#21 - Epidemiological, clinicopathological and immunohistochemical aspects of vulval squamous cell carcinomas in 33 cows

Abstract in English:

Vulvar squamous cell carcinomas (VSCCs) in cattle were retrospectively studied regarding the prevalence, epidemiology, clinicopathological, and immunohistochemical aspects. The degree of vulvar pigmentation was also evaluated. In the 48 years analyzed, necropsy and biopsy reports of 7,483 cattle were found. Out of these, 664 (8.87%) cases of various neoplasms were identified; 33 (4.97%) of these cases were of VSCCs. Nineteen cows were Holstein, three were Charolais, one was Jersey, and 10 were mix breed cows. Grossly, the main change was vulvar swelling, with bleeding and concomitant myiasis. The tumor masses were firm, ulcerated and with yellow areas. It was possible to reevaluate microscopically 30 out of the 33 cases. Eight of them were well differentiated, 17 were moderately differentiated, and five were poorly differentiated VSCCs. The evaluation of squamous intraepithelial lesions (SILs) was performed in 21 cases. Epithelial hyperplasia was observed in 10 cases, mild dysplasia in two, moderate in one, and severe in five cases; in three cases no SILs were observed. Fontana-Masson stain for melanin was performed in 21 cases. In 17 cases the epidermal pigmentation was absent; it was mild in two and moderate in other two cases. Independently of the degree of differentiation, most neoplastic keratinocytes were strongly positive for bovine pancytokeratine through the immunohistochemistry (IHC) technique. Bovine papillomavirus was not detected by IHC in this study.

Carcinomas de células escamosas vulvares (CCEVs) em bovinos foram estudados retrospectivamente quanto à prevalência, epidemiologia, quadro clinicopatológico e aspectos imuno-histoquímicos. O grau de pigmentação da pele vulvar foi também avaliado. Nos 48 anos analisados retrospectivamente, foram computados materiais de necropsias e biópsias de 7.483 bovinos recebidos no Laboratório de Patologia Veterinária da UFSM. Desses, em 664 (8,87%) casos de neoplasmas foram identificados, sendo 33 (4,97%) casos de CCEVs. Dezenove eram vacas da raça Holandesa, três da Charolesa, uma era Jersey e 10 eram sem raça definida. A principal alteração macroscópica foi aumento de volume vulvar, sangrante e com miíase concomitante. As massas tumorais eram firmes, ulceradas e com áreas amarelas. Foi possível reavaliar microscopicamente 30 dos 33 casos. Desses, oito eram CCEVs bem diferenciados, 17 eram moderadamente diferenciados e cinco eram pobremente diferenciados. A avaliação de lesões intraepiteliais escamosas (LIEs) foi realizada em tecidos de 21 casos que tinham epitélio de revestimento. Hiperplasia epitelial foi observada em 10 casos; displasia leve em dois, moderada em um e acentuada em cinco casos; em três casos não havia LIEs. A técnica de Fontana-Masson para melanina foi realizada em 21 casos. Desses, em 17 a pigmentação do epitélio da epiderme vulvar era ausente, em dois era leve e em outros dois era moderada. Independentemente do grau de diferenciação dos CCEVs, houve imunomarcação acentuada da maioria dos ceratinócitos neoplásicos para pancitoceratina bovina pela técnica de imuno-histoquímica (IHQ). Papilomavírus bovino não foi detectado pela IHQ neste estudo.


