#21 - Transferência de imunidade passiva em bezerros das raças Nelore e Limousin e proteinograma sérico nos primeiros quatro meses de vida, p.410-416

E-mail: janlisboa@uel.br To study the passive transfer of immunity, 90 healthy calves, 45 Nelore and 45 Limousin, were grouped (n=15) according to their dam’s parity: first, second or third or more calvings. Blood samples were draw from each calf with 24 to 36 hours of life and 15, 30, 60, 90, and 120 days. The total serum (TSP) and plasma (TPP) proteins, gamaglutamiltransferase activity (GGT), serum albumin, alpha, beta and gammaglobulin by electrophoresis in agarose gel and IgG estimated by the zinc sulphate turbidity test were determined. Two-way-ANOVA was used to compare the data in the first age. The age behaviour was analysed through repeated measures ANOVA. Correlations were established between the variables. The passive transfer of immunity was successful in both breeds and the parity of the dam had no effect on the calves’ serum gammaglobulin concentration. The levels of gammaglobulins were higher at the end of the first day of life, and decreased until 60 days. From there on, the increase due to the active production of antibodies was precocious in taurine calves and slower in Zebu calves. At the end of the first day of life, the gammaglobulin was correlated with IgG (r=0,859), TPP (r=0,807), TSP (r=0,811) and GGT (r=0,399). The variation of serum proteins followed the normal pattern throughout the first four months of life, with little differences between taurine and Zebu calves.

E-mail: febencli@usp.br The immunoglobulin variation profile in lavages from the broncoalveolar and tracheo-bronchial regions of 20 healthy newborn Holstein male calves was studied. They were fed with colostrum and distributed into 2 groups, 10 animals each. Group 1 underwent the nasotracheal catheterization technique to get the bronchoalveolar lavage (BAL), and Group 2 underwent the tracheocenthesis to collect the tracheobronchial lavage (TBL), both procedures being carried out at a 7-day-interval, starting on the first days up to about one month of life. Higher IgG contents, as compared to IgA, were noted across the respiratory tract. These immunoglobulins were impacted by the site of the respiratory tract washed, as well as by the calves’ life time in weeks. Higher immunoglobulin contents were detected in TBL, as well as higher IgM and IgA rates, as compared to BAL. The BAL immunoglobulins showed a tendency to be reduced in TBL.

#23 - Imunidade passiva, morbidade neonatal e desempenho de cabritos em diferentes manejos de colostro, p.219-224

[Passive immunity, neonatal morbidity and performance of kids in different colostrum management.] Imunidade passiva, morbidade neonatal e desempenho de cabritos em diferentes manejos de colostro. Pesquisa Veterinária Brasileira 25(4):219-224. Centro de Ciências Agrárias, UFPB, Campus III, Areia, PB 58397-000, Brazil. E-mail: saravilar@bol.com.br The experiment was performed to determine the kid management that ensures the best passive immunity, and to evaluate the relationship between passive immunity, neonatal morbidity and performance. Blood samples were obtained from 58 Saanen kids before colostrum ingestion and 30h after. The kids were submitted to five treatments: (T1) kids were allowed to nurse the dam for 24 hours ingesting colostrum ad libitum; (T2) kids were bottle-fed with 200mL colostrum in the first hour of life; (T3) kids were bottle-fed with 400 mL colostrum, 200mL in the first hour and 200mL 8 hours after birth; (T4) kids were bottle-fed with 400mL colostrum, 200mL in the first hour and 200mL 14 hours after birth; (T5) kids were bottle-fed with 600mL of colostrum, 200mL in the first hour, 200ml 12 hours after birth, and 200mL at 24 hours of life. Serum levels of gamaglobulins were determined by electrophoresis. The occurrence of diseases was monitored from birth to 28 days. Daily weight gains were used to evaluate the performance until weaning. The lowest concentration of g-globulin was observed in Group 2 (1,57g/dL) and the highest in Group 3 (2,60g/dL). In spite of higher levels of passive immunity Group 3 showed more animals with scours, however no statistical differences were observed. The immune status at 30 hours postpartum was not associated with neonatal weight gain.

#24 - Qualitative and semiquantitative studies on the immunoglubulin G-Fc-receptor activity of streptococci

Abstract in English:

These studies were conducted, in order to determine the nonimmune binding of human immunoglobulin (Ig) G to streptococci by qualitative and semiquantitative tests. In the qualitative slide method 28 (30.4%) of the 92 streptococcal cultures belonging to the serological group A bound IgG as well as 118 (50.2%) of the 235 group C cultures and 25 (31.6%) of the 79 group G cultures. The semiquantitative microtiter procedure revealed IgG-Fc-receptor activity in 31 (33.6%) of the 92 group A cultures, 171 (72.7%) of the 235 group C cultures and 32 (40.5%) of the 79 group G cultures in dilutions up to 1 :16000. Extraction of the streptococci by boiling or concentrated formic acid were suitable to obtain free IgG-Fc-receptors for the test. The semiquantitative microtiter procedure proved to be more sensitive for the demonstration of nonimmune IgG-binding to streptococci than the qualitative slide method and indicated the degree of IgG-Fc-receptor activity.

Foi conduzido um estudo qualitativo e semiquantitativo para verificar a incidência de Fc-receptores para imunoglobulina G (IgG) humana em Streptococcus spp. Na prova qualitativa em lâmina verificou-se Fc-receptor-atividade em 28 (30,4%) das 92 culturas do grupo sorológico A, em 118 (50,2%) das 235 culturas do grupo sorológico C e em 25 (31,6%) das 79 culturas do grupo sorológico G. Nas provas semiquantitativas em placas de microtitulação, 31 (33,6%) das 92 culturas do grupo A, 171 (82,7%) das 235 culturas do grupo sorológico C e 32 (40,5%) das 79 culturas do grupo sorológico G mostraram-se positivas, em diluições até 1 :16000. De um total de 611 culturas testadas, as provas semiquantitativas em placa de microtitulação detectaram 234 (38,3%) culturas positivas, enquanto a prova qualitativa em lâmina 171 (28,0%). Dentre as provas semiquantitativas, a extração por fervura das bactérias e o tratamento com ácido fórmico mostraram-se eficientes na detecção da IgG-Fc-receptor-atividade. Concluiu-se que as provas semiquantitativas em placas de microtitulação são eficientes na determinação do grau da IgG-Fc-receptor-atividade.
