E-mail: alexsantos@usp.br The connexin 32 (Cx32) is a protein that forms the channels that promote the gap junction intercellular communication (GJIC) in the liver, allowing the diffusion of small molecules through cytosol from cell-to-cell. Hepatic fibrosis is characterized by a disruption of normal tissue architeture by cellular lesions, and may alter the GJIC. This work aimed to study the expression and distribution of Cx32 in liver fibrosis induced by the oral administration of dimethylnitrosamine in female Wistar rats. The necropsy of the rats was carried out after five weeks of drug administration. They presented a hepatic fibrosis state. Sections from livers with fibrosis and from control livers were submitted to immunohistochemical, Real Time-PCR and Western-Blot analysis to Cx32. In fibrotic livers the Cxs were diffusely scattered in the cytoplasm, contrasting with the control livers, where the Cx32 formed junction plaques at the cell membrane. Also it was found a decrease in the gene expression of Cx32 without reduction in the protein quantity when compared with controls. These results suggest that there the mechanism of intercellular communication between hepatocytes was reduced by the fibrotic process, which may predispose to the occurrence of a neoplastic process, taken in account that connexins are considered tumor suppressing genes.

Sheep with conidiobolomycosis had nonregenerative, normocytic and normochromic anemia, leucocytosis with neutrophilia and increased nutrophil:lymphocyte rate, moderate monocytosis, moderate thrombocytosis, hypoproteinemia (hypoglobulinemia), hypomagnesemia, hypocalcemia and increased values of phosphorus. Serum activities of AST and GGT were increased and serum activity of FA decreased. Serum values of urea and creatinine were within normal values. Hypoglycemia and hyperbilirrubinemia were also observed. These results can be used for experimental studies with the disease, for treatments trials, and to detect early cases of the disease without clinical signs. Otherwise the hematologic and blood biochemistry values of healthy sheep can be used as reference values for hair sheep in the semi-arid region of Brazil.

These plants can be placed in three major groups: plants causing acute liver necrosis; plants causing liver fibrosis; and plants causing hepatogenous photosensitization. For some of these plants the toxic principles are known. Cestrum parqui and Xanthium cavanillesi that cause acute liver necrosis contain carboxy-atractylosides. Senecio spp., Crotalaria spp., and Echium plantagineum that cause liver fibrosis contain pyrrolizidine alkaloids. As for the group of plants causing hepatogenous photosensibilization, Myoporum spp. contain furanosesquiterpenes, Lantana spp contain triterpenes, and Brachiaria spp. and Panicum spp. contain steroidal saponins. The clinical and pathologic features of the toxicosis caused by these phytotoxins, general mechanisms of production for the production of the clinical signs and the methods for diagnosis of hepatic failure in farm animals are reviewed.

#54 - Spontaneous poisoning by larvae of Perreyia flavipes (Pergidae) in sheep, p.19-22

Spontaneous poisoning by larvae of Perreyia flavipes (Pergidae) in sheep. Pesquisa Veterinária Brasileira 28(1):19-22. Departamento de Patologia Clínica Veterinária, Universidade Federal do Rio Grande do Sul, Avenida Bento Gonçalves 9090, Porto Alegre, RS 91540-000, Brazil. E-mail: davetpat@ufrgs.br From a flock of 175 Texel sheep 25 animals died after consumption of a sawfly larvae subsequently identified as Perreyia flavipes. The disease occurred in June-July 2006 on a farm located in the county of Encruzilhada do Sul, Rio Grande do Sul, Brazil. Although there were 11 cattle in the same paddock, none of them was affected. High numbers of compact masses containing up to 150 larvae were scattered in the paddock where the animals were grazing. Most affected sheep showed severe apathy during 24-36 h before death, but weakness, muscular tremors and depression were also observed. Necropsy was performed on six sheep and the main macroscopic lesions were hemor-rhages in the subcutaneous tissues, endocardium, gallbladder wall, and abomasal mucosa. In all animals was found hydrothorax, hydropericardium, ascites, and mild jaundice. Edema in the abomasal folds, mesentery, perirenal tissues, and gallbladder wall were also seen. The livers were yellowish with disseminated pinpoint hemorrhages in the parenchyma and had an enhanced lobular pattern. Perreyia flavipes larval body fragments and heads were found in the forestomach contents of the six sheep. Feces were scant, dry and formed balls coated by mucus and streaks of blood. Similar contents were also present at the end of the cecum. Prominent microscopic lesions included severe and diffuse periacinar or massive necrosis of hepatocytes associated with multifocal random hemorrhages. Diffuse necrosis of lymphoid follicles in lymph nodes and Peyer´s patches, lymphoid depletion and necrosis in germinative centers of the spleen, and diffuse vacuolization in the renal tubular epithelia were also seen.

