#111 - Experimental poisoning by Brachiaria decumbens in feedlot sheep, 30(3):195-202

[Experimental poisoning by Brachiaria decumbens in feedlot sheep.] Intoxicação experimental por Brachiaria decumbens em ovinos confinados. Pesquisa Veterinária Brasileira 30(3):195-202. E-mail: rlemos@nin.ufms.br Foi realizado estudo experimental de intoxicação por Brachiaria decumbens em ovinos confinados que receberam apenas esta forrageira como alimento. Os animais foram avaliados em três períodos do ano, com duração de 60 dias cada. As análises incluíram exame clínico e amostras de sangue para mensuração da atividade sérica das enzimas gama glutamiltransferase e aspartato aminotransferase com o objetivo de avaliar sua importância e utilidade no diagnóstico de fotossensibilização hepatógena causada pela planta. Foram descritos ainda, achados de necropsia e histopatológicos. Dos vinte e quatro animais confinados, cinco morreram no primeiro período (fevereiro a abril), quatro no segundo (junho a agosto) e dois no terceiro (outubro a dezembro), tendo como principais sinais clínicos anorexia, icterícia, fotofobia, dermatite leve, além de desidratação e apatia. Nos achados de necropsia foram constatados icterícia generalizada, vesícula biliar repleta e distendida, padrão lobular hepático evidente e, em dois casos, opacidade de córnea. As alterações microscópicas foram mais significativas no fígado com bilestase, tumefação e vacuolização de hepatócitos, sinusóides com macrófagos, proliferação de ductos e dúctulos biliares com infiltração linfocítica, que variaram na sua severidade conforme o tempo decorrido da ingestão da planta, além de imagens negativas de cristais no citoplasma de macrófagos presentes nos sinusóides hepáticos e no espaço periportal e cristais refringentes oticamente ativos, ocluindo a luz de ductos biliares. Os achados foram característicos de fotossensibilização hepatógena por B. decumbens, principalmente pela presença de fotofobia, mesmo quando os animais não apresentaram fotodermatite. O aumento da atividade sérica da gama glutamiltransferase teve alta correlação com a morte dos animais. O surgimento da elevação da atividade sérica de gama glutamiltransferase foi, em média, 11 dias antes da constatação dos sinais clínicos, validando sua importância no diagnóstico e na prevenção. A aspartato aminotransferase teve resultados inconsistentes. Treze animais não apresentaram sinais clínicos, apesar de serem constatadas significativas elevações nos níveis séricos das enzimas em questão, sugerindo a existência de indivíduos tolerantes/resistentes dentro do intervalo estabelecido. O modelo experimental foi adequado na caracterização da intoxicação de ovinos por B. decumbens, oferecendo subsídios para estimar preventivamente o risco de morte de ovinos intoxicados pela planta, além de ser útil para a realização de novos estudos da intoxicação por B. decumbens.

E-mail: rlemos@nin.ufms.br Brachiaria decumbens was fed as exclusive source of food to feedlot sheep. Sheep were evaluated in three periods of the year, of 60 days each, regarding their clinical status and was performed collection of blood samples to measure enzymatic activity of gamma glutamyltransferase and aspartate aminotransferase to evaluate their diagnostic importance and usefulness besides the clinical signs, necropsy and histopathology finds in hepatogenous photosensitization Brachiaria related. Out of 24 feedlot sheep, five died in the first period (February-April), four in the second (June-August) and two in the third (October-December). Main clinical signs were anorexia, jaundice, photophobia and two cases of slight dermatitis, besides dehydration, apathy and corneal opacity. Necropsy revealed generalized jaundice, distended gall bladder and liver with evident lobular pattern. The histological changes varied in severity according the time after the beginning of the plant ingestion; they were more significant in the liver and included bilestasis, swelling and vacuolation of hepatocytes, foammy macrophages, bile duct proliferation and mononuclear inflammatory cell infiltration, further crystal negative images were found within cytoplasm of macrophages present inside of hepatic sinusoid and in the periportal space and also optically active refringent crystal in the lumen of bile tubules. These finds are characteristic of hepatogenous photosensitization, mainly by presence of photophobia, even when the sheep did not present photodermatitis. The results of serum biochemistry activity, during the course of poisoning with strong relation between increased gamma glutamyltransferase activity, and lethality. Onset of arising gama glutamyltransferase activity was 11 days, in average, before beginning of clinical signs. Aspartate aminotransferase had increased activity only close to the beginning of clinical signs. Serum biochemistry results of GGT suggest that they are useful for a precocious diagnosis preventing deaths caused by ingestion of B. decumbens. Thirteen sheep didn’t show clinical signs despite significant increases on serum enzyme activity, suggesting the existence of tolerant/resistant individuals. The experimental model of feedlot was adequate to study B. decumbens toxicosis in sheep.