#22 - Poisoning by Portulaca elatior (Portulacaceae) in goats, 31(6):465-470

[Poisoning by Portulaca elatior (Portulacaceae) in goats.] Intoxicação por Portulaca elatior (Portulacaceae) em caprinos. Pesquisa Veterinária Brasileira 31(6):465-470. Hospital Veterinário, Centro de Saúde e Tecnologia Rural, Universidade Federal de Campina Grande, Campus de Patos, 58700-000 Patos, PB, Brazil. E-mail: franklin.riet@pq.cnpq.br An outbreak of poisoning by Portulaca elatior was diagnosed in the state of Paraíba, northeastern Brazil, during the rainy season. Between February and April, 16 out of 316 goats were affected and 13 died. Fifteen of the affected animals were from a flock of 16 goats introduced on the farm at the beginning of the rainy season. Clinical signs were intense salivation, reluctance to move, lip tremors, slight uncoordinated gait, diarrhea, bloat, abdominal pain, groans, dehydration and constant bellowing. The course of the disease ranged from 2 to 48 hours; 13 goats died and three recovered. Gross lesions were reddening of the mucosa of rumen and reticulum, hemorrhages in the ruminal serosa, ulcerations of the abomasal mucosa, and ecchymoses in the small intestinal mucosa. Histological examination revealed degeneration and necrosis of epithelial cells with vesicle formation in the fore stomachs associated with infiltration by neutrophils. Suspected to have caused the outbreak, Portulaca elatior was administered experimentally to three animals at doses of 40, 20 and 10 grams of the fresh plant per kg bodyweight, respectively. The goats that ingested 20 and 40g/kg died, and the one that ingested 10g/kg showed clinical signs but recovered. These animals showed similar clinical signs and histological lesions as those seen in the goats poisoned spontaneously, demonstrating that the disease was caused by the ingestion of Portulaca elatior. Oxalate concentration were determined in P. elatior and in another plant identified as Blutaparon vermiculare. P. elatior contained 6.37% of total oxalates and B. vermiculare 5.29%. B. vermiculare was administered experimentally to one goat at a dose of 40g/kg, without causing clinical signs. These results and the absence of oxalate crystals in the digestive system and kidneys of the goats necropsied suggest that poisoning by P. elatior is not caused by high oxalate concentration in the plant.

E-mail: franklin.riet@pq.cnpq.br Um surto de intoxicação espontânea por Portulaca elatior foi diagnosticado no município de Aroeiras, Paraíba, na época das chuvas. Entre os meses de fevereiro e abril, em um rebanho de 316 caprinos, 16 adoeceram, sendo 15 de um lote de 16 caprinos que tinham sido introduzidos recentemente na fazenda e um animal do rebanho já existente na fazenda. Os caprinos apresentaram intensa salivação, relutância em se movimentar, tremores de lábios, marcha incoordenada, diarreia, timpanismo, dor abdominal, gemidos, desidratação e berros constantes. A evolução da enfermidade variou de 2 a 48 horas; 13 caprinos morreram e três se recuperaram. As lesões macroscópicas caracterizavam-se por mucosa ruminal e do retículo avermelhadas, serosa do rúmen hemorrágica, mucosa abomasal avermelhada com áreas ulceradas e mucosa do intestino delgado com equimoses. No exame histopatológico observou-se degeneração e necrose das células epiteliais dos pré-estômagos com formação de vesículas e infiltrado inflamatório neutrofílico. Suspeita de ter causado o surto, Portulaca elatior foi administrada a três caprinos nas doses de 40, 20 e 10 gramas de planta fresca por kg de peso animal (g/kg). Esses caprinos apresentaram sinais semelhantes aos casos espontâneos. Os animais que ingeriram 20 e 40g/kg morreram e o que ingeriu 10g/kg se recuperou. Microscopicamente foram observadas lesões do sistema digestivo semelhantes às dos casos espontâneos. A reprodução experimental da intoxicação, com sinais clínicos e lesões histológicas semelhantes às dos casos espontâneos, comprova que a doença foi causada por Portulaca elatior. P. elatior apresentou níveis de oxalatos de 6,37% e outra planta, identificada como Blutaparon vermiculare e que ocorria em grande quantidade nas pastagens, apresentou níveis de oxalatos de 5,29%. B. vermiculare foi administrada experimentalmente a um caprino na dose de 40g/kg, sem que o animal apresentasse sinais clínicos. Esse resultado e a ausência de cristais de oxalatos no sistema digestivo e rins dos caprinos necropsiados, sugerem que a intoxicação por P. elatior não é causada pela alta concentração de oxalatos na planta.