[Hepatic cirrhosis in dogs: 80 cases (1965-2003).] Cirrose hepática em cães: 80 casos (1965-2003). Pesquisa Veterinária Brasileira 27(11):471-480. Considering those cases in which the sex and age of the dogs were informed in the necropsy reports, males comprised 53.8% of the cases and 46.2% were females; while 50.0% of these dogs were aged, 48.6% were adults and 1.4% were puppies. The main observed clinical signs include ascites (39/80 [48.8%]), icterus (19/80 [23.8%]), anorexia (13/80 [16.2%]), neurological disturbances (12/80 [15.0%]), dyspnea (12/80 [15.0%]) and subcutaneous edema (10/80 [12.5%]). In 63 dogs for which there was a description of gross morphology in the necropsy reports, 76.2% had macronodular and 23.8% had micronodular cirrhosis. ABSTRACT.- Inkelmann M.A., Rozza D.B., Fighera R.A., Kommers G.D., Graça D.L., Irigoyen L.F. & Barros C.S.L. 2007. [Infectious canine hepatitis: 62 cases.] Hepatite infecciosa canina: 62 casos. Pesquisa Veterinária Brasileira 27(8):325-332. Departamento de Patologia, Universidade Federal de Santa Maria, 97105-900 Santa Maria, RS, Brazil. E-mail: claudioslbarros@uol.com.br Necropsy reports from 5,361 dogs necropsied over a 43-year period (1964-2006) were reviewed in search for cases of infectious canine hepatitis (ICH). Sixty two (1.2%) cases of the disease were found. Most of the 62 affected dogs (91.9%,) were 2-year-old or less. Clinical signs were recorded in the necropsy files of 45 ICH affected dogs and included anorexia (55.6%), apathy (35.6%), diarrhea (35.6%) (often with blood [43,8%]), neurological signs (33.3%), vomiting (26.7%), petechiae and echymosis in the mucous membranes and/or skin (24.4%), hypothermia (20.0%), abdominal pain (15.6%), icterus (13.3%), enlargement and congestion of the tonsils (11.1%), fever (11.1%) and ascites (6.7%). The clinical courses lasted from few hours to 15 days. The most frequent necropsy findings included hepatic changes (87.1%), edematous, congested and hemorrhagic lymph nodes (51.6%), bloodstained fluid, clear fluid or whole blood in the abdominal cavity (35.5%), and petechial or paint-brush hemorrhages over the pleural (27.4%) and gastrointestinal (24.2%) serosal surfaces. In 12.9% of the cases there was a granularity to the intestinal serosa. Hemorrhages in the leptomeninges and in the substance of the brain were observed in 9.7% of the cases. Hepatic gross changes included moderately enlarged and more friable livers with marked lobular pattern, congestion and multifocal pale or hemorrhagic foci of necrosis. Films and strands of fibrin covered the hepatic surface in 20.4% of the cases and in 27.8% of the cases the gall bladder was thickened by edema. Zonal or randomly distributed multifocal hepatic necrosis (93.5%) associated with intranuclear inclusion bodies were the most consistent microscopic findings. Intranuclear inclusion bodies were found in the liver in every case and their detection was the criterium for confirmation of the diagnosis. The most significant microscopic extra-hepatic lesions included hemorrhages and intranuclear inclusion bodies in endothelial and reticuloendothelial cells of the renal glomeruli (50.0%) lymph nodes (47.8%), brain (27.8%), tonsils (25.0%) and spleen (10.0%).

Histopathological changes in liver and gill epithelium of Nile tilapia, Oreochromis niloticus, exposed to waterborne copper. Pesquisa Veterinária Brasileira 27(3):103-109. Liver and gill samples were collected after 21 days of exposure to copper and lesions were analyzed by light microscopy. The main histopathological changes observed in gills exposed to the highest concentration were edema, lifting of lamellar epithelia and an intense vasodilatation of the lamellar vascular axis. Although less frequent, lamellar fusion caused by the filamentar epithelium proliferation and some lamellar aneurisms were also found. The liver of control group exhibited a quite normal architecture, while the fish exposed to copper showed vacuolation and necrosis. These hepatic alterations were more evident in fish exposed to 1.0 and 2.5mg L-1 copper concentrations. The number of hepatocytes nucleus per mm2 of hepatic tissue decreased with the increase of copper concentration. In contrast, the hepatic somatic index was high in fish exposed at 2.5mg L-1 of copper. In short, this work advance new knowledge as influence of copper in the gill and liver histology of O. niloticus and demonstrated that their effects could be observed at different concentrations.

