The fragment of msp5 gene, except the hydrophobic N-terminal region, was amplified by PCR, cloned in pTrcHis-TOPO plasmid and expressed in Escherichia coli. Solubilization of the recombinant protein was evaluated in different pHs and concentrations of urea. The sensibility and specificity of the assay were evaluated with 66 sera from cattle experimentally-infected and 96 sera from cattle free of A. marginale defined by polymerase chain reaction for msp5 gene. Serum samples from 1,666 cattle from Brazil - states of Rio Grande do Sul (73), Mato Grosso do Sul (91), Pernambuco (86), Bahia (314) and Minas Gerais (267), Uruguay (32) and Costa Rica (803), were tested by ELISAs with recombinant truncated MSP5 and with recombinant MSP1a, and the agreement between both ELISAs was calculated. ELISA with recombinant truncated MSP5 protein detected infected animals with sensibility of 96.97% and specificity of 100%. In cattle experimentally-infected, the ELISA detected antibodies from the 12th day post-infection (DPI) to the end of the experiment, at the 37th DPI. The agreement between the ELISAs with truncated MSP5 and MSP1a antigens was 95.67%, with a kappa index of 0.81. Disagreement results showed significative difference (p <0.001). Antibodies for A. marginale were detected in animals of the all the region analyzed. The ELISA with recombinant truncated MSP5 showed a good performance in ELISA for detention of antibodies against A. marginale, with high sensitivity and specificity, representing an important tool for the diagnosis of anaplasmose bovine in epidemiological studies.

E-mail: bombonat@usp.br The thymus is an organ with great immunological relevance during foetus life and neonatal period, It is the predecessor of lymphopoiesis and shows high lymphopoietic activity, being also the largest lymphoid organ during intrauterine life and at birth. In this study, the morphological aspects of the prenatal development of dog thymus were described in foetus of different ages and sexes. Twenty-four foetus of mongrel domestic dogs, males and females, were divided into four age groups. The thymus presented two lobes linked by a rose-colored tissue connection that was localized in the cranial mediastinal space. The cranial portion protrudes slightly beyond the first pair of ribs. The cells that formed the thymus parenchyma were well organized. Concentric aggregates named Hassal corpuscles were observed, involved by a thin capsule of connective tissue thickened by adipose tissue. The vessels size gradually increased with the foetus age, being larger in animals that were closer to birth. The volume (Vref) and size parameters of female thymus were larger than in males.

Email: fjhblazq@usp.br Two types of probiotics were used in piglets. One product is a mixed culture of viable Lactobacillus acidophilus, Enterococcus faecium e Bifidobacterium bifidum. The second product is composed of inactivated Lactobacillus acidophilus cells. The piglets received two weekly oral doses for 30 days while a control group did not receive probiotics. All piglets were euthanized at the 30th day of life and the mesenteric lymph nodes, the small intestine, and blood samples were collected. The tissue samples were studied by light microscopy and the blood serum was analyzed by ELISA method. The treatment with the probiotic with viable cells produced higher serum levels of IgA (P<0.05) and more IgA expressing cells were found in the mesenteric lymph nodes than observed in the inactivated cells treatment or control groups (P<0.05). Also, intestinal villi were longer, crypts were deeper (P<0.05) and fecal coliform count was lower than found in the inactivated product (P<0.05). These results suggest that viable probiotics are more efficient than inactivated probiotics to induce immunostimulation and intestinal modifications in piglets, thus improving their health and development.

