#101 - Cellular and microbiological profiles and risk factors for subclinical mastitis in goats in the semi-arid region of Paraíba, 30(5):379-384

E-mail: sergio.azevedo@pq.cnpq.br A subclinical mastitis study was conducted in nine dairy goat herds in the semi-arid region of Paraíba state, Northeastern Brazil, to determine the occurrence of infection, to evaluate microbiological and cellular profiles of the milk, to test the sensitivity of isolated microorganisms to antimicrobials, and to identify risk factors. One hundred thirty-one dairy goats were used, 261 samples were collected for microbiological culture and 131 samples for somatic cells count (SCC). During collection, the California Mastitis Test (CMT) was conducted and an epidemiological questionnaire was applied for each herd. There was bacterial growth in 30 samples (11.49%), with 25 (83.33%) coagulase-negative Staphylococcus and five (16.66%) Staphylococcus aureus isolated. The SCC mean was 1.39x106 cells/ml. CMT presented low sensitivity (46.7%) and low specificity (60.6%) compared with microbiological culture. Gentamicin and the association of neomycin, bacitracin and tetracyclin were the antimicrobials against which the microorganisms isolated showed 100% sensitivity. Penicillin and ampicillin had the greatest resistance rates (66.67% and 63.89%, respectively). Goat breeding is not the main activity on the farms and do not isolate diseased animals were identified as risk factors for caprine subclinical mastitis. Program for the control and prophylaxis of mastitis must be implemented focusing primarily on hygiene cares at milking and correction of the risk factors identified in this study.

E-mail: sergio.azevedo@pq.cnpq.br Foi realizado um estudo da mastite subclínica em nove rebanhos de cabras leiteiras no semiárido paraibano com o objetivo de determinar a ocorrência da infecção, avaliar o perfil microbiológico e celular do leite, testar a sensibilidade dos microorganismos isolados frente a antimicrobianos além de identificar os fatores de risco. Foram utilizadas 131 cabras leiteiras das quais foram colhidas 261 amostras de leite para exame microbiológico e 131 para contagem de células somáticas (CCS). Na ocasião das colheitas foi realizado o California Mastitis Test (CMT) e aplicado um questionário epidemiológico por propriedade. Houve crescimento bacteriano em 30 amostras (11,49%) com 25 (83,33%) dos isolados identificados como Staphylococcus coagulase negativa e cinco (16,66%) Staphylococcus aureus. A média de CCS foi de 1,39x106 células/ml. O CMT apresentou baixa sensibilidade (46,7%) e baixa especificidade (60,6%) quando comparado ao exame microbiológico. A gentamicina e a associação da neomicina, bacitracina e tetraciclina foram os antimicrobianos contra os quais os microrganismos isolados apresentaram 100% de sensibilidade. Penicilina e ampicilina foram os de maiores índices de resistência (66,67% e 63,89%, respectivamente). A caprinocultura não ser a atividade principal da propriedade e o não isolamento de animais doentes, foram identificados como fatores de risco para a mastite subclínica caprina nas propriedades estudadas. Programas de controle e profilaxia da mastite devem ser implementados enfocando as medidas de higiene na ordenha e correção dos fatores de risco identificados nesse estudo.

E-mail: savasco@usp.br Risk factors associated with leptospirosis were identified in cows in the state of Bahia, northeastern Brazil. A total of 10,823 cows with > 24 months of age from 1,414 herds were randomly sampled. For the serological diagnosis of Leptospira spp. infection, the microscopic agglutination test (MAT) was carried out using 24 serovars as antigens. A herd was considered positive when presented at least one seropositive animal. Of the 1,414 investigated herds, 1,076 (77.9%; 95% CI = 75.7-80.0%) presented at least one reactant animal at MAT to any serovar. Serovar Hardjo (Hardjoprajitno) was the most prevalent, with 34.49% (95% CI = 31.97-37.14%) of the positive herds. Presence of more than 28 bovine females in reproductive age (OR=2.11; p<0.001), presence of cervids (OR=2.02; p=0.010), animal purchase (OR=1.57; p<0.001), to slaughter animals in the property (OR=1.58; p=0.030) and to share pasture (OR=1.63; p<0.001) were identified as risk factors for leptospirosis due to any serovar. Risk factors for leptospirosis due to serovar Hardjo (Hardjoprajitno) were presence of swine (OR=1.28; p=0.040) and animal purchase (OR=1.48; p<0.001).