E-mail: luacristiny@yahoo.com.br With the aim to study the distribution of lesions the rabies virus in spontaneous cases of rabies in ruminants and to determine the efficiency of the direct fluorescent antibody test (DFA), mouse inoculation (MI) and presence of Negri bodies in the diagnosis of the disease, 48 cases of the rabies were examined. Samples of frontal, temporal, parietal and occipital cerebral cortex, hippocampus, thalamus, rostral and caudal colliculi, cerebellum, pons, medulla oblongata, basal nuclei and sections of the cervical, thoracic and lumbar spinal cord were examined. Of the 48 samples examined all were positive on DFA and MI, and in 30 (62.5%) Negri bodies were observed. However there were differences in the results of the three tests among different regions of the central nervous system. In the samples of the cerebral cortex in 38 cattle, the frequency of inclusion bodies was low (11-37%), and so was the positivity to DFA and MI (60-80%). In contrast, all samples of thalamus, pons and spinal cord were positive to DFA and MI. In other regions of the brain stem, positivity to these tests varied between 60% and 96.7%. On histologic examination, the major frequency of Negri bodies (88.2%) was observed in the cerebellum. In eight sheep the DFA and MI tests were positive in all sections of the CNS examined and Negri bodies were found in three sheep. Only two goats were examined; both were positive in DFA and MI tests and in one Negri bodies were found. These results suggest that the recommendations of the Brazilian Technical Manual for Rabies of Herbivores is adequate for rabies diagnosis, because their recommendations include the histologic study and the examination of cerebellum, and sections of the brainstem with high positivity to DFA and MI tests. However, a better recommendation is to send for DFA and MI half of the brain cut longitudinally and samples of the spinal cord, which will permit to examine one or two sections, and if those are negative to get back to the material and examine the rest of the sections. In contrast, to collect samples of the brain or half brain can be inappropriate for the diagnosis of other diseases of the CNS, for which the study of the whole fixed brain is necessary to recognize the symmetry or distribution of lesions. In these situations by the results obtained here, it can be recommended to send different sections of the spinal cord for DIF and MI tests and to fix the whole brain for gross and histologic examinations.

E-mail: luacristiny@yahoo.com.br Com o objetivo de identificar a distribuição das lesões do vírus rábico no sistema nervoso central de casos espontâneos de raiva em ruminantes e comparar as técnicas de imunofluorescência direta (IFD), inoculação em camundongos (ICC) e presença de corpúsculos de Negri para o diagnóstico da doença foram analisados materiais proveniente de 48 casos de raiva, incluindo amostras de córtex frontal, temporal, parietal e occipital, hipocampo, tálamo, colículo rostral e caudal, cerebelo, ponte, medula oblonga, núcleo da base e porções da medula cervical, torácica e lombar. De 48 amostras analisadas, todas foram positivas na IFD e na ICC e em 30 (62,5%) foram encontrados corpúsculo de Negri (CN). No entanto, houve diferenças importantes no resultados dos três testes nas diferentes regiões do SNC avaliadas. Nos cortes de córtex cerebral, em 38 bovinos, a presença de corpúsculos de inclusão foi baixa (11%-37%) assim como a positividade para IFD e ICC (60-80%). Pelo contrário, todas as amostras de ponte, tálamo e medula testados foram positivas para IFD e ICC. Em outras regiões do tronco encefálico e também no cerebelo a positividade para ICC e IFD foi de 60% a 96,7%. No cerebelo foi encontrada a maior frequência (88,2%) de corpúsculos de inclusão. Em oito ovinos as provas de ICC e IFD foram positivas em todos os cortes e foram observados corpúsculos de inclusão em três animais. Foram analisados somente dois casos de caprinos encontrando-se corpúsculos de inclusão em um e ambos foram positivos para IFD e ICC. Os resultados obtidos nesse trabalho sugerem que a conduta recomendada pelo Manual Técnico de Controle da Raiva dos Herbívoros (MTCRH) permite o diagnóstico de raiva associando o estudo histológico aos testes de IFD e ICC que incluem cerebelo, tálamo e tronco encefálico que apresentam alta positividade para as provas de IFD e ICC. No entanto, a melhor conduta é a de incluir metade do encéfalo cortado longitudinalmente e amostras de medula. Isto permite examinar por IFD e ICC uma ou mais regiões onde essas provas apresentam maior positividade e, posteriormente, se essas provas fossem negativas, retornar ao material original e examinar outras regiões. Por outro lado, a coleta de amostras dos locais recomendados pela MTCRH, assim com a coleta de metade do encéfalo, podem prejudicar o diagnóstico de outras doenças para o qual é necessário o estudo de todo o encéfalo após a fixação em formaldeído, para constatar a simetria e a distribuição das lesões. Nestes casos, com base nos resultados obtidos neste trabalho, pode ser recomendado para diagnóstico laboratorial de raiva o envio exclusivo de porções da medula cervical, dorsal e lombar, já que as três porções apresentaram 100% de positividade nas provas de IFD e ICC. Além disso, o estudo histológico de todas as porções do cérebro incluídas neste trabalho permitirá complementar o diagnóstico.