Pesquisa Veterinária Brasileira 27(5):191-193. Departamento de Cirurgia da Faculdade de Medicina Veterinária e Zootecnia, Universidade de São Paulo, Av. Bacteria present in dentinal tubules, apical foramens and apical delta is related to failure of this procedure. Some studies reveal that during endodontic treatment in one session bacteria still remain in dental structures. The aim of the present study was to evaluate the endodontic treatment in one or two sessions, using calcium hydroxide plus camphorated paramonochlorophenol (CMCP) with temporary endodontic medicaments. Four dogs were used, and two animals were submitted the treatment in one session only and the others in two sessions. Premolar teeth were used in this study: second and third upper premolar teeth and second, third and fourth lower premolar teeth were divided into two groups. Opening of the pulp chamber was performed with burns, and the pulp was exposed for 60 days. After this period the group treated in only session was obtured with gutta-percha and zinc oxide and eugenol, and the group treated in two sessions received the CMCP that remained in the canal per 30 days and was afterwards obtured with the material of the other group. Clinical and radiographical control was performed twice a week, and after 60 days a small block containing the teeth, surrounding periapical tissues and the periodontium was removed for histological and microbiological study. Histological analysis revealed intense inflammatory response in both groups. Microbiological analysis showed a more severe microbial reduction in the group treated in one session. However, bacteria were still observed in the two groups, indicating that none of the treatments presented total efficiency.

Four Kirschner wires were inserted through the bone cortices, being two proximally and two distally to the fracture. After the fracture reduction the ends of both proximal and distal wires were twisted in distal or proximal direction, respectively, being the wires connected by two bars of autopolymerizing acrylic resin, in the external lateral and medial faces of the limb. The return to full capability to use the member was observed in 20.00±7.09 days, and the bone healing occurred in 35.12±8.72 days. The results of this study showed that open reduction and use of Type II external skeletal fixator is an effective method for the treatment of tibiotarsus fractures in White Plymouth Rock chickens.

The reduction test was applied through egg counts per gram of feces and larval culture to evaluate resistance. The fecal samples were collected in the day when medication was accomplished (day base), at 7, 14 and 21 days after treatment. The following results for the reduction of Trichostrongyloidea eggs were obtained: Treatment of the females with Moxidectina reduced the egg counts to 92.8, 88.7 and 89.8%, of the males to 92.6, 96.2 and 98.1%; with Levamisol of the females to 96, 97.1 and 91%, of the males to 85.7, 94,2 and 100%; with Albendazol of the females 65, 60.3 and 75.4%, of the males 88.8, 88.8 and 55.5%; with Ivermectin of the females to 92.2, 68.6 and 70.6%, of the males to 41.7, 73.6 and 59.7%; and with the purgative potato of the females to 31.8, 34.1 and 49.4%, of the males to 61.5, 80.7 and 50%. Histopathological changes in liver and gill epithelium of Nile tilapia, Oreochromis niloticus, exposed to waterborne copper. Pesquisa Veterinária Brasileira 27(3):103-109. E-mail: fontain@utad.pt Nile tilapia, Oreochromis niloticus, of both sexes were reared in freshwater and exposed to 0.5, 1.0 and 2.5mg L-1 of waterborne copper for a period of 21 days. Liver and gill samples were collected after 21 days of exposure to copper and lesions were analyzed by light microscopy. The main histopathological changes observed in gills exposed to the highest concentration were edema, lifting of lamellar epithelia and an intense vasodilatation of the lamellar vascular axis. Although less frequent, lamellar fusion caused by the filamentar epithelium proliferation and some lamellar aneurisms were also found. The liver of control group exhibited a quite normal architecture, while the fish exposed to copper showed vacuolation and necrosis. These hepatic alterations were more evident in fish exposed to 1.0 and 2.5mg L-1 copper concentrations. The number of hepatocytes nucleus per mm2 of hepatic tissue decreased with the increase of copper concentration. In contrast, the hepatic somatic index was high in fish exposed at 2.5mg L-1 of copper. In short, this work advance new knowledge as influence of copper in the gill and liver histology of O. niloticus and demonstrated that their effects could be observed at different concentrations.

