aspects of experimental poisoning by sodium monofluoroacetate in sheep.] Projeto Sanidade Animal Embrapa/UFRRJ, Seropédica, RJ 23890-000, Brazil. E-mail: tiagocpeixoto@yahoo.com.br The objective of this study was to verify if the ingestion of single doses of sodium monofluoroacetate (MF) and daily fractions of 1/2.5 and 1/5 of the lethal dose causes the same lesion as the one observed in the kidney of cattle poisoned by Brazilian sudden death causing plants (BSDCP). MF was administered orally in single doses of 0.5 and 1.0mg/kg to four sheep, and repeated daily doses of 0.1 and 0.2mg/kg to two others. Death occurred in five of six animals. The course of poisoning lasted from 3min to 33h5min. Clinically the animals presented palpitation, abdominal breathing, slight balance loss with sometimes swaying gait, they laid down and placed the head on their flank. In the “dramatic phase”, all the sheep fell into lateral decubitus, stretched out the legs, made peddling movements, presented opistotonus, and died. The electrocardiographical examination showed heart dilatation and reduction of the systolic shortening fraction. Laboratory hematological exams revealed increased urea and creatinine. MF caused the clinical and pathological symptoms of “sudden death”. At postmortem examination, heart auricles and jugular, cava, azygos and pulmonary veins of all animals were moderately engorged, and in some sheep, pulmonary edema was observed. Histopathology revealed hydropic-vacuolar degeneration (HVD) of the epithelial cells of the distal convoluted kidney tubules associated with nuclear picnosis in all the sheep. Vacuolation and less often necrosis of liver cells was seen in some cases. No references to that peculiar type of lesion could be found in the literature, except the description of kidney lesions in animals associated with the ingestion of BSDCP, and recent studies of MF poisoning in cattle. The present study demonstrated in sheep that single lethal doses or repeated doses of fractions of the lethal dose of MF causes HVD of the distal convoluted kidney tubules, associated with nuclear picnosis.

E-mail: sheilacr@fmvz.unesp.br The aim of this study was to histologically evaluate the tissue reaction to implantation of polyurethane disks containing castor oil produced in premolded form, supplied commercially, and biomass molded at the moment of application; since a late tissue reaction may occur during the total hardening process of the biomaterial. Twenty female Wistar rats, about 3 months of age, weighing 300-350g, and polyurethane disks 0.8cm in diameter and 0.5cm in thickness were used. The premolded disk was implanted into the subcutaneous tissue at the right flank and the molded biomass disk at the left flank. Five rats each were submitted to euthanasia at 3, 7, 15 and 30 days after surgery, and the polyurethane disks and surrounding tissue were collected. The specimens were processed for HE staining and examined microscopically. A moderate inflammatory reaction was primarily composed of polymorphonuclear cells and macrophages. The lymphocytes varied from absent to discreet. The intensity of the inflammatory reaction decreased at the same time the formation of fibrous conjunctive tissue increased around the implants. However, the numbers of macrophages remained the same. In conclusion, both polyurethane disks induce the same type of inflammatory reaction that varies from slight to moderate according to evaluation time.

#95 - Histomorphometry of encephalic meninges of Wistar rats in different band, 30(11):996-1002

ABSTRACT.- Pereira K.F., Lima V.M., Conegero C.L. & Chopard R.P. 2010. [Histomorphometry of encephalic meninges of Wistar rats in different bands.] E-mail: kpereira@usp.br The development of the nervous sys tem is very complex and there are few studies about the organization of the brain envoltories related to the encephalus growing. Using the rat as an animal model, it was proposed to evaluate the several structural aspects of paquimeninge and leptomeninge in different ages. It was used 4 groups of different ages and processed according to the techniques of the light and transmission microscopy. It was verified that the adult rats present a higher area of collagen fibers of type I and III, if compared to the others groups. It was found that, the collagen fibers of type III occupy, in all analyzed groups, a higher area when compared to type I fibers. The results reveal that the Weigert Oxona’s staining, which shows elastics, elauninics, and oxitalanics fibers, showed a statistically difference when compared to the Weigert’s staining and Verhoeff’s staining that show elauninics and elastics fibers, respectively. The ultra-structural aspects demonstrated the presence of many fibroblasts and mitochondria in the paquimeninge and also in the leptomeninges of the neonats and adults groups, indicating the high cellular activity and consequently, an intense formation of conjunctive tissue. As collagen fibers of type III acting on the structural maintenance of delicate and expansive tissues, the study shows that the function of the encephalic meninges are not only related to the to resistance to tractions and tensions that the encephalus is subjected. But also the function related to the distensibility of the meningeos and brain vases according to the sanguineous apport in several specific functions of the nervous system.

