E-mail: proehe@gmail.com The serum neutralization (SN) test is the gold standard method to measure neutralizing antibodies to bovine herpesviruses. However, in view of the further subdivisions of bovine herpesviruses in types/subtypes, defining which virus to use at challenge in SN tests may be difficult. In view of that, this study was carried out to re-evaluate (SN) sensitivity with different types/subtypes of bovine herpesviruses types 1 (BoHV-1) and 5 (BoHV-5) as challenge viruses. Bovine sera (n=810) were collected from two distinct geographic regions and tested by SN with three type 1 viruses (BoHV-1.1 strains “Los Angeles” and “EVI123/98”; BoHV-1.2a strain “SV265/96”) and three type 5 viruses (BoHV-5a strain “EVI88/95”; BoHV-5b strain “A663” and BoHV-5c “ISO97/95”). SN tests were performed with a 1 hour incubation of the serum-virus mixtures at 37ºC against 100 TCID50 of each of the viruses. SN sensitivity varied greatly depending on the challenge virus used in the test. The highest sensitivity (327 positive/810 total sera tested; 40.37%) was attained when the positive results to the six viruses were added together. No association could be found between any particular type or subtype of virus and the sensitivity of the test. When positive results to each single strain were considered, SN sensitivity varied from 41.7% to 81.7%, depending on the virus and the geographic region of origin of the sera. Variation was detected even when challenge viruses belonged to the same subtype, where disagreement between positive results reached 41%. These results indicate that one hour incubation SN tests against single viruses, as performed here, may display a significantly low sensitivity (p=0.05); performing SN tests against a number of different viruses may increase considerably SN sensitivity. Furthermore, the choice of virus used for challenge is critical in SN tests. In addition, sera from different geographic regions may give rise to disagreeing results with different strains of BoHV-1 and BoHV-5. This might be particularly relevant for control programs and in international trade, were maximum sensitivity should be targeted.

Entretanto, com as subdivisões propostas destes agentes em tipos e subtipos, a definição de qual amostra utilizar como virus de desafio à SN pode ser difícil. Em vista disso, este estudo foi realizado para re-avaliar a sensibilidade de testes de SN utilizando diferentes tipos e subtipos de herpesvírus bovinos tipos 1 (BoHV-1) e 5 (BoHV-5) como amostras de desafio. Soros bovinos (n=810) foram coletados de duas regiões geográficas distintas e testados frente a amostras do tipo 1 (BoHV-1.1: amostras “Los Angeles” e “EVI123/98”, BoHV-1.2a: amostra “SV265/96”) e três amostras do tipo 5 (BoHV-5a: “EVI88/95”; BoHV-5b: “A663” e BoHV-5c “ISO97/95”). Os testes de SN foram realizados com incubação de 1 hora a 37ºC da mistura soro-vírus, frente a 100 doses infectantes para 50% dos cultivos celulares (DICC50) de cada um dos vírus. A sensibilidade da SN variou grandemente em função do vírus utilizado no teste. A maior sensibilidade (327 soros positivos/810 soros testados; 40.37%) foi alcançada quando os resultados positivos frente aos seis diferentes vírus foram somados. Nenhuma associação foi detectada entre determinado tipo/subtipo de vírus e a sensibilidade do teste. Quando resultados positivos frente a cada vírus foram considerados isoladamente, a sensibilidade da SN variou entre 41,7% a 81,7%, dependendo do vírus de desafio e da região geográfica de origem das amostras de soro. Variação foi detectada mesmo quando as amostras de desafio pertenciam a um mesmo subtipo; a discrepância entre os resultados positivos atingiu até 41%. Estes resultados indicam que testes de SN contra amostras isoladas de vírus podem apresentar uma sensibilidade notadamente baixa; o emprego de diferentes amostras de vírus de desafio pode aumentar consideravelmente a sensibilidade da prova. Além disso, a escolha da amostra de vírus para a realização do teste é crítica. Outro achado importante é que sorors de diferentes regiões geográficas podem dar resultados discordantes frente a diferentes amostras de BoHV-1 e BoHV-5. Estes achados são particularmente relevantes para programas de controle destas infecções e para o comércio internacional, onde a sensibilidade deve ser maximizada.

