Silva L.A.F., Fioravanti M.C.S., Trindade B.R., Silva O.C., Eurides D., Cunha P.H.J., Silva M.L. & Moura M.I. 2004.[Foot diseases in dairy cows: association with clinical mastitis, metrites and predisposing factors.] Postal 131, Goiânia, GO 74001-970, Brazil. E-mail: lafranco@vet.ufg.br With the objective to investigate a possible association between foot diseases, clinical mastitis and/or metritis and predisposing factors for foot diseases, 5300 dairy cows from 80 intensive and semi-intensive farms were used. In 325 (6.13%) cows only foot disease was diagnosed, in 35 (0.66%) foot disease and clinical mastitis, in 52 (0.98%) foot disease and metritis, in 28 (0.53%) foot disease, clinical mastitis and metritis, in 128 (2.42%) only metritis, in 165 (3.11%) only clinical mastitis, and in 89 (1.68%) cows metritis and clinical mastitis. Rapid changes in the diet, high exposure time of hoof horn to slurry and wet conditions, concrete floors, use of footbaths, low frequency of claw trimming, irregular quarantine, and no attention to health aspects during acquisition of the animals were considered as major risk factors for the occurrence of those diseases. The McNemar test for dependent samples showed significant statistical differences between the occurrence of foot disease, clinical mastitis and metritis, in spite of the poor association between those diseases. It was concluded that there was no expressive relationship between foot diseases, clinical mastitis and metritis in lactent cows.

E-mail: davetpat@vortex.ufrgs.br The study assessed differences of pathogenicity among 19 strains of Brachyspira pilosicoli isolated from cases of diarrhea in swine in the State of Rio Grande do Sul, Brazil, using an one-day-old chick model. The chicken inoculated with live bacterial cultures were submitted to euthanasia 21 days later, and the cecum was examined histologically using hematoxylin-eosin, silver staining and immunohistochemistry with a polyclonal antibody anti-Brachyspira pilosicoli. With silver staining and immunohistochemistry, respectively, 21.59% and 70.96% of the chicken showed colonization of the cecal epithelium with B. pilosicoli. Differences in the type of colonization characterized by continuous adhesion, focal adhesion or free bacteria in the intestinal lumen were observed. Immunohistochemistry was more efficient to assess intestinal colonization, because 49.37% more colonized chicken could be detected with silver staining. With three strains, elongated figures inside the cytoplasm of epithelial cecal cells in the inoculated chicken were observed.

E-mail: nslazaro@aol.com Thirty-eight strains of Salmonella Muenster, isolated from swine and the abattoir environment, in the State of Rio de Janeiro, Brazil, from March 1991 to February 1992, were analyzed for the presence of plasmids. The strains were selected according to their profile regarding the antimicrobials: streptomycin, tetracycline, sulphonamide and sulfametoxazole-trimethoprim. Thirteen strains were resistant to one or several antimicrobials, 18 with intermediate degree and seven were sensitive. Plasmids varying in size from 1.2 Kb to 42 Kb were detected in 37 (97.36%) of the 38 samples, corresponding to 11 different profiles (P1- P11), varying from 1 to 6 plasmids per model. The number and plasmids diversity was greater than the resistance marks for strains. The plasmid of 2.85 Kb was the most frequent, being present in 83.78% of the 37 strains; only the one of 7.5 Kb was detected at the two slaughterhouses. There was no parallelism between resistance pattern and plasmidial profile, and a same antibiotype was found in several plasmidial profiles. The results of the present investigation, allowed us to conclude that the plasmid characterization is an useful and simple tool for the epidemiological typing of this sorovar.

