E-mail: peixotop@ufrrj.br São descritos aspectos toxicológicos, clínico-patológicos e ultraestruturais de coelhos intoxicados iatrogênica e experimentalmente por vitamina D por via subcutânea. Clinicamente, os animais evidenciaram sinais de insuficiência cardiovascular como ascite e edema pulmonar, hiporexia, anorexia, diarréia mucosa, emagrecimento e apatia. As clássicas alterações de mineralização e, por vezes, osseificação, do sistema cardiovascular, bem como as alterações de rins, pulmões, estômago, entre outros órgãos, foram reproduzidas com administrações subcutâneas de solução oleosa de colecalciferol (vitamina D3 não-ativada).

E-mail: peixotop@ufrrj.br Toxicological, clinic-pathological and ultrastructural aspects of iatrogenic and experimental subcutaneous poisoning in rabbits by vitamin D are described. Clinically the animals showed signs of cardiovascular insufficiency, as ascite and lung edema, hyporexia, anorexia, mucous diarrhoea, loss of weight and apathy. The classical alterations of minera-lization and, occasionally, ossification of the cardiovascular system, as well the lesions of kidneys, lungs, stomach, among other organs, were reproduced by the subcutaneous administration of an oily solution of cholecalciferol (non-activated vitamin D3).

E-mail: olney@uenf.br Pythiosis is caused by Pythium insidiosum and the occurrence of disease in horses was described in the North and Northwest State of Rio de Janeiro, Brazil. The disease was described in cattle, sheep, humans, and horses in different states and regions across the country. This paper describes the development of IgY and IgG polyclonal antibodies, in chicken and rabbits, respectively against proteins extracted from kunkers and hyphae of P. insidiosum from affected horses. The proteins were recognized by chicken, rabbit and horse antibodies by immunodiffusion and Western blot against majority bands of 27 and 43 KDa, and titrated by ELISA. The antibodies IgY developed by the first time against Brazilian strains of P. insidiosum may represent a valuable tool in the detection of antigens of the pathogen and contribute to further studies aimed at immunotherapy and knowledge about this disease in endemic areas in Rio de Janeiro and in Brazil.

Este trabalho descreve o desenvolvimento de anticorpos policlonais, IgY e IgG, em galinha e coelho, respectivamente, contra proteínas extraídas de kunkers e hifas de P. insidiosum de cavalos doentes. As proteínas foram reconhecidas por anticorpos de galinha, coelho e cavalos contra as bandas majoritárias de 27 e 34 KDa em imunodifusão e Western blot tituladas por ELISA. Os anticorpos IgY desenvolvidos pela primeira vez contra cepas brasileiras de P. insidiosum podem representar um valioso instrumento na detecção de antígenos de patógenos e contribuem para novos estudos baseados na imunoterapia e no entendimento sobre esta doença em áreas endêmicas no Rio de Janeiro e em todo o país.

E-mail: anafranco.ufrgs@gmail.com Bovine herpesvirus type 5 (BoHV-5) is a major cause of viral meningoencephalitis in cattle. The expression of different viral proteins has been associated with BoHV-5 neuropathogenesis. Among these, gI, gE and US9 have been considered essential for the production of neurological disease in infected animals. To evaluate the role of gI, gE and US9 in neurovirulence, a recombinant from which the respective genes were deleted (BoHV-5 gI-/gE-/US9-) was constructed and inoculated in rabbits of two age groups (four and eight weeks-old). When the recombinant virus was inoculated through the paranasal sinuses of four weeks-old rabbits, neurological disease was observed and death was the outcome in 4 out of 13 (30.7 %) animals, whereas clinical signs and death were observed in 11/13 (84.6%) of rabbits infected with the parental virus. In eight weeks-old rabbits, the BoHV-5 gI-/gE-/US9- did not induce clinically apparent disease and could not be reactivated after dexamethasone administration, whereas wild type BoHV-5 caused disease in 55.5% of the animals and was reactivated. These findings reveal that the simultaneous deletion of gI, gE and US9 genes did reduce but did not completely abolish the neurovirulence of BoHV-5 in rabbits, indicating that other viral genes may also play a role in the induction of neurological disease.