Um rebanho foi considerado foco quando apresentou pelo menos um animal soropositivo. Das 1.414 propriedades investigadas, 1.076 (77,9%; IC 95% = 75,7-80,0%) apresentaram pelo menos um animal reagente na SAM para qualquer sorovar. O sorovar Hardjo (Hardjoprajitno) foi o mais prevalente, com 34,49% (IC 95% = 31,97-37,14%) das propriedades positivas. Presença de mais de 28 fêmeas bovinas em idade reprodutiva no rebanho (OR=2,11; p<0,001), presença de cervídeos (OR=2,02; p=0,010), compra de animais (OR=1,57; p<0,001), abate de animais na própria fazenda (OR=1,58; p=0,030) e utilização de partos compartilhados (OR=1,63; p<0,001) foram identificados como fatores de risco para leptospirose por qualquer sorovar. Os fatores de risco para leptospirose pelo sorovar Hardjo (Hardjoprajitno) foram a presença de suínos (OR=1,28; p=0, 040) e a compra de animais (OR=1,48; p<0,001).

E-mail: viv_vet@yahoo.com.br Erythrograms aid the veterinary practitioner in the diagnosis of diseases that affect domestic animals. However, hematological studies involving buffaloes are scarce, with little information in the literature on reference values for blood components in this species. Therefore, the objective of this study was analyze erythrograms of Murrah breed buffaloes of different ages, both males and females. Buffaloes in the study were distributed into four experimental groups, according to their ages: Group 1, animals from birth to 3 months of age (n=15); Group 2, animals from 4 to 6 months of age (n=50); Group 3, animals from 7 to 12 months of age (n=50); and Group 4, animals from 13 months to five years of age (n=50). The following values were obtained, for Group 1: 7.9x106 erythrocytes/mL (He), 13.0g/dL hemoglobin (Hb), hematocrit (Ht) 38.9%, Mean Corpuscular Volume (MCV) 49.0 fl, Mean Hemoglobin Corpuscular Concentration (MCHC) 33.6%, and Mean Corpuscular Hemoglobin (MCH) 16.4 pg; for Group 2: He7.1x106/mL, Hb12.5g/dL, Ht 36.8%, MCV 52.4 fl; MCHC 33.9%; and MCH 17.8 pg; for Group 3: He 7.9x106/mL, Hb12.0g/dL, Ht 33.8%, MCV 43.1 fl, MCHC 35.4%, and MCH 15.34pg; for Group 4: He 6.7x106/mL, Hb 11.7g/dL, Ht 34.4%, MCV 53.4 fl, MCHC 34.4%, and MCH 17.4 pg. Statistical analysis of the results for the different age ranges permitted the conclusion that when the animals got older there was a decrease in the mean red cell counts (He), hemoglobin concentration (Hb) and hematocrit values (Ht). Although hematometric indexes showed significant variations in G3.