[Congenital malformations in ruminants in the semiarid of the Brazilian Northeast.] Malformações congênitas em ruminantes no semiárido do Nordeste Brasileiro. E-mail: dantas.af@uol.com.br Congenital malformations caused by the ingestion of Mimosa tenuiflora have been reported in ruminants in the semiarid of the Brazilian Northeast. This paper reports malformations diagnosed in ruminants, from 2000 to 2008, by the Veterinary Pathology Laboratory of the Federal University of Campina Grande, Patos, PB, in municipalities of the states of Paraíba, Pernambuco and Rio Grande do Norte. During the period, 47 (3.48%) out of 1.347 ascensions were reported as malformations. Based in the type of malformation and in the origin of the animals, malformations were divided in: 1) caused by the ingestion of M. tenuiflora, and 2) sporadic malformations of unknown causes. In sheep, 21 out of 418 ascensions were malformations, being 18 (4.3%) of malformations caused by M. tenuiflora and 3 (0.71%) of sporadic malformations. In cattle, 14 out of 434 ascensions were malformations, from these 8 (1.84%) were caused by M. tenuiflora and 6 (1.38%) were sporadic malformations. In goats, 12 out of 495 ascensions were malformations, being 9 (1.81%) malformations related with the ingestion of M. tenuiflora and 3 (0.6%) sporadic malformations. More frequent malformations caused by M. tenuiflora were arthrogryposis, micrognatia, palatoschisis, microphtalmia and unilateral or bilateral hypoplasia or aplasia of the incisive bones. Sporadic malformations were acephaly and hermaphrodite, dicephaly and malformations of mesenteric vessel in sheep; atresia ani in three goats; and hydranencephaly, atresia ani, ribs malformation with eventracion, cerebellar hypoplasia with hydrocephalus, pulmonary choristoma and meningocele, and siamese twins in cattle. A case of cerebellar hypoplasia with hydrocephalus was negative on immunohistochemistry to bovine viral diarrhea virus. Malformations caused by M. tenuiflora occurred during the whole year. The highest frequency in sheep seems to be associated with the consumption of the plant by ewes after first rains, in the first two months of gestation, when they are supplemented with concentrates, and M. tenuiflora is the main green forage available. Malformations occur mainly in degraded areas of native forest (caatinga) invaded by M. tenuiflora, with lesser variety of other species.

E-mail: dantas.af@uol.com.br Malformações congênitas causadas pela ingestão de Mimosa tenuiflora têm sido observadas em ruminantes no semiárido do Nordeste Brasileiro. Neste trabalho foram estudadas as malformações congênitas em ruminantes diagnosticadas entre 2000 e 2008, em municípios da Paraíba, Pernambuco e Rio Grande do Norte. Durante o período foram recebidos 1.347 materiais de ruminantes para diagnóstico, desses 47 (3,48%) foram dignosticados como malformações congênitas. Com base no tipo de malformação e na procedência do animal as malformações foram divididas em: 1) causadas pelo consumo de M. tenuiflora; e 2) malformações esporádicas, sem causa conhecida. De 418 materiais de ovinos, 21 corresponderam a malformações, sendo 18 (4,3% do total de materiais) de malformações causadas por M. tenuiflora e 3 (0,71%) de malformações esporádicas. De 434 materiais de bovinos, 14 foram diagnosticados como malformações, sendo 8 (1,84%) causadas por M. tenuiflora e 6 (1,38%) malformações esporádicas. De 495 materiais de caprinos, 12 apresentaram malformações, sendo 9 (1,81%) causadas pela ingestão de M. tenuiflora e 3 (0,6%) malformações esporádicas. As principais malformações causadas por M. tenuiflora foram artrogripose, micrognatia, palatosquise, microftalmia e hipoplasia ou aplasia unilateral ou bilateral dos ossos incisivos. As malformações esporádicas incluiram: acefalia e hermafroditismo, dicefalia e malformações de vasos intestinais em ovinos; atresia anal em três caprinos; e hidranencefalia, atresia anal, malformações de costelas com eventração, hipoplasia cerebelar e hidrocefalia, coristoma pulmonar e meningocele, e gêmeos siameses em bovinos. O caso de hipoplasia cerebelar com hidrocefalia foi negativo pela imuno-histoquímica para o vírus da diarreia viral bovina. Malformações congênitas causadas por M. tenuiflora ocorreram durante todo o ano. A maior frequência em ovinos está aparentemente associada ao consumo da planta, na primeira fase da gestação, após as primeiras chuvas, quando as ovelhas estão sendo suplementadas e a planta é o principal volumoso disponível. As malformações ocorrem principalmente nas áreas mais degradadas, onde existe maior disponibilidade da planta e menor variedade de plantas da caatinga.