E-mail: carlalopes.mendonca@gmail.com The aim of the present study was to evaluate the effect of Staphylococcus aureus experimentally induced mastitis on proteinogram and serum concentrations of cupper, iron and zinc levels of Santa Ines primiparous ewes . The right mammary gland of ten healthy ewes was inoculated with 1,0x104 UFC/mL of S. aureus. Clinical examination and determination of serum concentrations of proteins by electrophoresis in polyacrylamide gel (SDS-PAGE), cupper, iron and zinc, as well plasma level of fibrinogen were measured before the inoculation (control) and 12h, 24h, 36h, 48h, 60h, 72h, 84h, 96h, 108h, 120h, 132h, 168h, 180h, 288h and 336h after bacteria inoculation. All animals experimentally infected presented clinical mastitis and subsequent loss of mammary gland function. The electrophoretogram allowed the identification of 23 proteins with molecular weights (MW) ranged from 26.000 to 185.000 daltons (Da) including acute-phase proteins, IgG and IgA. A significant increase (P<0,05) in haptoglobin, ceruloplasmin, IgG and IgA concentrations was observed. Antitrypsin and acid glicoprotein concentrations did not alter. The levels of iron and zinc decreased and the cupper concentration increased . A positive correlation between plasma fibrinogen and serum ceruloplasmin (r=0.74), haptoglobin (r=0.62) and IgA (r=0.62) was also identified. Results showed the importance of ceruloplasmin and haptoglobin as acute-phase proteins in ewes with intramammary infections and confirms fibrinogen as an inflammatory marker because its high correlation with specific proteins. The alterations in the serum levels of Cu, Fe and Zn suggest the action of inflammatory mediators triggered by S. aureus.

Foram utilizadas 10 ovelhas da raça Santa Inês, primíparas, recém-paridas, com aproximadamente dois anos de idade e bom estado nutricional. Inoculou-se na metade direita da glândula mamária 1,0x104UFC/mL da bactéria, enquanto que a metade esquerda serviu como controle. Os animais foram acompanhados diariamente e a partir do diagnóstico clínico de mastite, procedeu-se colheita do material para realização do proteinograma sérico em gel de poliacrilamida contendo dodecil sulfato de sódio (SDS-PAGE) e para determinação do teor plasmático de fibrinogênio e das concentrações séricas de cobre, ferro e zinco em 16 momentos a saber: antes da inoculação (controle) e 12h, 24h, 36h, 48h, 60h, 72h, 84h, 96h, 108h, 120h, 132h, 168h, 180h, 288h e 336h após a inoculação (p.i.). Todas as ovelhas apresentaram quadro clínico de mastite, com perda da funcionalidade da glândula mamária. O proteinograma permitiu a identificação de 23 proteínas, cujos pesos moleculares (PM) variaram de 26.000 a 185.000 dáltons (Da), incluindo proteínas de fase aguda, IgG e IgA. Notou-se aumento significativo nas concentrações de haptoglobina e ceruloplasmina, assim como de IgG e IgA. Não se constatou alteração nos teores de antitripisina e de glicoproteína ácida .Verificou-se diminuição nos teores de ferro e zinco e elevação na concentração de cobre. Constatou-se correlação positiva entre o teor plasmático de fibrinogênio e as concentrações séricas de ceruloplasmina (r=0,74), a haptoglobina (r=0,62) e IgA(r=0,62). Estes resultados mostram a importância das proteínas de fase aguda ceruloplasmina e haptoglobina como indicadores auxiliares da infecção intramamária de ovelhas, assim como ratifica a relevância do fibrinogênio como marcador inflamatório em razão de sua alta correlação com as proteínas especificas. As alterações nas concentrações séricas de Cu, Fe e Zn sugerem a ação de mediadores inflamatórios, estimulados por S. aureus.

E-mail: fonteque@cav.udesc.br The objective was to evaluate the electrophoretic profile of proteins and serum concentration of immunoglobulin G (IgG) in Saanen goats with mastitis experimentally induced by Staphylococcus aureus (dl-a-tocopherol acetated). 14 adult goats, (supplemented with vitamin E DL-a-tocopherol) primiparous pregnant, seronegative for caprine arthritis encephalitis (CAEV), clinically healthy, were divided into two groups of seven animals: Not supplemented group (G1) and group supplemented with 2.000 UI of DL-a-tocopherol (G2 Vit E), by intramuscular injection on the day of the parturition and seven days later. At the 9th day after delivery 300 UFCs of the S. aureus ATCC 225923 strain were inoculated into the left half of the mammary gland of each animal. The mastitis was determined through collection of milk samples for evidence of infection by means of bacteriological examination, somatic cell count (SCC) and California Mastitis Test (CMT). Then samples were collected after 12, 24, 48 and 72 hours, antimicrobial intra-mammary gland treatment was initiated, with new evaluation 48 hours after treatment. The electrophoretic profile of serum protein of the goats, showed five fractions, as follows: albumin and globulin (a, b1, b2 e g-globulin). There was an increase in the production of g-globulin and lower production of b2-globulin fraction 12 hours after infection, and faster decrease in the supplemented group, showing the influence of vitamin E in the production of acute phase proteins. There was no influence of vitamin E in the serum concentration of immunoglobulin G (IgG) in supplemented animals. The supplementation with vitamin E increased the concentration of immunoglobulin and decreased the production of acute phase proteins, probably the antioxidant effect minimizing the tissue injury during the inflammatory process in the mammary gland.