#112 - Fireweed (Senecio madagascariensis) poisoning in cattle, 30(1):10-12

E-mail: vetcruz@terra.com.br In a dairy cattle herd in southern Brazil, 7 out of 554 cattle were affected and died due to Senecio madagascariensis poisoning. Clinical, pathological, and epidemiological findings in the affected cattle were indistinguishable from those usually seen in poisoning caused by other Senecio species. The plant invaded extensive areas in heavily stocked paddocks. Senecio madagascariensis had been spreading in this farm for the last three years, with no control strategy, because neither the farmers nor the local veterinarian knew about the potential risks of this Sernecio species.

E-mail: vetcruz@terra.com.br Em um rebanho bovino leiteiro no Rio Grande do Sul, sete de 554 bovinos foram afetados e morreram devido à intoxicação por Senecio madagascariensis. Aspectos clínicos, patológicos e epidemiológicos observados nos bovinos afetados foram indistinguíveis daqueles usualmente observados em casos de intoxicações por outras espécies de Senecio. A planta invadiu extensas áreas em potreiros mantidos constantemente com altas lotações de bovinos. Senecio madagascariensis esteve se estabelecendo nesta propriedade pelos últimos três anos, sem que qualquer medida de controle fosse adotada pela equipe técnica do estabelecimento, que desconhecia o potencial tóxico dessa espécie de Senecio.

Os corações foram examinados em seu terço médio para acesso ao complexo valvar, que foi submetido a estudos de área, número e tipificação de cordas tendíneas. Os resultados demonstram que este complexo é formado por duas cúspides principais e quatro cúspides acessórias, apresentam em sua formação de 2-3 músculos papilares, nos quais se inserem de 10-25 cordas tendíneas, que se fixam em cúspides voltadas para a parede do ventrículo. A área total deste complexo apresenta uma média de 38,56cm2 com um mínimo de 24,96cm2 e um máximo de 55,54cm2. Estatisticamente não há relação entre número de cordas e a área da cúspide onde estas estão inseridas, nem com o número de músculos papilares dos quais elas provem.

E-mail: ceambrosio@usp.br Atrioventricular valve complex of 30 Jafarabadi water buffaloes, adult males were studied in this research with no heart diseases. The animals were obtained from a slaughterhouse in Brazilian State of Parana. The hearts were opened at the third portion affording access to the valve complex. The complexes had its area, number and type of tendinous cords submitted to analysis. The results showed that the complex is composed by two cusps and four accessory cusps, two or three papillary muscles in which 10-25 tendinous cords fix on the cusps that face the ventricle wall. The total area of the complex was on average 38.56cm2, with a minimum of 24.96cm2 and a maximum of 55.54cm2. Statistically, no relation between the number of cords and the cusps’ area where they are inserted or with the number of papillary muscle where they originated from was observed.

Considerando a importância deste fator no âmbito da reprodução animal foram estudados os anexos embrionários e fetais bovinos fecundados por monta natural de 15-70 dias de gestação, com o objetivo de estabelecer parâmetros morfométricos da placenta na fase inicial da gestação. Com uso de um paquímetro foram realizadas mensurações do comprimento (crânio caudal), largura (latero lateral) e altura (dorso ventral) das membranas corioalantóide e amniótica. O início da formação dos cotilédones foi observado e quantificado, assim como, o peso placentário. O peso médio do saco gestacional aumentou com o evoluir da idade gestacional, entretanto, o crescimento foi acelerado a partir de 20-30 dias de gestação. O comprimento crânio caudal e dorso ventral da membrana corioalantóide e do âmnio apresentaram crescimento lento e gradual com o evoluir dos períodos gestacionais analisados. Com 30-40 dias de gestação, os primeiro cotilédones já eram visualizados e contatos com facilidade na superfície coriônica. Os períodos de crescimento coincidiram com os maiores índices de perdas gestacionais em bovinos. Os parâmetros aqui analisados poderão servir para futuras investigações dos anexos embrionários de organismos manipulados em laboratório.