Este vírus foi inoculado em coelhos de idades diferentes (quatro e oito semanas de idade). Quando o vírus recombinante foi inoculado nos seios paranasais de coelhos de quatro semanas de idade, doença neurológica e morte foram observadas em 4 dos 13 (30,7 %) animais, enquanto que sinais clínicos e morte foram observados em 11/13 (84,6%) dos coelhos infectados com o vírus parental. Em coelhos de oito semanas de idade, o BoHV-5 gI-/gE-/US9- não induziu sinais clínicos aparentes e, após tentativa de reativação viral por tratamento com dexametasona, o vírus não foi re-excretado. Por outro lado, o vírus selvagem causou doença clínica em 55,5 % dos coelhos e foi re-excretado após tratamento com dexametasona. Estes achados revelam que a deleção simultânea dos genes gI, gE e US9 reduziu mas não aboliu completamente a neurovirulência do BoHV-5 em coelhos, indicando que outros genes virais possam ter papel na indução da doença neurológica.

E-mail: leydsonf@fmva.unesp.br O presente trabalho visou avaliar a vitalidade e os níveis séricos de cortisol de bezerros neonatos, nascidos de partos eutócicos e auxiliados, bem como determinar os índices de partos distócicos em vacas receptoras de embriões nelores. Cerca de 310 bezerros foram avaliados. Observou-se a ocorrência de 38 partos distócicos (38/310) e de oito abortos (8/310). Os bezerros nascidos de partos distócicos mais duradouros, entre quatro e seis horas, apresentaram os maiores valores médios de frequência respiratória e dos níveis séricos de cortisol. A temperatura retal foi mais elevada em animais nascidos de partos laboriosos do que naqueles nascidos de partos normais. As taxas de morbidade e mortalidade em animais neonatos nelores foram de 77,93% e 3,44%, respectivamente. O emprego do escore APGAR, ao nascimento, demonstrou que 90% dos bezerros apresentavam-se deprimidos, independente do tipo de nascimento.

E-mail: leydsonf@fmva.unesp.br The purpose of present study was to evaluate the vitality and the serum levels of cortisol from newborn calves by normal and laborious deliveries, and to obtain the distocia register in Nelore embryo recipient cows. Three hundred ten calves were evaluated. The register of 38 dystocic calvings (38/310) and 8 abortions (8/310) were observed. In newborn animals the morbity and mortality rates were 77.93% and 3.44%, respectively. Calves born after long dystocic calving, lasting 4-6 hours, showed the highest mean values of respiratory frequency and serum cortisol levels. Animals born after laborious calving presented higher rectal temperature than those born at normal calving. The APGAR score showed that 90% calves had signs of dullness independent of parturition type.

De um modo geral, a indução de diabetess modifica o diâmetro vascular nas fêmeas e que a administração de vitamina C interfere nos dados métricos relativos ao diâmetro vascular somente nas fêmeas.

In a general way, the induction of diabetes modifies the vascular diameter in the females and that the vitamin C administration only intervenes with relative metric data to the vascular diameter in the females.


#116 - Intoxicações natural e experimental por salinomicina em coelhos, 29(9):695-699

Projeto Sanidade Animal Embrapa/UFRRJ, Seropédica, RJ 23890-000, Brazil. E-mail: pfpeixoto@terra.com.br An outbreak of salinomycin poisoning in rabbits is described. At least 27 out of 2,000 rabbits reared on a farm died after the coccidiostatic drug sulfaquinoxaline was substituted by salinomycin in the feed. An average of 26.9ppm salinomycin was detected in the ration given to the rabbits. Clinical signs included anorexia, apathy and bradykinesia, which progressed to incoordination and recumbency. Gross lesions consisted of pale areas in the skeletal muscles. The histopathological findings showed severe necrotic degenerative myopathy in association with infiltration of neutrophils and macrophages. One rabbit exhibited similar alterations in the myocardium. Mineralization was observed in the affected skeletal muscles in some cases. In order to verify if the poisoning was due to salinomycin, 20 rabbits were divided into five groups and a ration containing the drug at doses of 10, 25, 50, 75 and 100ppm was given. The administration of doses higher than 50ppm resulted in manifestation of the clinical signs seen in the outbreak of poisoning. It was concluded, that probably an error related to the mixture of salinomycin in the feed was the cause of deaths in the spontaneous outbreak of poisoning on the rabbit farm.

E-mail: pfpeixoto@terra.com.br Relata-se, pela primeira vez, um surto de intoxicação por salinomicina em coelhos. De 2000 animais, no mínimo 27 morreram após troca do coccidiostático sulfaquinoxalina pela salinomicina. A análise de parte da ração detectou 26,9ppm de salinomicina. Os sinais clínicos observados foram anorexia, apatia e lentidão com evolução para incoordenação dos movimentos e decúbito. As lesões macroscópicas consistiram de áreas pálidas na musculatura esquelética. O exame histopatológico evidenciou miopatia degenerativo-necrótica. Adicionalmente, verificou-se reação inflamatória constituída por neutrófilos e macrófagos. Um coelho apresentou lesões similares no miocárdio. Em alguns casos, mineralização estava presente nos músculos esqueléticos afetados. Vinte coelhos experimentais foram divididos em 5 grupos que receberam 10, 25, 50, 75 e 100ppm de salinomicina por via oral, com a finalidade de reproduzir a intoxicação. Os animais que receberam a partir de 50ppm de salinomicina apresentaram sinais clínicos semelhantes aos observados no surto espontâneo. Nossos resultados indicam que, provavelmente, erro na mistura da substância à ração causou a morte dos coelhos.