Grupo não suplementado (G1) e grupo suplementado com 2.000 U.I. de acetato de dl-a-tocoferol (G2 Vit E) via intramuscular no dia do parto e sete dias após o parto. Ao nono dia do pós-parto foram inoculados 300 UFCs da cepa de S. aureus ATCC 225923, na metade esquerda da glândula mamária de cada animal. A mastite foi determinada pela colheita das amostras de leite para a comprovação da infecção, por meio de exames bacteriológicos, contagem de células somáticas (CCS) e California Mastitis Test (CMT), a partir deste momento foram efetuadas colheitas às 12, 24, 48 e 72 horas, sendo posteriormente instituído o tratamento intramamário com antimicrobiano e nova avaliação 48 horas após o tratamento. O perfil eletroforético em gel de agarose das proteínas séricas das cabras, apresentaram cinco frações, sendo: albumina e globulinas (a, b1, b2 e g). Houve aumento na produção de g-globulina e menor produção da fração b2-globulina 12 horas após a infecção, com os valores reduzindo mais rapidamente no grupo suplementado, evidenciando a influência da vitamina E na diminuição da produção das proteínas de fase aguda. Não houve influência da vitamina E na concentração sérica de imunoglobulina G (IgG) nos animais suplementados. A suplementação com vitamina E aumentou a concentração de imunoglobulinas e diminuiu a produção de proteínas de fase aguda, provavelmente pelo efeito antioxidante minimizando a lesão tecidual durante o processo inflamatório localizado na glândula mamária.

E-mail: leydsonf@fmva.unesp.br O presente trabalho visou avaliar a vitalidade e os níveis séricos de cortisol de bezerros neonatos, nascidos de partos eutócicos e auxiliados, bem como determinar os índices de partos distócicos em vacas receptoras de embriões nelores. Cerca de 310 bezerros foram avaliados. Observou-se a ocorrência de 38 partos distócicos (38/310) e de oito abortos (8/310). Os bezerros nascidos de partos distócicos mais duradouros, entre quatro e seis horas, apresentaram os maiores valores médios de frequência respiratória e dos níveis séricos de cortisol. A temperatura retal foi mais elevada em animais nascidos de partos laboriosos do que naqueles nascidos de partos normais. As taxas de morbidade e mortalidade em animais neonatos nelores foram de 77,93% e 3,44%, respectivamente. O emprego do escore APGAR, ao nascimento, demonstrou que 90% dos bezerros apresentavam-se deprimidos, independente do tipo de nascimento.

E-mail: leydsonf@fmva.unesp.br The purpose of present study was to evaluate the vitality and the serum levels of cortisol from newborn calves by normal and laborious deliveries, and to obtain the distocia register in Nelore embryo recipient cows. Three hundred ten calves were evaluated. The register of 38 dystocic calvings (38/310) and 8 abortions (8/310) were observed. In newborn animals the morbity and mortality rates were 77.93% and 3.44%, respectively. Calves born after long dystocic calving, lasting 4-6 hours, showed the highest mean values of respiratory frequency and serum cortisol levels. Animals born after laborious calving presented higher rectal temperature than those born at normal calving. The APGAR score showed that 90% calves had signs of dullness independent of parturition type.

Serum activities of AST and GGT were increased and serum activity of FA decreased. Serum values of urea and creatinine were within normal values. Hypoglycemia and hyperbilirrubinemia were also observed. These results can be used for experimental studies with the disease, for treatments trials, and to detect early cases of the disease without clinical signs. Otherwise the hematologic and blood biochemistry values of healthy sheep can be used as reference values for hair sheep in the semi-arid region of Brazil.