E-mail: antonioassis@dracena.unesp.br The main goal of this morphometrical study was to characterize the development of the extra-embryonic membranes of 15 to 70-day-old bovine embryos obtained by natural mating. With a millimeter paquimeter the cranio-caudal, latero-lateral and dorso-ventral measurements of chorion and amnion were determined. The development of the cotyledons and weight of the gestational sac were observed and quantified. The weight of the gestational sac increased during gestation; however, the growth was faster then 20 to 30 days. The cranial-caudal and dorsal-ventral length of the choriallantois and the amnion developed slowly and gradually with progress of the gestational period, and the cranio-caudal length decreased after 50 to 60 days of gestation, increasing in the next period. After 30 to 40 days of gestation, the first cotyledons were visualized and counted easily in the chorionic surface. The growth periods were the same of the main gestational losses in cattle. The parameters analyzed could be useful for investigations of extra-embryonic membranes in organisms manipulated in the laboratory.

Clinical signs were abdominal discomfort, increased cardiac and respiratory rate, anorexia, hypermotility of the gut, cecal tympany, and diarrhea. The clinical course was of 18-36 hours. One affected mare survived after symptomatic treatment. Gross lesions were severe congestion, hemorrhages, edema and ulcers of the glandular stomach. Congestion, edema and hemorrhages were also observed in ileum, cecum and large colon. Histologic examination revealed degeneration and necrosis of the aglandular epithelium of the stomach, gastritis and enteritis with infiltration by mononuclear cells and neutrophils, edema of the mucosa, and dilatation of lymphatic vessels. One horse was poisoned experimentally with 1g/kg body weight of B. coridifolia. Clinical signs and lesions were similar to those observed in the spontaneous cases.

E-mail: marinabrito@usp.br Duchenne muscular dystrophy (DMD) is a neuromuscular disorder characterized by a continuous muscle necrosis and degeneration with eventual fibrosis and fatty tissue infiltration. Progressive increase in muscle interstitial fibrosis prevents the movement of myogenic cells, which is necessary for myotube formation. Canine model is the best phenocopies of the disease in humans when comparing with others animal models with dystrophy. Anti-fibrotic treatment of DMD patients, targeting the cytokine mediators, TGF-beta, and the treatment with antiinflammatories, may limit muscle degeneration and contribute for the improvement of the course of the illness. This work aimed to verify the possible adverse effects in renal physiology by means of evaluation sanguineous biochemist and arterial pressure, in order to verifying the viability of Losartan (a TGF-beta inhibiter) in affected dogs by muscle dystrophy. It was used four adults dogs, two of each gender. A dose of 50mg of Losartan was orally given once a day. The clinical exams, the kidney function, arterial blood pressure and potassium level did not show any adverse effect through the experimental period. Losartan utilization showed to be a safe therapy for the antifibrotic treatment in adults dogs, not affecting neither the kidney function nor the arterial blood pressure.

E-mail: crisruiz@scamilo.edu.br The coronary arteries of the goats have been used to researches about to pharmacological actions, heart failure after connection of coronary arteries and others. Twenty-seven goat hearts, both sexes, fixed in formalin at 10% has been used in this study that analyzed the coronary arteries pattern in goats and if the same is similar to the human coronary arteries pattern. The left coronary artery is dominant and in relation to the length demonstrated average 1.15cm emitting two branches: interventricular paraconalis and circumflex. The interventricular paraconalis branch with average length 10.5cm issued branches to the both ventricles fairly, ending more frequently in the interventricular subsinuosis sulcus. The circumflex branch with average length 8.5cm issued branches to left ventricle and right atrium fairly. The left marginal branch is not constant. The right coronary artery, with average length 6.1cm issued branches to right ventricle and right atrium fairly, but the interventricular subsinuosis branch with average length 3.1cm is variable and can present one long branch, one short branch or one double branch, with the huge part for the right ventricle.