#25 - Focal symmetrical encephalomalacia in sheep, 30(5):423-427

E-mail: franklin.riet@pq.cnpq.br Focal symmetrical encephalomalacia (FSE) is the most prominent lesion seen in the chronic form of enterotoxemia by Clostridium perfringens type D. This paper reports FSE in sheep in Brazil. Six deaths occurred within a seven days period in a flock of 70, four to 30-month-old Santa Inês sheep in the state of Paraíba in the Brazilian semiarid. The flock was grazing a paddock of irrigated sprouting Cynodon dactylon (Tifton grass), and supplemented, ad libitum, with a concentrate of soybean, corn and wheat. Nervous signs included blindness and recumbence. A 19 month-old sheep was examined clinically and necropsied after a clinical course of three days. Gross lesions were herniation of the cerebellar vermis and multifocal, bilateral, symmetric brownish areas in the internal capsule, thalamus and cerebellar peduncles. Histologic lesions were multifocal, bilateral malacia with some neutrophils, swelling of blood vessels endothelium, perivascular edema, and hemorrhages. The flock was vaccinated, before the outbreak, with only one dose of Clostridium perfringens type D vaccine. Two factors are suggested to be important for the occurrence of the disease: insufficient immunity due to the incorrect vaccination; and high nutritional levels by the supplementation with highly fermentable carbohydrates.

E-mail: franklin.riet@pq.cnpq.br Encefalomalacia focal simétrica (EFS) é a lesão mais proeminente vista nas formas subaguda ou crônica da enterotoxemia por Clostridium perfringens tipo D. Este trabalho relata EFS em ovinos no semiárido do estado da Paraíba. Seis ovinos morreram, em um período de sete dias, dentro de um rebanho de 70 animais, da raça Santa Inês, entre 4-30 meses de idade, que pastavam em piquete de Cynodon dactylon (capim Tifton), que estava rebrotando. Os ovinos eram suplementados com um concentrado de soja, trigo e milho. Os sinais nervosos incluíam cegueira e decúbito lateral. Um ovino de 19 meses de idade foi examinado clinicamente e necropsiado, depois de um curso clínico de 3 dias. Macroscopicamente foram observadas herniação do cerebelo e áreas acastanhadas, multifocais, simétricas e bilaterais na cápsula interna, tálamo e pedúnculo cerebelar. Histologicamente observou-se malacia, bilateral e simétrica, com alguns neutrófilos, tumefação das células endoteliais dos vasos sanguíneos, edema perivascular e hemorragia. O rebanho foi vacinado, antes do surto, com uma única dose de vacina para Clostridium perfringens tipo D. Dois fatores são sugestivos quanto a importância da ocorrência da enfermidade: imunidade insuficiente devida à vacinação incorreta; e altos níveis nutricionais da suplementação com carboidratos altamente fermentáveis.

E-mail: franklin.riet@pq.cnpq.br Para determinar as doenças que ocorrem no sistema nervoso de bovinos no semiárido nordestino, foi realizado um estudo retrospectivo em 411 necropsias de bovinos realizadas no Hospital Veterinário da Universidade Federal de Campina Grande, Patos, Paraíba, entre janeiro de 2000 a dezembro de 2008. Dos 411 casos analisados 139 (33,81%) apresentaram alterações clínicas do sistema nervoso e as fichas foram revisadas para determinar os principais achados referentes à epidemiologia, aos sinais clínicos e às alterações macroscópicas e microscópicas. Em 28 (20,14%) casos o diagnóstico foi inconclusivo. As principais enfermidades foram raiva (48,7% dos casos com sinais nervosos), abscessos cerebrais (7,2%) incluindo três casos de abscesso da pituitária, febre catarral maligna (6,3%), botulismo (6,3%), alterações congênitas (4,5%), traumatismo (4,5%), tuberculose (2,7%), tétano (2,7%), infecção por herpesvírus bovino-5 (2,7%), encefalomielite não supurativa (2,7%), intoxicação por Prosopis juliflora (2,7%), status spongiosus congênito de causa desconhecida (1,8%) e polioencefalomalacia (1,8%). Outras doenças diagnosticadas numa única oportunidade (0,9%) foram criptococose, listeriose, encefalite tromboembólica, linfossarcoma, tripanossomíase e babesiose por Babesia bovis.