The total serum (TSP) and plasma (TPP) proteins, gamaglutamiltransferase activity (GGT), serum albumin, alpha, beta and gammaglobulin by electrophoresis in agarose gel and IgG estimated by the zinc sulphate turbidity test were determined. Two-way-ANOVA was used to compare the data in the first age. The age behaviour was analysed through repeated measures ANOVA. Correlations were established between the variables. The passive transfer of immunity was successful in both breeds and the parity of the dam had no effect on the calves’ serum gammaglobulin concentration. The levels of gammaglobulins were higher at the end of the first day of life, and decreased until 60 days. From there on, the increase due to the active production of antibodies was precocious in taurine calves and slower in Zebu calves. At the end of the first day of life, the gammaglobulin was correlated with IgG (r=0,859), TPP (r=0,807), TSP (r=0,811) and GGT (r=0,399). The variation of serum proteins followed the normal pattern throughout the first four months of life, with little differences between taurine and Zebu calves.

Twelve 5-month-old male lambs maintained on alfalfa hay (3% BW) and non-fluorinated water ad libitum were used. Animals were allocated into Control, receiving 5g NaCl/animal/day + 0.2mg I/kg dry matter) and Treated group, receiving the same treatment plus sodium fluoride (4.7mg F/kg body weight). Mineral treatment was given by gavage, daily for 150 days. Blood, urine and fecal samples were collected during and the end of the experiment. At the end of treatment period animals were euthanized and kidney, pineal and bone samples were collected. Urine F was higher in treated animals throughout the experiment. Bone F levels were also increased in treated animals; pineal F content however, was not different between groups. Kidney histology revealed no differences. It is concluded that chronic F administration induces accumulation of the element in the skeleton. However such fact appears not to be detrimental to animals. Rates of F accumulation in bone and urine excretion obtained in other species can not be used in growing lambs.

#69 - Clinical and laboratorial evaluation of Nellore cattle experimentally infected with Trypanosoma vivax, 21(4):157-161

[Clinical and laboratorial evaluation of Nellore cattle experimentally infected with Trypanosoma vivax] Avaliação clínico and laboratorial de bovinos nelore infectados experimentalmente com Trypanosoma vivax. Pesquisa Veterinária Brasileira 21(4):157-161. Embrapa Gado de Corte, Cx. Postal 154, Campo Grande, MS 79002-970, Brazil. In arder to evaluate the clinical-laboratorial alterations, six Neil ore calves were inoculated with 107 Trypanosoma vivax isolated from Poconé region, Mato Grosso, Brazil. The animals were evaluated daily for rectal temperature, packed cell volume (PCV), parasitemia, antibody production, calor of mucous membranes, behavior and appetite. Blood and serum samples for biochemical evaluation for aspartate aminotransferase (AST), alkaline phbsphatase (AF), gamma glutamyltransferase (GGT), cholesterol, urea, creatinine, creatine kinase (CK), calcium, phosphorus and proteinogram were collected on days 4, 8, 12, 16, 23 and 30 post inoculation (DPI). During the following 6 months rectal temperature, PCV and parasitemia were evaluated weekly. T. vivax was evidenced from 1 DPI in all calves and persisted until day 30 in five of six animals. A remarkable decrease (p<0.05) of PCV mean value (25%) was observed on 10 DPI. The animals presented no alterations in their clinical or serum biochemical state during the trial. Seroconversion took place 6 and 8 DPI, and all the animals remained seropositive during the 30 days of experiment. In all the experimental animals the occurrence of T. vivax infection was verified, characterized by the increase of corporal temperature, presente of the blood protozoa and reduction of the globular volume, without altéations in the other variables analyzed. Nellore calves, when experimentally inoculated with T. vivax, are able to establish a balance between host-parasite relationship.

Os animais foram observados diariamente, durante 30 dias, quanto aos parâmetros de temperatura retal, volume globular (VG), parasitemia, produção de anticorpos, coloração de mucosas, comportamento e apetite. Deteminaram-se os níveis séricos de aspartato aminotransferase (AST), fosfatase alcalina (FA), gama glutamiltransferase (GGT), creatina kinase (CK), colesterol, uréia, creatinina, cálcio, fósforo e o perfil eletroforético das proteínas séricas aos 4, 8, 12, 16, 23 e 30 dias pós-inoculação (DPI). Durante os 6 meses seguintes, os animais foram observados semanalmente, avaliando-se a temperatura retal, o VG e a parasitemia. T. vivax foi evidenciado a partir do terceiro e quarto DPI em todos os bezerros e persistiu até o 30º DPI em cinco dos seis animais em estudo. Ocorreu um decréscimo significativo (p<0,05) do valor médio do VG (25%) aos dez DPI. Os animais não apresentaram qualquer alteração no quadro clínico, bem como na avaliação da bioquímica sérica durante o período experimental. A soroconversão ocorreu aos 6 e 8 DPI, permanecendo todos os animais soropositivos nos 30 dias experimentais. Bovinos nelores jovens, infectados experimentalmente com T. vivax, foram capazes de estabelecer um equilíbrio na relação hospedeiro-parasita.


