#1281 - The materno-fetal interface in llama (Lama guanicoe glama), p.221-228

Abstract in English:

ABSTRACT.- Iturrizaga D.M., Verechia F.T., Santos T.C., Bombonato P.P., Teixeira D.G. & Miglino M.A. 2007. The materno-fetal interface in llama (Lama guanicoe glama). Pesquisa Veterinária Brasileira 27(6):221-228. Departamento de Cirurgia, Faculdade de Medicina Veterinária e Zootecnia, Universidade de São Paulo, Av. Prof. Dr. Orlando Marques de Paiva 87, São Paulo, SP 05508-270, Brazil. E-mail: miglino@usp.br Samples from 9 llamas (28 through 36 weeks of gestation) were collected and fixed in 4% buffered paraformaldehyde (light microscopy) and in 2.5% buffered glutaraldehyde (transmission and scanning electron microscopy). The material was processed in paraplast and slides (5mm) were stained with HE, PAS, Masson-Trichrome, acid phosphatase and Perl´s. The uteroferrin was immunolocalized. The results show that llama placenta is chorioallantoic, diffuse, folded and epitheliochorial, and the fetus is covered with an epidermal membrane. The trophoblast cells have variable morphology: cubic, rounded and triangular cells, with cytoplasm containing PAS-positive granules. Binucleated cells with large cytoplasm and rounded nuclei, as well as giant trophoblastic cells with multiple nuclei were also observed. Numerous blood vessels were observed beneath the cells of the uterine epithelium and around the chorionic subdivided branches. Glandular activity was shown by PAS, Perl´s, and acid phosphatase positive reactions in the cytoplasm and glandular lumen, and by immunolocalization of the uteroferrin in the glandular epithelium. The uterine glands open in spaces formed by the areoles, which are filled by PAS-positive material. The llama fetus was covered by the epidermal membrane, composed of stratified epithelium, with up to seven layers of mono-, bi- or trinucleated cells. The high level of maternal and fetal vascularization surfaces indicates an intense exchange of substances across both surfaces. The metabolic activity shown in the uterine glands suggests an adaptation of the gestation to the high altitudes of the natural habitat of this species.

Abstract in English:

ABSTRACT.- Silva M.C., Fighera R.A., Brum J.S., Graça D.L., Kommers G.D., Irigoyen L.F. & Barros C.S.L. 2007. [Clinicopathological features in 620 neurological cases of canine distemper.] E-mail: claudioslbarros@uol.com.br The files of 5,361 necropsies performed in dogs in the Veterinary Pathology Laboratory of the Federal University of Santa Maria during 1965-2006 were reviewed in search of cases of canine distemper. Six hundred and eighty three cases (12.7%) of the disease were found, 620 of which had neurological signs. From those 620, the following data on each case were retrieved: age, clinical signs, histopathology and concomitance or not of another disease. Age groups were classified as puppies (up to 1 year of age), adults (from 1 to 9 years) and aged (from 10 years on). In 565 out of the 620 (91.1%) neurological cases of canine distemper, histopathological brain changes were observed and in 554 of those 565 the age was registered in the files with following age group distribution: 45.9% of puppies, 51.4% of adults, and 2.7% of aged dogs. Neurological clinical signs encompassed a large spectrum of motor, postural and behavioral disturbances which could occur together or individually. Most frequent clinical signs were myoclonus (38.4%), motor incoordination (25.0%), seizures (18.5%), and paraplegia (13.4%). In 98.4% of the 565 dogs with histopathological changes in the brain demyelination, non-suppurative encephalitis or a combination of these two were found. Intranuclear eosinophilic inclusion bodies were observed in different brain cells of 343 of the 565 dogs with histopathological changes. In 170 (49.6%) the cellular type bearing the inclusions was not mentioned in the file and in the remaining cases the inclusions were seen in astrocytes (94.8% of the cases), neurons (3.5%), oligodendrocytes (1.1%), and ependyma cells (0.6%). Taking in consideration the type of lesions and the age groups, cases with combined demyelination and non-suppurative encephalitis occurred in 40.0% of the puppies, 51.2% of the adult dogs and 72.7% of the aged dogs. Demyelination alone occurred in 48.4% of the puppies, 41.3% of the adults and in 35.7% of the aged dogs. Non-suppurative encephalitis alone occurred 11.6% of the puppies, 7.5% of the adults and in 7.1% of the aged dogs.