Four outbreaks were caused by Ipomoea asarifolia, 3 by Brachiaria spp., 2 by Crotalaria retusa, 2 by Tephrosia cinerea, 1 by Panicum dichotomiflorum, 1 by Mascagnia rigida, and 20 by Mimosa tenuiflora. In goats, 6.4% of the diseases were caused by toxic plants. Seven outbreaks were caused by Mimosa tenuiflora, 1 by Ipomoea asarifolia, 1 by Ipomoea carnea, 1 by Ipomoea riedelli, 3 by Prosopis juliflora, 1 by Arrabidaea corallina, 2 by Aspidosperma pyrifolium, and 2 by Turbina cordata. In horses, 14% of the diagnosed diseases were due to plants poisonings including 12 outbreaks caused by Crotalaria retusa and one by Turbina cordata. Annual losses in the state of Paraíba by deaths of livestock are estimated in 3,895 cattle, 8,374 sheep, 6,390 goats, and 366 horses, which represent about US$ 1,380,000. Epidemiologic, clinical and pathologic aspects of poisonings by Crotalaria retusa in cattle, Brachiaria spp. in sheep, Prosopis juliflora in cattle and goats, Nerium oleander in cattle, Opuntia ficus-indica in goats, and Turbina cordata in horses and goats are reported.

Em bovinos 7,4% dos diagnósticos realizados pelo LPV foram intoxicações por plantas. Foram diagnosticadas intoxicações por Centhraterum brachylepis (um surto), Brachiaria spp. (um surto), Crotalaria retusa (dois surtos), Ipomoea batatas (um surto), Marsdenia sp. (um surto), gramíneas contendo nitratos e nitritos (um surto por Echinochloa polystachya e dois surtos por Pennisetum purpureum), Palicourea aeneofusca (um surto), Prosopis juliflora (três surtos), Nerium oleander (um surto) e Mimosa tenuiflora (sete surtos). Na espécie ovina 13% dos diagnósticos foram intoxicações por plantas. Os surtos foram causados por Ipomoea asarifolia (quatro surtos), Brachiaria spp. (três surtos), Crotalaria retusa (dois surtos), Tephrosia cinerea (dois surtos), Panicum dichotomiflorum (um surto), Mascagnia rigida (um surto) e malformações associadas à ingestão de Mimosa tenuiflora (20 surtos). Nos caprinos, 6,4% dos diagnósticos corresponderam à intoxicação por plantas. Sete surtos foram causados por Mimosa tenuiflora, um por Ipomoea asarifolia, um por Ipomoea carnea, um por Ipomoea riedelli, três por Prosopis juliflora, um por Arrabidaea corallina, dois por Aspidosperma pyrifolium, dois por Turbina cordata e um por Opuntia ficus-indica. Na espécie equina 14% das doenças diagnosticadas foram devidas a intoxicações por plantas, sendo 12 surtos por Crotalaria retusa e um por Turbina cordata. As perdas na Paraíba por plantas tóxicas são estimadas em 3.895 bovinos, 8.374 ovinos, 6.390 caprinos e 366 equinos, que representam uma perda econômica anual, por morte de animais, de R$ 2.733.097,00. E-mail: franklin.riet@pq.cnpq.br This paper reports paratuberculosis in sheep and goats for the first time in Brazil. On Farm 1 in a flock of 33 goats and 13 sheep, one adult female goat presented progressive weight loss during about one year, and soft feces during one month before death. All the animals from the flock were submitted to the comparative tuberculin test. One sheep was positive to the avian tuberculin (2.2%) and in two the results were inconclusive. On Farm 2, in a flock of 200 sheep and 80 goats, one adult ewe presented progressive weight loss during about one year, and soft feces during 20 days before death. All sheep older than four months of age and 23 goats were submitted to tuberculin test with avian tuberculin; 47 (25.4%) sheep were positive, 115 (61.5%) had inconclusive results, and 25 (13.4%) were negative. No positive tuberculin test were observed in the goats, but in 11 (47.8%) the test was inconclusive and in 12 (52.2%) was negative. At necropsy both affected animals had enlarged mesenteric lymph nodes. In the affected sheep, the gut wall was thickened and corrugated, mainly in the ileum and ileocecal valve. Histologically, granulomas with mainly foamy macrophages were observed in the lamina propria and submucosa of the gut, in mesenteric lymph nodes, and in the goat also in the liver. One tuberculin positive sheep and one tuberculin negative goat from Farm 1 were euthanized and necropsied. A mild thickened of the intestinal wall was observed grossly in both animals. Inflammatory infiltrate with mainly lymphocytes was observed on the submucosa and lamina propria of the gut. In the four cases acid-alcohol positive rods, positive for Mycobacterium spp. on immunohistochemical examination were observed in macro-phages and lymphocytes. New studies are necessary to know the prevalence and economical impact of paratuberculosis in goats and sheep in Brazil, to determine control measures, and to evaluate the risk of human transmission.