#70 - Clinical pathological data and analyses of mineral elements of cattle affected by epizootic botulism in the State of São Paulo

Abstract in English:

In arder to study laboratorial aspects of beef cow mortality, a syndrome popularly known as "doença da vaca caída", examens were made of blood, cerebrospinal fluid, serum, bone and liver samples from 32 naturally affected 4 to 9 year old cows, 27 belonging to the Nellore breed and5 were crossbred Nellore, all originating from farms located in municipalities near Botucatu, State of São Paulo. Laboratory determinations were analysed by descriptive statistics and included hematological values, total plasma protein, plasma fibrinogen, cerebrospinal fluid analysis, and concentration measurements of serum calcium, phosphorus, magnesium, sodium, potassium, chloride, total protein, albumin, globulin, alkaline phosphatase, aspartate aminotransferase, gama-glutamyltransferase and creatine kinase activities, included bane ash percentage and concentrations of calcium, phosphorus and magnesium, and also hepatic levels of copper, zinc, iron, manganese and cobalt. In addition, mouse bioassays and complement micro-fixation tests were performed to detect botulinum toxins in liver samples. The results indicated leukocytosis (13,3±3,9 x103/mm3) with neutrophilia (8,9±3,2 x103/mm3), hypocalcemia (7,8±1,7mg/dl), hypophosphatemia (3,6±1,6mg/dl), hypoalbuminemia (2,9±0,9g/cll), increased creatine kinase activity (691,0±829,7 UI/1), and reduced ash percentage (60,3±1,9%) and low phosphorus (17,2±0,4%) in bane. The other values were all within normal limits. The diagnosis of botulism, involving type C and D toxins, was confirmed as the cause of the mortality in the region of study, what is strongly consistent with the other laboratorial findings.

Com o objetivo de verificar alguns aspectos laboratoriais da mortalidade de vacas de corte, popularmente conhecida como "doença da vaca caída", estudaram-se amostras de sangue total, líquor, soro sanguíneo, osso e fígado colhidas de 32 fêmeas bovinas (27 da raça Nelore e 5 mestiças), entre 4 e 9 anos de idade, naturalmente acometidas e pertencentes a propriedades rurais localizadas em municípios próximos a Botucatu, Estado de São Paulo. Procederam-se as determinações e o estudo estatístico descritivo das variáveis: 1) hematológicas, de proteínas plasmáticas totais e fibrinogênio; 2) dos aspectos físicos, bioquímicos e celularidade no líquor; 3) de cálcio, fósforo, magnésio, sódio, potássio; cloretos, proteína total, albumina, globulina e atividades da fosfatase alcalina, aspartato aminotransferase, gama-glutamiltransferase e creatina quinase no soro sanguíneo; 4) de cinzas, cálcio, fósforo e magnésio no osso, e, 5) de cobre, zinco, ferro, manganês e cobalto no fígado, empregando-se as provas de inoculação em camundongos e de microfixação de complemento para a detecção da presença de toxinas botulínicas nas amostras de tecido hepático. Os resultados apontaramcomo alterações, leucocitose (13,3±3,9 x103/mm3) com neutrofilia (8,9±3,2 x103/mm3), hipocalcemia (7,8±1,7mg/ dl), hipofosfatemia (3,6±1,6mg/dl), hipoalbuminemia (2,9±0,9g/dl), elevação na atividade da creatina quinase (692,0±829,7 UI/1) e reduções na cinza (60,3±1,9%) e fósforo (17,2±0,4%) ósseos, como indicativos do estado carencial de fósforo, apresentando-se as demais variáveis dentro dos limites de variação considerados normais para a espécie. O diagnóstico de botullsmo, com envolvimento das toxinas C e D, foi confirmado como a causa da mortalidade investigada na área de abrangência do estudo, e encontra coerência com o conjunto de resultados.