E-mail: crisruiz@scamilo.edu.br The coronary arteries of the goats have been used to researches about to pharmacological actions, heart failure after connection of coronary arteries and others. Twenty-seven goat hearts, both sexes, fixed in formalin at 10% has been used in this study that analyzed the coronary arteries pattern in goats and if the same is similar to the human coronary arteries pattern. The left coronary artery is dominant and in relation to the length demonstrated average 1.15cm emitting two branches: interventricular paraconalis and circumflex. The interventricular paraconalis branch with average length 10.5cm issued branches to the both ventricles fairly, ending more frequently in the interventricular subsinuosis sulcus. The circumflex branch with average length 8.5cm issued branches to left ventricle and right atrium fairly. The left marginal branch is not constant. The right coronary artery, with average length 6.1cm issued branches to right ventricle and right atrium fairly, but the interventricular subsinuosis branch with average length 3.1cm is variable and can present one long branch, one short branch or one double branch, with the huge part for the right ventricle.

Golden Retriever dogs show similar genotypic and phenotypic characteristics to Human Muscular Dystrophy and are considered a proper animal model for DMD studies. Latero-lateral and dorso-ventral thoracic radiographies were obtained from 10 Golden Retriever dogs affected by muscular dystrophy, to investigate possible radiographic alterations. Thorax radiographic examination revealed (a) interstitial and alveolar pattern, (b) initial phases of pneumonia and pulmonary edema, (c) cardiomegaly as a principal alteration in the thoracic cavity, (d) megaesophagus displacing the trachea and heart silhouette, and (e) cranial protrusion of the diaphragm lining into the thorax with development of a hiatus hernia displacing the stomach to the caudal mediastinum. Postmortem examination showed pleural effusion, pulmonary emphysema, degenerative and metaplasic processes in the diaphragm and intercostal muscles. Radiographic examination was considered essential for the diagnosis of cardiac and respiratory disease in Golden Retriever dogs affected by muscular dystrophy, and to identify the primary pulmonary process and to provide the establishment of suitable therapeutic treatment, with a reserved prognosis in advanced stage of the disease.

E-mail: davetpat@ufrgs.br Samples of gastric lymph nodes and the stomachs from 24 pigs selected from herds affected by postweaning multisystemic wasting syndrome (PMWS) and sudden death associated with gastric ulcers were studied. Pigs were selected on the basis of unthriftiness, decreased feed intake, and wasting. The stomachs were opened, inverted, and classified into 0-3 score according the severity of the gross lesions present in pars oesophagica (non-glandulargastric mucosa). Selected samples were processed for paraffin embedding and stained with hematoxylin and eosin. Immunohistochemistry using anti-PCV2 (porcine circovírus type 2) antibody, anti-Helicobacter pylori antibody and a wide-spectrum anti-cytokeratin antibody was performed. Gross changes in pars oesophagea were classified according to the severity of lesions as score 3, 2, and 1 in 8, 6, 5 stomachs respectivelly. Microscopically, hyperplastic lymphoid follicles, lymphohistiocytic inflammatory infiltrates and focci of necrosis in the gastric mucosa were common findings. Large amounts of PCV2 antigen were observed in the cytoplasm and nuclei from intralesional cells and debris from the gastric glandular mucosal zone; however, in the fundus, anti-PCV2 immunostaining was restricted to the surface mucosal cells and foveolar compartment. All gastric lymph nodes were positive for PCV2 antigen. Anti-H. pylori immunostaining was seen in eleven cases, mainly in the antrum, on the mucosal surface and foveolar compartment. The association of the anti-PCV2 immunostaining with the glandular mucus-producing cells suggests a role for PCV2 as an additional factor for the swine ulcer development.

Both of them have many cytoplasmic and reticule endoplasmic vesicles on transmission electron microscopy. The allantoic has many vessels filled with eritrocytes, elongated cells which form a stratified simple epithelium. The amnion is transparent or sometimes whitish, consisting of a stratified simple epithelium. The main difference between the allantoic and amnion is that the latter is avascular. The yolk sac is an opaque membrane that disappears during gestation, and is the only membrane that is not in contact with the others; it presents three different types of cells which form three distinct layers (endoderm, mesothelium and mesenquimal).