ABSTRACT.- Rodrigues A.B., Athayde A.C.R., Rodrigues O.G., Silva W.W. & Faria E.B. 2007. [Evaluation of the efficacy of antihelmintics to control gastrointestinal nematodes in goats raised in the state of Paraíba.] E-mail: adrianabonfimr@yahoo.com.br The objective was to evaluate conventional and alternative anthelminthic action on the sensibility of gastrointestinal nematodes in goats. Hundred and twenty goats of both sexes were distributed into 10 groups of 24 animals each (12 males and 12 females) and submitted to treatments of specific anthelminthic composition. The commercial drugs used were moxidectin 0.2%, albendazole, levamisol chlorhydrate, ivermectin and an aqueous extract of the purgative potato (Operculina hamiltonii). The reduction test was applied through egg counts per gram of feces and larval culture to evaluate resistance. The fecal samples were collected in the day when medication was accomplished (day base), at 7, 14 and 21 days after treatment. The following results for the reduction of Trichostrongyloidea eggs were obtained: Treatment of the females with Moxidectina reduced the egg counts to 92.8, 88.7 and 89.8%, of the males to 92.6, 96.2 and 98.1%; with Levamisol of the females to 96, 97.1 and 91%, of the males to 85.7, 94,2 and 100%; with Albendazol of the females 65, 60.3 and 75.4%, of the males 88.8, 88.8 and 55.5%; with Ivermectin of the females to 92.2, 68.6 and 70.6%, of the males to 41.7, 73.6 and 59.7%; and with the purgative potato of the females to 31.8, 34.1 and 49.4%, of the males to 61.5, 80.7 and 50%. In larval culture Haemonchus, followed by Bunostomum, Trichostrongylus, and Oesophagostomum, were identified even after treatments.

Abstract in English:

ABSTRACT.- Pescador C.A., Oliveira E.C., Pedroso P.M.O., Bandarra P.M., Okuda L.H., Corbellini L.G. & Driemeier D. 2007. [Reproductive losses linked to Toxoplasma gondii infection in goats in southern Brazil.] E-mail: davetpat@ufrgs.br Toxoplasma gondii was implicated with reproductive losses in a goat herd in Rio Grande do Sul, Brazil. Gross changes were present in 2 out of 6 fetuses/offsprings submitted for diagnosis and included enlarged, pale mesenteric lymph nodes, and edematous, mottled red and tan lungs. Microscopic changes were observed in most fetuses and were especially characterized by lymphoplasmacytic infiltration in the brain and lungs. Other histological changes included lymphoplasmacytic interstitial nephritis, necrotizing lymphadenitis and periportal lympho-plasmacytic cell infiltrates. While bacteriological and direct immunofluorescent antibody tests for Leptospira sp. were negative in samples from all cases, immunohistochemical and PCR procedures for Toxoplasma gondii reacted positively in most of them. Antibody titles against T. gondii varying from 1:512 to 1:2048 were detected in serum samples from the mother goats of these aborted (1), stillborn (3) or dead newborn (2) kids. This paper describes the clinical, pathological, serological, molecular and immunohistochemical findings of a Toxoplasma gondii-infected goat flock.

E-mail: ceambrosio@usp.br Fifteen papillae from female water buffaloes and one bovine female were obtained in a slaughter house for this study, which consisted in morphologic and biometrical analyses, histology and histometric and mesoscopical analyses. The means were: right cranial papilla (RCP) 20.95g; right caudal papilla (RCaP) 21.49g; left cranial papilla (LCP) 23.52g; and left caudal papilla (LCaP) 21.35g. Papillary duct average length, numbers of folds and papillary cisterns length were, respectively: 0.67/5,5 e 2.17cm for RCP; 0.62/6.0 e 2.26cm for RCaP; and for the left antimere: RCP: 0.71/6.0 e 2.26cm and for RCaP: 0.74/5.7 e 2.57cm. Pearson’s analysis revealed negative correlation between body weight and mammary glandular weight. Sampling of proximal, middle and distal area was taken and fixed into a 10% formaldehyde solution, followed by mesoscopical investigation through 10, 20 and 40µm thickness cuts stained by Azan, F3BA Picrosirus and Weigert-Van Gieson techniques. Histological and morphometric analyses were applied on 5µm thick samples and showed that the muscular layer was disposed into two arrangements, internal circular and external longitudinal with a bi-stratified cuboidal epithelium. A well developed muscular layer were related to the middle portion revealing thickness of 22.90mm with a great amount of connective tissue (7.10mm).