#364 - Comparison of the sensibility of cattle and buffaloes to poisoning by Palicourea marcgravii (Rubiaceae), 23(4):167-172

Abstract in English:

ABSTRACT.- Barbosa]. D., Oliveira C. M. C., Tokarnia C. H. & Riet-Correa E 2003. [Comparison of the sensibility of cattle and buffaloes to poisoning by Palicourea marcgravii (Rubiaceae).] E-mail: diomedes@ufpa.br In order to verify if buffaloes are more resistant than cattle to poisoning by Palicourea marcgravii, the plant was given per os simultaneously to buffaloes and cattle. Seven buffaloes and three head of cattle were used. Doses of 0.5, 1.0 and 2.0 g/kg did not cause signs of poisoning in buffaloes; but doses of 3.0, 4.0 and 6.0 g/kg caused death. In cattle 0.25 g/kg did not cause symptoms; but 0.5 and 2.0 g/kg were lethal. The influence of exercise on the onset of symptoms, the time which elapsed from the beginning of the administration of the plant to the appearance of symptoms, and the symptoms themselves, were similar in the two animal species. The clinical course was longer in buffaloes; whilst in cattle the period between the onset of severe symptoms and death lasted 9 to 17 minutes; in buffaloes it varied from 10 minutes to 1 hour 28 minutes. It can be concluded, that buffaloes are about six times more resistant than cattle to the toxic action of P. maracgravii. The lower incidence of deaths in buffaloes by poisonous plants in the Amazon region is, at least in part, due the lower sensibility of buffaloes to poisoning by this plant. Another factor responsible for the smaller number of buffaloes which die due to plant poisoning in the Amazon region, is possibly that buffaloes prefer the Jowlands, e.g. temporarily flooded areas, which are the habitat of Arrabidaea bilabiata, the second most important poisonous plant of the Amazon region. This plant is less poisonous than P. marcgravii, and its habitat are the higher not flooded areas. So it would be safer, in order to diminish losses, to breed buffaloes instead of cattle in areas where P. marcgravii occurs. The reason for the greater resistance of buffaloes should be further investigated as a possible prophylactic measure to avoid poisoning by P. marcgravii in cattle. On the other hand, Australian investigators modified genetically the ruminai bacteria Butyrivibrio fibrisolvens by introducing a gene, isolated from a Moraxella sp, which codifies a dehalogenase, capable of hydrolyzing fluoroacetate, the poisonous principle of P. marcgravii. Transferring the genetically modified B. fibrisolvens into the rumen of animal, which consume plants that contain fluoroacetate, would be a viable method to control that poisoning through a ruminal detoxifying process for the toxin. Through preliminary contacts, the director of the consorcium in Australia, responsible for the modification of the bacteria, declared that he is interested to sell the technology to our country, but it would be necessary to know if it is permitted to introduce into Brazil the genetically modified bacteria. If allowed, it would be necessary to perform investigations on the viability and the methodology for using this bacteria to control P. marcgravii poisoning in our country.

RESUMO.- Barbosa]. D., Oliveira C. M. C., Tokarnia C. H. & Riet-Correa E 2003.

Abstract in English:

ABSTRACT.- Vianni M.C.E. & Lázaro N.S. 2003. [Profile of antimicrobial susceptibility in strains of Gram positive cocci, negative catalase, isolated from buffalo subclinical mastitis.] The susceptibility of antimicrobials was studied in Gram positive and catalase negative cocci (21 samples of Lactococcus garvieae and 6 Enterococcus gallinarum), isolated from the milk of cows with subclinical mastitis, belonging to six buffalo herds in the State of Rio de Janeiro. The test used was diffusion of disks in agar Müller Hinton, according to recommendations of the National Committee for Clinicai Laboratory Standards - NCCLS. There were tested disks with ampicillin (10mg), cefalotin (30mg), cefotaxime (30mg), cefoxitin (30mg), doranfenicol (30mg), eritromycin (15mg), gentamycin (10mg), nitrofurantoin (300mg), norfloxacin (10mg), penicillin (1 O IU), tetracydin (30mg) and vancomycin (30mg). The results showed that with Lactococcus garvieae, the most efficient antimicrobial was nitrofurantoin, revealing 85.71% sensibility, followed by cefotaxime (61.90%), vancomycin (52.38%), norfloxacin (47.62%) and cefalotin (47.62%). The highest resistance was developed against penicillin and ampicillin, with 95.24% resistance for the two antimicrobials. The susceptibility profile developed by the strains of Enterococcus gallinarum showed low sensibility against the tested antimicrobials; the highest resistance observed was against eritromycin and gentamycin, with 33.34% sensibility for both. The antimicrobial evaluation showed 100% resistance against vancomycin and tetracyclin, followed by cloranfenicol, penicillin, ampicillin, cefoxitin, cefotaxim, norfloxacin and nitrofurantoin; all of them showed a resistance of 83.33% with the samples tested.