E-mail: lucianelaskoski@hotmail.com The gastrointestinal diseases of horses are aggravated by complications such as laminitis. The laminitis etiopathogeny are connected with lamellar basement membrane degradation by matrix metalloproteinases (MMPs). Inflammatory cytokines and leukocytes enzymes can active MMPs. The object of this study was to evaluate morphological changes on lamellar tissue of horses with colic syndrome and its association with clinical and laboratorial parameters. It was observed intensive destruction of lamellar architecture, mainly on animals with severe physical and laboratorial alterations, such as delayed capillary refill time, congested mucous membrane, tachycardia, hemoconcentration and low count of platelet and leukocytes. The results sign to the most likely moment of development of lamellar tissue injuries in horses with colic syndrome, which can be adopted preventive measures against laminitis.

E-mail: lucianelaskoski@hotmail.com As afecções gastrintestinais dos cavalos são agravadas por complicações como a laminite, cuja etiopatogenia está relacionada à degradação da membrana basal do tecido laminar por metaloproteinases (MMPs). A ativação das MMPs pode ocorrer devido à liberação local de citocinas inflamatórias ou enzimas provenientes de leucócitos infiltrados no tecido laminar. O objetivo deste trabalho foi avaliar as alterações morfológicas do tecido laminar de equinos com síndrome cólica letal e sua provável associação com parâmetros clínicos e laboratoriais. Observou-se intensa destruição da arquitetura laminar, principalmente nos animais com alterações físicas e laboratoriais mais acentuadas, como tempo de preenchimento capilar prolongado (TPC), membranas mucosas congestas, taquicardia, hemoconcentração e redução nas contagens de plaquetas e leucócitos. Os resultados sinalizam o provável momento do desenvolvimento de lesões do tecido laminar em equinos com síndrome cólica, no qual é possível adotar medidas preventivas contra a laminite.

E-mail: saulo-andrade@bol.com.br The aim of this study was to determine the clinical-pathological alterations and laboratory findings in cattle inoculated with Bothrops alternatus venom, with the intention of providing information for the establishment of diagnosis and differential diagnosis procedures, as well as to elucidate some obscurities observed in the pertinent literature. The lyophilized venom was diluted in 1 ml of physiologic solution. It was administered to 5 bovines by the subcutaneous route at doses of 0.0625, 0.125 and 0.25mg/kg body weight, and to 2 bovines by the intramuscular route at doses of 0.25 e 0.45mg/kg. Six bovines died and the only animal that survived, who had subcutaneously received the venom at a dose of 0.0625mg/kg, recovered. The first clinical signs were observed from 25min to 5h30min after the inoculation. The clinical evolution time varied from 7 hours 18 minutes to 92 hours. Regardless of the dose, the clinical picture was characterized by swelling (hemorrhage/hematoma) at the site of inoculation, increase in bleeding time and capillary refill time, paleness of mucous membranes and apathy. The laboratory exams revealed progressive normocytic normochromic anemia, thrombocytopenia, reduction in fibrinogen and total plasma proteins, decreased hematocrit and hemoglobin, and slight increase in creatine kinase and lactate dehydrogenase. When subjected to necropsy, the animals that received the venom through the subcutaneous route showed large hematomas and hemorrhagic areas in the subcutaneous tissue, extending from the site of inoculation. The animals in which inoculation was performed intramuscularly exhibited, in addition, intramuscular hemorrhage. The left endocardium showed extensive hemorrhagic lesions, and petechiae were found on the serosae of rumen, omasum, abomasum and gall bladder. The colon, rectum and perirenal areas were surrounded by clotted blood in three animals. Besides hemorrhage, the histological examination revealed hemorrhage and coagulative muscle necrosis in the vicinities of the inoculation site in the animals that received the venom by the intramuscular route. These lesions were mild in the muscles close to the site of inoculation in the animals inoculated by the subcutaneous route. The swelling on the site of inoculation and its surroundings was a consequence of blood accumulation and not merely edema. Myoglobinuria and gross or microscopic lesions were not observed in the kidneys. This study indicates that B. alternatus can lead adult bovines to death if they release all their venom during a bite. Conversely, since venomous ophidians can control the amount of poison they inoculate and they probably do not identify bovines as potential preys, it is likely that the number of accidents involving this species is small, which is in agreement with the findings of the majority of the centers for anatomical-pathological diagnosis in Brazil.