E-mail: ceambrosio@usp.br In 60 hemiheads of sheep of the Santa Ines breed the apparent origin in the skull of itinerary, ramification and distribution of the glossopharingeal nerve (Gf) and the vagus nerve (Vg) in the retropharyngeal region (Rr) were studied. By fixation and dissection of the specimens it was seen that the glossopharyngeal nerve and the vagus nerve arise from the jugular foramen in 100% of the cases. The right and the left glossopharingeal nerve (Glde) are frequently (86.6%) located more medially to the tympanic bulla, pass caudally to the stylohyoid bone, bypass the margin of the caudal stylopharyngeal muscle, the tonsilla, of the pharyngeal and the lingual mucous membrane. These branches are distributed, respectively, in the carotid sinus, pharyngeal musculature, soft palate, stylopharyngeal muscle, palatine tonsil, pharyngeal mucosa and the caudal third of the tongue (100%). The right and the left vagus nerve run caudally in association with the accessory nerves (Ac) up to the atlas (70%) and go then medially to the occipital artery, dorsally to the common carotid and the sympathetic trunk in the cervical region (80%). The vague nerves have pharyngeal (86.6%) and cranial laryngeal (100%) branches.

#1287 - Histopathological and histomicrobiological study of root canal therapy medication, comparison of calcium hydroxide versus gutta-percha with zinc oxide/eugenol in the teeth of dogs, p.71-74

Abstract in English:

ABSTRACT.- Domingues-F. L.M., Gioso M.A. & Padilha Filho J.G. 2007. Histopathological and histomicrobiological study of root canal therapy medication, comparison of calcium hydroxide versus gutta-percha with zinc oxide/eugenol in the teeth of dogs. Pesquisa Veterinária Brasileira 27(2):71-74. Departamento de Cirurgia da Faculdade de Medicina Veterinária e Zootecnia, Universidade de São Paulo, Av. Prof. Dr. Orlando de Marques de Paiva 87, Bloco 8-superior, Cidade Universitária, São Paulo, SP 05508-000, Brazil. E-mail: lesliedf@usp.br The presence of microorganisms in dental structures with experimentally induced necrosis was evaluated. The materials were tested to evaluate their antimicrobial activity and tissue repair efficacy. Four dogs were used in this experiment, with a total of 64 roots of premolar teeth, divided into three groups. The root canals of Group I were filled with gutta-percha and zinc oxide/eugenol cement; Group II were filled with calcium hydroxide, and Group III were not filled. All animals were clinically and radiographically examined 15 days after surgery and then again every subsequent 15 days until 120 days, when the teeth were extracted en bloc. Histopathological analysis showed inflammatory infiltration, cement and bone resorption and necrotic tissue in the apical delta in different proportions. Histomicrobiological analysis showed the presence of microorganisms inside the teeth structures, with different concentrations according to the treatment used. There was statistical significance between the groups (p>0.05). Gutta-percha with zinc oxide/eugenol demonstrated good antimicrobial activity; calcium hydroxide was not efficient. The conclusion of this study is that gutta-percha with zinc oxide/eugenol is the better protocol for filling root canals in dogs.