Na Fazenda 1, de um rebanho de 33 caprinos e 13 ovinos, uma cabra adulta apresentou emagrecimento progressivo por aproximadamente um ano e fezes pastosas um mês antes da morte. Todos os animais do rebanho foram tuberculinizados com a prova comparativa. Um ovino (2,2%) teve resultado positivo à tuberculina aviar e em dois o teste foi inconclusivo. Na Fazenda 2, com 200 ovinos e 80 caprinos, foi afetada uma ovelha adulta que apresentou emagrecimento progressivo por aproximadamente um ano e fezes pastosas por aproximadamente 20 dias. Todos os ovinos com mais de 4 meses de idade e 23 caprinos foram tuberculinizados com tuberculina aviar; em 47 (25,4%) o resultado foi positivo, em 115 (61,5%) inconclusivo e em 25 (13,4%) negativo. Entre as cabras não houve nenhuma positiva à tuberculina aviar, mas em 11 (47,8%) o teste foi inconclusivo e em 12 (52,2%) foi negativo. Na necropsia dos dois animais com sinais clínicos os linfonodos mesentéricos estavam aumentados de tamanho e edemaciados. O ovino afetado da Fazenda 2, apresentou espessamento e enrugamento da mucosa do intestino, principalmente no íleo e válvula íleo-cecal. Microscopicamente o caprino e o ovino com sinais clínicos apresentaram lesões semelhantes, caracterizadas por granulomas com predominância de macrófagos espumosos, na lâmina própria e submucosa do intestino e linfonodos mesentéricos. O ovino positivo à tuberculina e um caprino negativo na Fazenda 1 foram eutanasiados e apresentaram discreto espessamento da mucosa do íleo. Na histologia foi observado infiltrado preferentemente linfocítico. Em todos os casos dentro de macrófagos e linfócitos havia bacilos ácool-ácidos resistentes, positivos na imuno-histoquímica para Mycobacterium spp. Sugere-se a necessidade de estudar a prevalência de paratuberculose em rebanhos de caprinos e ovinos no Brasil, determinar medidas de controle e avaliar a possibilidade de transmissão para humanos.

The outbreaks occurred at beginning of the raining season, when the plant sprouts ahead other forages, or after the end of the raining season, when M. rigida stayed green and other forages had dried. In the experimental reproduction of the poisoning doses of 10 and 20g of M. rigida per kg body weight from two different regions were lethal to 3 goats and 3 sheep. One goat that ingested 20g/kg and a sheep that ingested 10g/kg recovered. Two sheep and two goats that ingested 5g/kg had mild clinical signs and recovered. Clinical signs of experimental and spontaneous cases were ingurgitation of the jugular veins, reluctance to move, sternal recumbence, incoordination, respiratory distress, depression, instability, and muscular tremors. Death occurred after a clinical manifestation period of about 4 min to 27h40min. The main lesions were pulmonary edema and vacuolization and necrosis of epithelial cells in some renal tubules. To test if the active principle of M. rigida causes sudden deaths in newborn lambs and kids, 2g/kg of the plant were given daily to two goats and five sheep in the 15 days previous to parturition. One sheep aborted two lambs, 5 days before parturition. The four lambs of the other four sheep ingested the colostrum without problems. The kid from one goat ingested the colostrum and died suddenly 5 minutes after. The kid from the other goat died immediately after parturition before ingestion of colostrum. These results suggest that the active principle of M. rigida was eliminated through the milk at toxic doses for the kids. Management measures to prevent the poisoning are recommended.