#366 - Clinical and hematological evaluation of Nelore calves experimentally infected with isolates of Babesia bigemina from the Southeastern, Northeastern and Northern regions of Brazil, 23(2):52-60

Abstract in English:

ABSTRACT.- Mendonça C.L., Vieira D., Kohayagawa A., Schenk M.A.M., Madruga C.R. & Afonso J.A.B. 2003. [Clinical and hematological evaluation of Nelore calves experimentally infected with isolates of Babesia bigemina from the Southeastern, Northeastern and Northern regions of Brazil.] Fed. Rural de Pernambuco, Cx. Postal 152, Garanhuns, PE 55292-901, Brazil. A comparative study was made regarding the clinical and hematological alterations caused by isolates of Babesia bigemina from southeastern, northeastern and northern Brazil in experimentally infected Nelore calves. Eighteen calves between 7 and 9 months of age, without antibodies against Babesia sp and raised free from ticks, were used. Three animais were previously inoculated with 2.0x109 parasitic erythrocytes (PE) for each stabilate. The other 15 calves were subdivided into three groups, with five animais each, that were subinoculated with 1.0x1010 PE of the respective isolates. The clinical and hematological alterations were evaluated by the determination of parasitaemia, haemogram, plasmatic fibrinogen, reticulocyte count, descriptive analysis of the bone marrow and erythrocytic osmotic fragility, for 30 days, totalizing seven moments of observation. The follow-up of the immunological response by the indirect fluorescent antibody test was carried out daily until the 10th day after inoculation (DAI) and after that, on the 151 20th, 25th and 30th DAI. A mild clinical manifestation of the disease was observed. The laboratory findings revealed low leveis of parasitaemia; decrease of the erythrogram values; absence of reticulocytes, initial decrease in the total count of leukocytes, neutrophils and lymphocytes with a posterior elevation of the number of these cells; hypercellularity of the erythrocytic series and decrease of the myeloid: erythroid relation which was more accentuated between the 8th and 12th DAI, and an increase of the erythrocytic osmotic fragility in the groups inoculated with the Southeast and Northeast isolates. None of the three isolates of B. bigemina gave rise to the clinical characteristic form of the disease, although they induced an humoral immune response.

RESUMO.- Mendonça C.L., Vieira D., Kohayagawa A., Schenk M.A.M., Madruga C.R. & Afonso J.A.B. 2003. [Clinical and hematological evaluation of Nelore calves experimentally infected with isolates of Babesia bigemina from the Southeastern, Northeastern and Northern regions of Brazil.]

E-mail: proehe@orion.ufrgs.br In order to determine the capacity of bovine herpesvirus type 1 and 5 (BHV-1 and BHV-5) to invade, multiply and spread along the central nervous system (CNS) (neuroinvasiveness), as well as their potential to induce neurological illness (neurovirulence), 30 to 35 days old rabbits were inoculated with the BHV-5 strain EVI 88 / 95 and Los Angeles and Cooper BHV-1 strains, by the intrathecal (IT) and intranasal (IN) routes. The BHV-5 strain induced severe neurological clinical signs in 100% (12/12) of the rabbits inoculated by both routes. Histopathological examination revealed multifocal non-suppurative meningoencephalitis, characterized by multifocal gliosis and perivascular cuffing. Virus was recovered from many parts of the brain. Both BHV-1 strains, when inoculated via 1T route, were not neurovirulent. The strain Los Angeles, after IN inoculation, induced signs of severe respiratory disease (7/7), as well as signs of neurological impairment, indistinguishable from those induced by BHV-5, in 57% (4/7) of the infected rabbits. However, the rabbits with nervous signs revealed at histopathology vasculitis and thrombosis in lungs and brain, the latter with foci of neuronal necrosis, but no lesions indicative of encephalitis, suggesting that neural damage was probably consequent to tissue anoxia. The BHV-1 strain Cooper, after IN inoculation, induced only mild signs of respiratory disease. These findings indicate that the BHV-5 strain was both neuroinvasive and neurovirulent, since it was capable of invading, spreading and multiplying in the rabbits brains by both routes of inoculation, yet causing neurological disease, apparently consequent to vírus induced neural damage. The BHV-1 Los Angeles strain was not neuroinvasive, whereas its neurovirulence was probably consequent to tissue anoxia, which histologically seemed not to be related to direct viral pathogenic effect. The BHV-1 strain Cooper was neither neurovirulent nor neuroinvasive for rabbits. It is possible that these observations bear relationship with the frequent association of BHV-5 with encephalitis in cattle, as opposed to BHV-1 encephalitis, which is a rare event in nature.