E-mail: paulaaragao.vet@gmail.com The purpose of this study was to establish the clinic-pathological and laboratory changes in sheep inoculated with Bothropoides jararaca and Bothrops jararacussu venom to provide subsidies for the differential diagnosis of snake bites. The liofilized venoms were diluted in 1 ml saline and administrated subcutaneously to four sheep. Three of the animals died, and the one that received 0.5mg/kg (B. jararaca venom) recovered. First symptoms were observed from 7 minutes to 1 hour after inoculation, and the clinical course varied from 7 hours and 9 minutes to 21 hours and 59 minutes. The symptoms, independent of the dosage, were swelling of the inoculation site, increased bleeding time and capillary filling, tachycardia, dyspnea, pale mucous membranes and diminished reaction to external stimuli. Laboratory tests revealed pronounced normocytic and normochromic anemia, trombocytopenia, slight reduction of fibrogen and total plasmatic protein, in two animals diminished hematocrit, besides pronounced increase of creatinaquinase and lactic dehydrogenase. At necropsy, the main findings at the inoculation site and adjacent tissues were extensive hemorrhages in the sheep inoculated with jararaca venom, and predominantly edema in the two animals inoculated with jararacussu venom. In two sheep which received jararacussu venom, acute pulmonary edema was observed. Hemorrhage and edema as the main histopathological changes, besides necrosis of muscle fibers and vessels at the inoculation site and adjacent tissue was observed. The renal tubular necrosis was attributed to shock. The volume increase at the inoculation site and surroundings was mainly due to hemorrhage (B. jararaca) or edema (B. jararacussu).

#97 - Interference of age and repetition of the same noxious stimulus on hyperalgesia, 30(9):777-782

ABSTRACT.- Ibañez J.F., Posso I.P. & Wallace V. 2010. Interference of age and repetition of the same noxious stimulus on hyperalgesia. Pesquisa Veterinária Brasileira 30(9):777-782. Departamento de Patologia Geral, Curso de Medicina Veterinária, Universidade Estadual do Norte do Paraná, Campus Luiz Meneghel, Rodovia BR 369 Km 54, Bandeirantes, PR 86360-000, Brazil. E-mail: ibanez@uenp.edu.br Pain in animals has been recognized for less than one century. Several authors confirm that animals are capable to process, register and modulate nociceptive stimuli in a very similar way to human kind and there are several evidences registering the impact of pain sensation over vital systems interfering on disease outcome. Nevertheless, despite some evidences that animals, as human beings, can store information from past painful experiences less is known about how this so called pain memory works. The aims of this study were: to evaluate if the response to a painful stimuli differs during different stages of life and if repetition of a same acute stimuli in the same animal interferes with expression of hyperalgesia. Thus, 60 rats were selected and arranged in 3 equal groups: 3 months, 6 months, and 9 months of age. All animals were injected 5% formalin solution in the plantar face of hind paw under volatile general anesthesia. Von Frey filaments were applied at 1h, 24h and 48h after sensitization. Injection was repeated twice with a 30-day interval, each time in a different hind paw. Results showed that younger rats express lower hyperalgesia thresholds in the first stimulation compared to elder animals and that repetition of same stimulus diminishes hyperalgesia thresholds when it begins during infant period and augments hyperalgesia thresholds when it begins during elder ages.