No entanto, estudos hematológicos em bubalinos são escassos, havendo poucas informações na literatura a respeito de valores de referência dos constituintes sanguíneos desta espécie. Assim, este trabalho teve como objetivo a análise do eritrograma de búfalos de diferentes faixas etárias da raça Murrah, machos e fêmeas. Os animais em estudo foram distribuídos em quatro grupos experimentais, de acordo com as idades: Grupo 1: animais com idade entre o nascimento e 3 meses (n=15); Grupo 2: animais com idade entre 4 e 6 meses (n=50); Grupo 3: animais com idade entre 7 e 12 meses (n=50); e Grupo 4: animais com idade superior a 12 meses (n=50). Como resultados das análises foram encontrados os seguintes valores médios, no Grupo 1: 7,9x106 hemácias/mL de sangue (He); 13,0 g/dL de hemoglobina (Hb); hematócrito (Ht) de 38,9%; Volume Corpuscular Médio (VCM) de 49,0 fl; Concentração Hemoglobínica Corpuscular Média (CHCM) de 33,6 %, e Hemoglobina Corpuscular Média (HCM) de 16,4 pg; no Grupo 2: He: 7,1 x106/mL; Hb: 12,5 g/dL; Ht: 36,8%; VCM: 52,4 fl; CHCM: 33,9%, e HCM: 17,8 pg; no Grupo 3: He: 7,9 x106/mL; Hb: 12,0 g/dL; Ht: 33,8%; VCM: 43,1 fl; CHCM: 35,4%, e HCM: 15,34 pg; e no Grupo 4: He: 6,7 x106/mL; Hb: 11,7 g/dL; Ht: 34,4%; VCM: 53,4 fl; CHCM: 34,4%. e HCM: 17,4 pg. E-mail: karinamedici@yahoo.com.br The particular apocrine secretion of goat milk different from the merocrine one observed in cows, may lead to errors in interpreting cellularity evaluations in the milk of this species. Thus, the objective of the present trial was to determine Somatic Cell Counts by means of one indirect methods, the California Mastitis Test (CMT), and direct methods, flow cytometry and direct microscopic count using methyl green-pyronine-Y stain, beyond comparing the methods of cellular counting. A total of 102 samples from 51 Saanen, Brown Alpine and Toggenburg female goats, bred in the state of São Paulo, were analyzed. Goats were separated in groups according to the phase of lactation and to physical examination of the mammary gland, and milk examination. Samples were divided into two aliquots, and were collected after California Mastitis Test evaluation. One aliquot was used in automatic cell counts, and the other, in direct microscopic count using methyl green-pyronine-Y stain. CMT results were as follows: 74.5% of the samples were negative, 8.8% yielded traces, 8.8% were weak positive (1), 6.8% were distinct positive (2) and 0.9% were strong positive (3). Medians of somatic cell counts in goat milk as evaluated by means automatic cell counter and direct microscopy, and grouped according to the different CMT scores, were as follows: 181,000, 578,000, 628,000, 1,421,500, and 5,542,000 cells/mL of milk and 74,991, 271,396, 71,420, 640,995, and 5,049,394 cells/mL of milk in scores negative, traces, 1, 2 and 3, respectively. Medians obtained in automatic cell counts and direct microscopic counts, grouped according to the phase of lactation were 159,500; 508,000; and 277,500 cells/mL of milk and 62,493; 89,275; and 146,411 cells/ml of milk, respectively. The correlation between the automatic and microscopic methods for somatic cell counts was 88%. Based on the results obtained, it could be concluded that there were differences between the automatic and microscopic methods for somatic cell counts, being this most adequate for the determination of the celularidade in the cellularity of goat milk.

E-mail: karinamedici@yahoo.com.br A particularidade da secreção láctea caprina, do tipo apócrina, diferente da secreção merócrina da vaca, leva a erros de interpretação durante a realização de técnicas de avaliação da celularidade do leite de fêmeas desta espécie. Portanto, o presente trabalho teve o objetivo de determinar a contagem de células somáticas pelo método indireto California Mastitis Test (CMT), e por métodos diretos, incluindo a contagem por citometria de fluxo e a contagem microscópica direta, através da coloração de verde de metil e pironina-Y, além de comparar os métodos de contagem celular. Foram analisadas 102 amostras de 51 fêmeas caprinas, das raças Saanen, Parda Alpina e Toggenburg, criadas no Estado de São Paulo. Os animais foram categorizados segundo a fase da lactação, exame físico da glândula mamária e exame do leite. As amostras foram colhidas, após a realização do exame Califórnia Mastitis Test, em duas alíquotas, uma destinada à contagem celular automática e a outra, a contagem microscópica direta, utilizando-se o corante verde de metil e pironina-Y. De acordo com os diferentes escores do CMT, observou-se 74,5% de amostras negativas, 8,8% de amostras com escore traços, 8,8% de amostras ligeiramente positivas (+), 6,8% de amostras fracamente positivas (++) e 0,9% de amostras fortemente positivas (+++). Os valores medianos das contagens de células somáticas presentes no leite de cabras, avaliadas através de contador automático e microscopia direta, e analisadas de acordo com os diferentes escores do CMT, foram, respectivamente, 181.000, 578.000, 628.000, 1.421.500 e 5.542.000 células/mL de leite e 74.991, 271.396, 71.420, 640.995 e 5.049.394 células/mL de leite, nos escores negativo, traços, +, ++ e +++. Os valores medianos obtidos através da contagem de células somáticas pelo método automático e microscópico direto, de acordo com as fases de lactação foram de 159.500, 508.000 e 277.500 células/mL de leite, e 62.493, 89.275 e 146.411. A correlação obtida entre a contagem celular automática e microscópica direta foi de 88%. A partir dos resultados observados pode-se concluir que existe diferença na contagem celular determinada através do método automático e microscópico sendo este último o mais adequado para a determinação da celularidade no leite de cabras.