ABSTRACT.- Leite L.G., Riet-Correa F., Medeiros R.M.T., Piacenti A., Aragão M. & Schons S.V. 2002. Susceptibility of laboratory animals to the intoxication by Ateleia glazioviana (Leg. Papilionoideae).] Papilionoideae). Pesquisa Veterinária Brasileira 22(2):73-78. Laboratório Regional de Diagnóstico, Faculdade de Veterinária, Universidade Federal de Pelotas, 96010-900 Pelotas, RS, Brazil. E-mail: lgmv@alternet.com.br Rabbits, guinea pigs, rats and mice were used to reproduce the cardiac lesions observed in ruminants in the intoxication by Ateleia glazioviana. Four animals of each species were fed during four months with a ration containing 10% of dry A glazioviana. The pellets were made by mixing 700 g of commercial ration, 200 g of cornstarch, 1000 mi of distilled water and 100g of the dry plant. Later this mixture was dried at 100ºC during 16-20 hours. Four control animals of each species were given similar food, but containing dry ryegrass (Lolium multiflorum) instead of A glaziovidna. Water was offered ad libitum and a daily amount of green oats (Avena sativa) was given to rabbits and guinea pigs. No clinical signs of disease were observed during the experiment. Ali animals were weighted weekly in the last 5 weeks of the experiment and no differences were observed in weight gains between treated and control groups. At the end of dosing ali animals were euthanatized and necropsied. No macroscopic or histologic lesions were observed in the heart or other organs. The abortive properties of A glazioviana were studied in rats. One group of 11 pregnant rats was fed, from day 7 to day 21 of gestation, ration containing 10% of A. glazioviana, prepared in the sarne way than in the previous experiment. A control group received the ration with 10% ryegrass. Ali rats delivered normal litters. To determine a possible loss of toxicity of the plant during the pellet preparation, dry A. glazioviana was heated for 16-20 hours at 100ºC and administered orally to one ewe. This sheep ingested daily 2.65 g/kg bw of dry A. glazioviana for 26 days. During the experimental period the animal had tachycardia and cardiac arrhythmia. At the end of the experiment bradycardia and severe depression were observed. It was euthanatized 7 days after the last plant administration. The sheep was 2.5-3 months pregnant and the fetus was apparently normal with no autolysis. Ascites and hydrothorax were observed at necropsy. Whitish areas were observed in the myocardium and the liver was pale. Degeneration and necrosis of cardiac fibers and proliferation of fibrous tissue were observed histologically in the heart. The liver had centrilobular degeneration of hepatocytes and congestion. These experiments indicate that A. glazioviana is not cardiotoxic for laboratory animals and suggest that the plant has a heat-stable active principie similar to the active principie of plants causing cardiac fibrosis (gousiekte) in Southern Africa.

RESUMO.- Leite L.G., Riet-Correa F., Medeiros R.M.T., Piacenti A., Aragão M. & Schons S.V. 2002. Susceptibility of laboratory animals to the intoxication by Ateleia glazioviana (Leg. Papilionoideae).]