E-mail: eduardofurtadoflores@gmail.com The activity of Foscarnet (PFA) against three bovine herpesviruses (BoHVs) was previously demonstrated in cell culture. Herein we evaluated the effects of PFA on the infection and disease by BoHV-1 and BoHV-5 in a rabbit model. Rabbits inoculated with BoHV-5 in the conjunctival sac (IC) and treated with PFA (100 mg/kg/day) from day 1 to 17 post-inoculation (pi) shed less virus between days 2 and 6 pi comparing to untreated controls; this difference was significant at day 3 pi [F(9,108) = 2,23; P<0.03]. The morbidity and mortality rates of rabbits inoculated with BoHV-5 IC or intranasally (IN) were also significantly reduced in PFA-treated rabbits (50%; 11/22) comparing to untreated controls (95.4%; 21/22) (P<0.0008). In rabbits inoculated IC with BoHV-1, a reduction in virus shedding was observed in PFA-treated animals between days 1 and 4 pi; 6 and 7 pi. In addition, PFA-treated rabbits presented a longer incubation period and a shorter clinical course comparing to untreated controls (P<0.005 and P<0.04, respectively). The frequency and severity of ocular signs were also reduced in the PFA-treated group. These results demonstrate that PFA is effective against BoHV-1 and BoHV-5 in vivo and open the way towards its use in experimental therapy of herpetic infections in domestic animals.

Abstract in English:

ABSTRACT.- Vailati M.C.F., Rocha N.S., Matsubara L.S., Padovani C.R., Schwartz D.S. & Matsubara B.B. 2010. AT1-receptor mediated vascular damage in myocardium, kidneys and liver in rats. Pesquisa Veterinária Brasileira 30(7):605-611. Departamento de Clínica Veterinária, Faculdade de Medicina Veterinária e Zootecnia, Universidade Estadual Paulista, Distrito de Rubião Júnior s/n, Botucatu, SP 18618-000, Brazil. E-mail: mfvailati@yahoo.com.br The systemic aspect of vascular damage induced by angiotensin II (ANG II) has been poorly explored in the literature. Considering the presence of ANG II and its specific receptor AT1, in several organs, all tissues might be potentially affected by its effects. The aims of this study were: To evaluate the early histological changes in the heart, liver and kidneys, produced by ANG II infusion, to evaluate the protective effect of losartan. Wistar rats were distributed into three groups: control (no treatment), treated with ANG II, and treated with ANG II + losartan. ANG II was continuously infused over 72 hours by subcutaneous osmotic pumps. Histological sections of the myocardium, kidneys and liver were stained and observed for the presence of necrosis. There were ANG II-induced perivascular inflammation and necrosis of the arteriolar wall in the myocardium, kidney, and liver by, which were partially prevented by losartan. There was no significant correlation between heart and kidney damage. Tissue lesion severity was lower than that of vascular lesions, without statistical difference between groups. ANG II causes vascular injury in the heart, kidneys and liver, indicating a systemic vasculotoxic effect; the mechanisms of damage/protection vary depending on the target organ; perivascular lesions may occur even when anti-hypertensive doses of losartan are used.

[Structure and cellularity of the fresh menisci (Oryctolagus cuniculus) of rabbits and the menisci preserved in glycerin.] E-mail: ricarlo@ufv.br In the present study was evaluated the tissue architecture, the percentage of cellular population, as well as viability and distribution of cells in fresh menisci of rabbits and preserved in 98% glycerin. Were analyzed medial menisci of rabbits freshly slaughtered, which were distributed into three groups: the MF group (n=7), composed of fresh menisci, corresponded to the control group; the MG group (n=7), composed by menisci preserved in 98% glycerin, for 30 days, and the MR group (n=7) by menisci preserved in 98% glycerin and rehydrated in NaCl 0.9% for 12 hours. In all menisci were identified and quantified the different cell types: fibroblasts/fibrocytes and condrocytes. The cell population percentage was statistically similar in all groups. All menisci preserved in the MG and MR groups showed a lower intensity of color and shrinkage of collagen fibers, reduced volume and higher intensity of staining of nucleus (chromatin condensation), as compared to fresh menisci (MF), featuring the phenomenon of cell lysis. The cartilaginous matrix of preserved menisci proved to be well preserved because the tissue architecture was maintained. It was concluded that 98% glycerin is an optional preservation mean for meniscal allografts with a devitalized collagenous matrix.