#105 - Treatment of dermatophytosis in cats with lufenuron?, 30(2):132-138

Pesquisa Veterinária Brasileira 30(2):132-138. Departamento de Medicina e Cirurgia Veterinária, Instituto de Veterinária, Universidade Federal Rural do Rio de Janeiro, Seropédica, RJ 23890-000, Brazil. E-mail: regina@vetskin.com.br Due to discrepancies regarding the effectiveness of lufenuron in treating dermato-phytosis caused by Microsporum canis the effect of this drug was checked in 46 cats (30 with cutaneous lesions and 16 asymptomatic carriers) treated in the Veterinary Hospital of the Federal Rural University of Rio de Janeiro, Brazil. The diagnosis was based on Wood’s lamp examination and fungal cultures. Biopsies were only performed in symptomatic animals. The animals were treated with lufenuron (120mg/kg every 21 days), for 4 times. The drug was efficient in 29 of the 30 affected felines and as well as in all of the asymptomatic carriers. The cat that did not respond, had received several dexameta-zone doses prior to the treatment with lufenuron. The drug was given to one animal during the first stages of pregnancy and no abnormalities or neonatal disorders were found in any of the kittens. None of the treated animals showed side effects. Twenty days after the last administration of lufenuron, 45 of the studied animals (98%) had negative fungal culture. The cost of treating dermatophytosis with lufenuron is a little higher than with ketoconazole, but the drug has some advantages regarding its practicability and security. The correct drug prescription and animal’s medication as well as environmental decontamination are very important for the success of the treatment.

Pesquisa Veterinária Brasileira 30(2):132-138. E-mail: regina@vetskin.com.br Em função de controvérsias sobre a eficácia do lufenuron no tratamento da dermatofitose causada por Microsporum canis, o efeito da droga foi avaliado em 46 gatos (30 com lesões cutâneas e 16 portadores assintomáticos) atendidos no Hospital Veterinário da Universidade Federal Rural do Rio de Janeiro, Brasil. O diagnóstico foi estabelecido através da lâmpada de Wood, da cultura fúngica e, adicionalmente, na maioria dos animais sintomáticos, pela avaliação histopatológica da pele. Os animais foram tratados com 120mg/kg de lufenuron, a cada 21 dias (quatro doses); a droga mostrou-se eficaz no tratamento de 29 dos 30 felinos que apresentaram a forma clínica da dermatofitose, bem como no de todos os animais assintomáticos. O único felino que não teve cura clínica completa havia recebido várias doses de dexametasona antes do inicio do tratamento. Em um animal gestante, foram utilizadas duas doses do lufenuron e não foi observada qualquer alteração clínica e morfológica nos filhotes. Nenhum dos animais tratados teve qualquer reação adversa ao medicamento. Vinte dias após o último tratamento, o exame micológico resultou negativo em 45 dos felinos estudados (98%). Embora o lufenuron tenha o custo um pouco mais elevado do que o do cetoconazol, é uma droga que apresenta praticidade e margem de segurança bem maiores. Vale ressaltar que o sucesso do tratamento está diretamente relacionado à correta utilização da droga, assim como a perfeita higienização do ambiente e dos utensílios dos animais.


#106 - Occurrence of parasitism by Dioctophyma renale in ring-tailed coatis (Nasua nasua) of the Tiete Ecological Park, São Paulo, Brazil, 29(12):959-962

E-mail: lmilanelo@gmail.com Dioctophymosis is a worldwide renal parasitosis caused by the Dioctophyma renale nematode, which results in progressive destruction of renal tissue. Aquatics annelids are considered the main intermediate hosts and the literature refers as permanent hosts of dogs, wild mammals and even humans. During procedures for population control of coatis (Nasua nasua) in the Ecological Park of Tietê (PET), was noticed the presence of parasitosis by D. renale. From 68 animals, males and females, young and adults, submitted to exploratory laparotomy, 51 were positive for the presence of worms, 9 were found only in the right kidney. In 10 cases, in addition to right kidney parasitism, worms were also observed in the abdominal cavity. In 24 cases D. renale was found only in the abdominal cavity and in 8 animals the right kidney was reduced to a small rigid structure. The study showed that the preferred site for parasitism of the worm, considered erratic, was the abdominal cavity in 66.66% of the cases.