ABSTRACT.- Brito L.G., Regitano L.C.A., Huacca M.E.F., Carrilho E. & Moya Borja G.E. 2007. Genotype characterization of the horn fly Haematobia irritans from different Brazilian geographic regions based on randomly amplified polymorphic DNA (RAPD) analysis. Pesquisa Veterinária Brasileira 27(1):1-5. Laboratório de Sanidade Animal, Embrapa Rondônia, BR 364 Km 5,5, Porto Velho, RO 78900-970, Brazil. E-mail: luciana@cpafro.embrapa.br Blood-sucking diptera are important parasites in bovine production systems, especially regarding confinement conditions. Haematobia irritans, the horn fly, is one of the most troublesome species within bovine production systems, due to the intense stress imposed to the animals. An important aspect while studying the variability within a species is the study of the geographic structure of its populations and, attempting to find out the genetic flow of Brazilian populations of horn fly, the RAPD technique, which is suited for this purpose, has been used. The use of molecular markers generated from RAPD made it possible to identify the geographic origin of samples from different Brazilian geographic regions, as well as to estimate the genotypic flow among the different Brazilian populations of the horn fly.

Abstract in English:

ABSTRACT.- Mathias L.A., Meirelles R.B. & Buchala F.G. 2007. [Stability of Brucella abortus whole cell antigen for use in the serological diagnosis of bovine brucellosis by the complement fixation test.] E-mail: lmathias@fcav.unesp.br The complement fixation test is used worldwide in the confirmatory diagnosis of bovine brucellosis. For this technique the antigen is the same as the one used in the tube agglutination test. However, literature is poor in information about the stability of the whole cell Brucella antigen for use in the complement fixation test to establish a time of validity of the antigen. Hence the aim of this investigation was to evaluate the stability of this antigen under refrigeration for use in the complement fixation test. Fourteen batches of antigen prepared with Brucella abortus strain 1119/3, produced from 9 months to 23 years and 11 months before, were analysed. One hundred and sixty-seven cattle sera with varying titres of antibodies to Brucella were tested through the warm complement fixation microtechnique with five 50% haemolytic units of complement. Sera with at least 25% of complement fixation in dilution 1:4 were considered positive. The results with 13 of the antigen batches were compared with the results obtained with the batch produced 9 months before by the McNemar c2 test and kappa statistic. The oldest antigen batch gave a higher proportion of sera titres which were exactly the same observed with the 9-month-batch (90.4%), and the antigen produced 4 years and 3 months before the test gave de lowest proportion of sera with the same titre of the 9-month-antigen (73.7%). The comparison of the results after being classified as positive and negative showed that the highest proportion of agreed results was observed with the antigen produced 21 years and 4 months before (98.8%, kappa 0.98). The antigen with the lowest proportion of agreed results was the one produced 3 years and 2 months before (91.6%, kappa 0.84). The results of the study show that most sera gave very similar results with all antigen batches evaluated, and that there was no relationship between the period of antigen production and the difference in test results.

Centro de Saúde e Tecnologia Rural, Universidade Federal de Campina Grande, 58700-000 Patos, PB, Brazil. E-mail: franklin.riet@pesquisador.cnpq.br The knowledge of the diseases of domestic animals in the different Brazilian regions is important to determine measures for their control and prevention. The objective of this paper is to report the epidemiology, clinical signs and pathology of the diseases of the central nervous system (CNS) of goats and sheep in the Brazilian semiarid, mainly in the state of Paraíba, diagnosed at the Veterinary Hospital of the Federal University of Campina Grande, from January 2000 to May 2006. During the period, 365 cases or outbreaks were diagnosed in goats and 270 in sheep. From these, 63 (9.92%) were of diseases of the CNS, being 34 (9.31%) in goats and 29 (10.7%) in sheep. The main diseases were abscesses (19.04%), tetanus (15.9%), rabies (9.52%) poisoning by Ipomoea asarifolia (7.93%), listeriosis (6.34%), traumatism (6.34%), polioencephalo-malacia (4.77%), pregnancy toxemia (3.17%), enzootic ataxia (3.17%), and meningitis (3.17%). Other diseases diagnosed in only one opportunity (1.59%) were intoxications by Crotalaria retusa, Ipomoea carnea, Ipomoea sericophylla and Prosopis juliflora, otitis with encephalitis, malformation, lymphoblastic lymphosarcoma, medulloblastoma, and focal symmetric necrosis. In 6.34% of the cases diagnosis was unknown.