Laboratory tests of liver function are not always reliable indicators of subclinical affected animals. Liver biopsy could be useful to identify cattle with hepatic lesions but without clinical signs and would have also a prognostic value since it is generally believed that hepatic lesions will eventually cause liver failure and death. Such animals could be picked out by liver biopsy before clinical signs develop and be sent to slaughter, minimizing economic losses. This study was aimed to evaluate the liver biopsy as a diagnostic and prognostic tool in cases of Senecio spp. poisoning in cattle. An outbreak of Senecio brasiliensis was diagnosed in dairy calves which ingested hay contaminated by 5-10% of this Senecio species. Liver biopsy using a Menghini needle by right transthoracic approach was carried out in 135 calves that ingested the contaminated hay. Biopsed calves were followed up for 26 months after the biopsy. Seventeen biopsied calves had typical lesions of Senecio spp. poisoning (positive calves) and 118 had histologically normal livers (negative calves). Hepatic lesions of positive calves included fibrosis, hepatomeglocytosis, and biliary hyperplasia. Fifteen out of the 17 positive calves died with typical clinical signs of Senecio spp. poisoning within 17-149 days after the biopsy; 13 of those were necropsied and had typical gross and histopathological lesions of Senecio spp. poisoning. Two positive calves were clinically normal at the end of the post-biopsy observation period. The prognostic value (sensibility) of the test was considered high since 88.23% of the positive calves died. The specificity of the test was considered very high (99.16%) since only one of the 118 negative calves died in the observation period. In none of the biopsed calves a negative effect related to the biopsy technique was observed.

The main gross lesions were observed in the liver, and included friable consistency, pronounced lobular pattern and areas of haemorrhages. The liver of one bovine was homogeneously dark reddened. Petechial hemorrhages in serosal membranes and edema in the gall bladder were frequently seen. Pale kidneys with red spots in the cortex were observed in one animal. Microscopically, the most striking lesion in the liver was massive coagulative necrosis, associated with centrolobular haemorrhages, observed in four animals. In the liver of one bovine centrolobular necrosis was observed . Tubular renal necrosis was noted in two animals. Additional microscopic lesions were found in the central nervous system of five bovines, especially in the frontal cortex, and included perineuronal and perivascular edema with basophilia and retraction of the neurons. T T. micrantha caused clinical signs with 50g/kg and death with doses of 54g/kg or higher. The fractionated administration of the green leaves as well as the dried leaves did not cause poisoning.

E-mail: bonel-raposo@brturbo.com Green leaves of Myoporum laetum were collected during spring and summer, and administered to five sheep and six steers at dosages of 20 and 30 g/kg. Liver biopsies were taken before (Controls) and 1, 3 and 7 days after dosage. ln sheep, the clinicai signs were depression, rumen hypomotility, dried feces, tenesmus, teeth grinding, dyspnea and typical lesions of photosensitization. ln cattle, the clinicai picture was much less pronounced. The main histological findings in sheep were vacuolization of hepatocytes, portal ftbrosis, bile duct proliferation and necrosis of periportal hepatocytes; the ultrastructural examination revealed hyperplasia of the smooth endoplasmic reticulum, hepatocellular hydropic degeneration, presence of crystals and severa! other degenerative changes. ln cattle both, the histological and the ultrastructural findings, were less evident.

Pesquisa Veterinária Brasileira 23(4):149-155. Depto Patologia Animal, Faculdade de Veterinária, UFPel, Pelotas, RS 96010-900, Brazil. E-mail: bonel-raposo@brturbo.com Amostras de Myoporum laetum foram colhidas durante a primavera e verão e administradas a cinco ovinos e seis bovinos em doses únicas de 20 e 30 g/kg. Biópsias hepáticas foram colhidas antes (controles) e 1, 3 e 7 dias após a dosagem da planta. Estas biópsias foram analisadas histológica e ultraestruturalmente. Os sinais clínicos, em ovinos, caracterizaramse, especialmente; pordepressão, diminuição dos movimentos ruminais, fezes ressequidas, tenesmo, ranger de dentes, dispnéia e lesões típicas de fotossensibilização. Em bovinos, o quadro clínico foi discreto. Os principais achados histológicos, em ovinos, incluíram vacuolização de hepatócitos, fibrose portal, proliferação de duetos biliares e necrose de hepatócitos periportais. Os estudos ultra-estruturais, em ovinos, revelaram hiperplasia do retículo endoplasmático liso, tumefação de hepatócitos, degranulação e vesiculação do retículo endoplasmático rugoso, presença de cristais aciculares, retenção biliar, tumefação de mitocôndrias e várias outras alterações degenerativas. Em bovinos, tanto os achados histológicos, quanto os ultra-estruturais foram menos evidentes.