E-mail: lmilanelo@gmail.com RESUMO.- [Ocorrência de parasitismo por Dioctophyma renale em quati (Nasua nasua) do Parque Ecológico Tietê, São Paulo.] A dioctofimose é uma parasitose renal causada pelo nematóide Dioctophyma renale conhecida por gerar a destruição progressiva do parênquima renal. Anelídeos de água doce são considerados os principais hospedeiros intermediários e a literatura refere como hospedeiros definitivos cães domésticos, mamíferos selvagens e até seres humanos. Durante procedimentos de controle populacional de quatis (Nasua nasua) no Parque Ecológico do Tietê (PET), evidenciou-se a presença do parasitismo por D. renale. Sessenta e oito animais foram submetidos à laparotomia, machos e fêmeas, jovens e adultos, dos quais 51 foram positivos para presença do parasita. Em 9 animais o parasita esteve presente apenas no rim direito; em 10 animais D. renale parasitava o rim direito e a cavidade abdominal simultaneamente. Em outros 24 quatis o parasita foi encontrado apenas na cavidade abdominal e em 8 animais o rim direito foi reduzido apenas a uma pequena estrutura rígida. O estudo demonstrou que para este hospedeiro (Nasua nasua), o rim direito não parece ser local de escolha do parasita, já que o encontro do verme em 66,6% dos casos foi a cavidade abdominal.

The animals had the arterial system filled with aqueous colored solution in a 50% of Neoprene Latex, and than were fixed in 10% aqueous solution formaldehyde. The thoracic lobes of thymus were vascularization by direct branches of the right (53.33%) and left (63.33%) internal thoracic arteries, left subclavian (3.33%), left vertebral (3.33%), right (3.33%) and left (3.33%) superficial cervical, left common carotid (3.33%), right coronary (3.33%) and brachiocephalic trunk (33.33%) and costocervical trunk (3.33%). The right internal thoracic arteries (70%) and left internal thoracic (76.67%), left subclavian (23.33%), left superficial cervical (3.33%) of the aortic arch (6.67%) provided indirect branches for the correspondent thoracic lobes of the thymus in each side.

Interpretation of the CCT was checked by calculation of the difference obtained between skin measures correspondent to respective tuberculins, before and 72 hours after inoculation: negative, when bovine PPD is smaller than avian PPD or larger in up to 1.8mm; suspicious or inconclusive, reaction to bovine PPD larger than avian PPD between 1.9 and 2.4mm; positive, reaction to bovine PPD larger than or equal to avian PPD in 2.5mm. From goats submitted to CCT, 0.47% (9/1866) reacted positively (95% CI = 0.23%-0.94%). The prevalence of positive herds to caprine tuberculosis was 10.71% (95% CI = 5.02-19.37%). When submitted to necropsy and histopathologic examination, a positive goat at tuberculin test presented tuberculosis-like lesions. The presence of Acid-Alcohol Resistant Bacilli (BAAR) was also detected by direct bacterioscopy; in relation to herd size, herds with larger number of animals (>25) with high incidence of the disease, and presence of the infection was evidenced in properties that raised cattle in association with goats and did not carry out tuberculin tests in cattle.

Serum samples (n=290) were analyzed to detect Chlamydophila spp. antibodies in 12 farms. Questionnaires were applied to identify risk factors. Frequency of serum-reactive animals were 10.3% (12.0% in ewes and 8.1% in goats) and 1/12 (91.6%) infection focuses were identified. This is the first report on anti-Chlamydophhila abortus antibodies in goats and sheep in Pernambuco and Brazil, respectively. Risk factors associated with goat infection were breed (OR=9.10) and management (OR=6.41). No significant associations in any of the analyzed factors were found for sheep. In summary, Chlamydophila sp. infection is disseminated in sheep and goat herds in the region. Control measures should be established, focusing primarily risk factor identified in this study, to reduce the possibility of infection by the agent.