#119 - Susceptibility of laboratory animals to the intoxication by Ateleia glazioviana (Leg. Papilionoideae), 22(2):73-78

E-mail: lgmv@alternet.com.br Rabbits, guinea pigs, rats and mice were used to reproduce the cardiac lesions observed in ruminants in the intoxication by Ateleia glazioviana. Four animals of each species were fed during four months with a ration containing 10% of dry A glazioviana. The pellets were made by mixing 700 g of commercial ration, 200 g of cornstarch, 1000 mi of distilled water and 100g of the dry plant. Later this mixture was dried at 100ºC during 16-20 hours. Four control animals of each species were given similar food, but containing dry ryegrass (Lolium multiflorum) instead of A glaziovidna. Water was offered ad libitum and a daily amount of green oats (Avena sativa) was given to rabbits and guinea pigs. No clinical signs of disease were observed during the experiment. Ali animals were weighted weekly in the last 5 weeks of the experiment and no differences were observed in weight gains between treated and control groups. At the end of dosing ali animals were euthanatized and necropsied. No macroscopic or histologic lesions were observed in the heart or other organs. The abortive properties of A glazioviana were studied in rats. One group of 11 pregnant rats was fed, from day 7 to day 21 of gestation, ration containing 10% of A. glazioviana, prepared in the sarne way than in the previous experiment. A control group received the ration with 10% ryegrass. Ali rats delivered normal litters. To determine a possible loss of toxicity of the plant during the pellet preparation, dry A. glazioviana was heated for 16-20 hours at 100ºC and administered orally to one ewe. This sheep ingested daily 2.65 g/kg bw of dry A. glazioviana for 26 days. During the experimental period the animal had tachycardia and cardiac arrhythmia. At the end of the experiment bradycardia and severe depression were observed. It was euthanatized 7 days after the last plant administration. The sheep was 2.5-3 months pregnant and the fetus was apparently normal with no autolysis. Ascites and hydrothorax were observed at necropsy. Whitish areas were observed in the myocardium and the liver was pale. Degeneration and necrosis of cardiac fibers and proliferation of fibrous tissue were observed histologically in the heart. The liver had centrilobular degeneration of hepatocytes and congestion. These experiments indicate that A. glazioviana is not cardiotoxic for laboratory animals and suggest that the plant has a heat-stable active principie similar to the active principie of plants causing cardiac fibrosis (gousiekte) in Southern Africa.

E-mail: lgmv@alternet.com.br Coelhos, cobaias, ratos e camundongos foram utilizados com a finalidade de reproduzir a forma cardíaca da intoxicação por Ateleia glazioviana. Quatro animais de cada espécie receberam por 4 meses a planta, secada à sombra, na concentração de 10% na ração, fornecida na forma de pelets. Os pelets foram preparados misturando 700 g de ração comercial, 200 g de amido de milho, 1000 mi de água destilada e 100g de planta seca e, posteriormente, secados em estufa a 100°C, durante 16 a 20 horas. Quatro animais de cada espécie serviram como testemunhas, recebendo a ração preparada da mesma forma, porém com a utilização de azevém (Lolium multijlorum) em lugar de A glazioviana. Era fornecida água à vontade e, para os coelhos e cobaias, também aveia (Avena sativa) verde diariamente. Esses animais não apresentaram nenhuma alteração clínica, e foram pesados semanalmente nas últimas 5 semanas do período experimental, não apresentando diferença significativa no ganho de peso. À necropsia não foram observadas alterações macroscópicas e pelo exame histológico também não foram detectadas lesões significativas no coração e em outros órgãos. Para testar a atividade abortiva de A glazioviana, ração contendo 10% de planta seca, preparada da mesma forma que no experimento anterior, foi administrada a 11 outros ratos, fêmeas, prenhes, nos dois últimos terços da gestação. Um grupo controle de 11 fêmeas recebeu a ração com azevém a 10%, durante o mesmo período. Esses animais tiveram filhotes normais e no prazo correto. Para determinar a possível perda de toxidez da planta durante a preparação dos pelets, A glazioviana foi aquecida por 16 a 20 horas a 100ºC e, posteriormente, administrada a um ovino em 26 doses diárias de 2,65 g/kg de planta seca. Durante o período experimental, a ovelha apresentou batimentos cardíacos mais fortes, taquicardia e arritmia cardíacas, depois bradicardia e, no período final, apatia acentuada. Foi sacrificada 7 dias após a última administração da planta. Esse animal apresentava gestação de 2,5 a 3 meses e o feto não apresentava sinais de autólise. As alterações macroscópicas mais evidentes à necropsia foram ascite, hidrotórax, áreas esbranquiçadas no músculo cardíaco e fígado de coloração clara. Na histologia do coração foram observadas degeneração e necrose de fibras cardíacas e proliferação de tecido conjuntivo fibroso. O fígado apresentava congestão e degeneração de hepatócitos nas áreas centrolobulares. Os resultados indicam que esses animais de laboratório não foram susceptíveis à ação cardiotóxica de A. glazioviana, por via oral, e sugerem a possibilidade de que o princípio ativo da planta, que é resistente ao calor, seja semelhante ao princípio ativo das plantas do sul da África que causam fibrose cardíaca.