E-mail: fontain@utad.pt Nile tilapia, Oreochromis niloticus, of both sexes were reared in freshwater and exposed to 0.5, 1.0 and 2.5mg L-1 of waterborne copper for a period of 21 days. Liver and gill samples were collected after 21 days of exposure to copper and lesions were analyzed by light microscopy. The main histopathological changes observed in gills exposed to the highest concentration were edema, lifting of lamellar epithelia and an intense vasodilatation of the lamellar vascular axis. Although less frequent, lamellar fusion caused by the filamentar epithelium proliferation and some lamellar aneurisms were also found. The liver of control group exhibited a quite normal architecture, while the fish exposed to copper showed vacuolation and necrosis. These hepatic alterations were more evident in fish exposed to 1.0 and 2.5mg L-1 copper concentrations. The number of hepatocytes nucleus per mm2 of hepatic tissue decreased with the increase of copper concentration. In contrast, the hepatic somatic index was high in fish exposed at 2.5mg L-1 of copper. In short, this work advance new knowledge as influence of copper in the gill and liver histology of O. niloticus and demonstrated that their effects could be observed at different concentrations.

Fifty samples were collected, using an esophageal tube, during winter (rainy season) and summer (dry season). The predominant colors were an olive green in the rainy season and a nut-brown in the dry season. The smell was aromatic, but was stronger in winter. A slight viscous consistence was found in most of the samples, with a greater proportion in winter. The sedimentation and flotation time was 6.73min (±1.63) in the rainy period and 3.15min (±0.72) in the dry period. In the biochemical tests, the average values found in winter and summer were, respectively: pH 6.76±0.21 and 6,59±0.14; methylene blue reduction, 3.20min (±0.76) and 7.76min (±3.00); chloride, 28.14±4,16mEq/L and 24.97±5.65mEq/L; acidity, 21.90±4.38UC and 13.68±2.97UC. Ruminal microbiotic analyses revealed abundant and moderate density of protozoa in winter and summer, respectively. The motility was very active and there were almost 90% of live protozoa in both seasons. Protozoa numbers were 425.373±217.258/mL in winter and 155.375±83.113/mL in summer. There was a mixed population of bacteria with prevalence of Gram-negative forms in both seasons.

Animals weighing less than 6 kg show well known difficulties regarding radius and ulna fracture repair mainly with bone non-union, due to poor vascularization of the distal portion of the radius. Therefore this study aimed to compare the velocity of bone callus formation in the treatment of fracture repair with plates and screws alone or with plates and screws plus BMP. Thirty three dogs with radius and ulna fractures were distributed into two groups, where animals of the control group received the conservative treatment performed with screws and plates alone, whilst the other group received the conservative treatment and BMP. The time of bone callus formation was evaluated comparatively through radiographic exams 30, 60, 90, 120, 180 and 210 days after the surgical procedure. Animals treated with BMP showed a healing time of 32±15 days, which was significantly different (p <0.001) from the control group which required 127±34 days. With the results obtained, it can be concluded that the distal radio-ulna fractures of dogs weighing less than 6 kg suffered a significant reduction of the bone callus formation time, which was around 90 days.

The materials were tested to evaluate their antimicrobial activity and tissue repair efficacy. Four dogs were used in this experiment, with a total of 64 roots of premolar teeth, divided into three groups. The root canals of Group I were filled with gutta-percha and zinc oxide/eugenol cement; Group II were filled with calcium hydroxide, and Group III were not filled. All animals were clinically and radiographically examined 15 days after surgery and then again every subsequent 15 days until 120 days, when the teeth were extracted en bloc. Histopathological analysis showed inflammatory infiltration, cement and bone resorption and necrotic tissue in the apical delta in different proportions. Histomicrobiological analysis showed the presence of microorganisms inside the teeth structures, with different concentrations according to the treatment used. There was statistical significance between the groups (p>0.05). Gutta-percha with zinc oxide/eugenol demonstrated good antimicrobial activity; calcium hydroxide was not efficient. The conclusion of this study is that gutta-percha with zinc oxide/eugenol is the better protocol for filling root canals in dogs.