#120 - Clinical and laboratorial evaluation of Nellore cattle experimentally infected with Trypanosoma vivax, 21(4):157-161

[Clinical and laboratorial evaluation of Nellore cattle experimentally infected with Trypanosoma vivax] Avaliação clínico and laboratorial de bovinos nelore infectados experimentalmente com Trypanosoma vivax. Pesquisa Veterinária Brasileira 21(4):157-161. Embrapa Gado de Corte, Cx. Postal 154, Campo Grande, MS 79002-970, Brazil. In arder to evaluate the clinical-laboratorial alterations, six Neil ore calves were inoculated with 107 Trypanosoma vivax isolated from Poconé region, Mato Grosso, Brazil. The animals were evaluated daily for rectal temperature, packed cell volume (PCV), parasitemia, antibody production, calor of mucous membranes, behavior and appetite. Blood and serum samples for biochemical evaluation for aspartate aminotransferase (AST), alkaline phbsphatase (AF), gamma glutamyltransferase (GGT), cholesterol, urea, creatinine, creatine kinase (CK), calcium, phosphorus and proteinogram were collected on days 4, 8, 12, 16, 23 and 30 post inoculation (DPI). During the following 6 months rectal temperature, PCV and parasitemia were evaluated weekly. T. vivax was evidenced from 1 DPI in all calves and persisted until day 30 in five of six animals. A remarkable decrease (p<0.05) of PCV mean value (25%) was observed on 10 DPI. The animals presented no alterations in their clinical or serum biochemical state during the trial. Seroconversion took place 6 and 8 DPI, and all the animals remained seropositive during the 30 days of experiment. In all the experimental animals the occurrence of T. vivax infection was verified, characterized by the increase of corporal temperature, presente of the blood protozoa and reduction of the globular volume, without altéations in the other variables analyzed. Nellore calves, when experimentally inoculated with T. vivax, are able to establish a balance between host-parasite relationship.

Avaliaram-se as alterações clínico-laboratoriais de seis bezerros Nelore, de ambos os sexos, inoculados experimentalmente com 107 organismos viáveis de Trypanosoma vivax, isolados de bovinos da região de Poconé, Estado de Mato Grosso. Os animais foram observados diariamente, durante 30 dias, quanto aos parâmetros de temperatura retal, volume globular (VG), parasitemia, produção de anticorpos, coloração de mucosas, comportamento e apetite. Deteminaram-se os níveis séricos de aspartato aminotransferase (AST), fosfatase alcalina (FA), gama glutamiltransferase (GGT), creatina kinase (CK), colesterol, uréia, creatinina, cálcio, fósforo e o perfil eletroforético das proteínas séricas aos 4, 8, 12, 16, 23 e 30 dias pós-inoculação (DPI). Durante os 6 meses seguintes, os animais foram observados semanalmente, avaliando-se a temperatura retal, o VG e a parasitemia. T. vivax foi evidenciado a partir do terceiro e quarto DPI em todos os bezerros e persistiu até o 30º DPI em cinco dos seis animais em estudo. Ocorreu um decréscimo significativo (p<0,05) do valor médio do VG (25%) aos dez DPI. Os animais não apresentaram qualquer alteração no quadro clínico, bem como na avaliação da bioquímica sérica durante o período experimental. A soroconversão ocorreu aos 6 e 8 DPI, permanecendo todos os animais soropositivos nos 30 dias experimentais. Bovinos nelores jovens, infectados experimentalmente com T. vivax, foram capazes de estabelecer um equilíbrio na relação hospedeiro-parasita.
