#251 - Effects of infection with Toxoplasma gondii oocysts on the intestinal wall and the myenteric plexus of chicken (Gallus gallus), 30(9):787-792

E-mail: debora@unipar.br This paper aims to analyze the effects of the Toxoplasma gondii infection in the intestinal wall and myenteric plexus of chicken (Gallus gallus). Ten 36-day-old chickens were separated into two groups: control and experimental, orally inoculated with oocysts of the T. gondii strain M7741 genotype III. After 60 days the birds were submitted to euthanasia and had their duodenum removed. Part of the intestinal segments was submitted to histological routine, HE staining, PAS histochemical technique, and Alcian Blue. Qualitative analysis of the intestinal wall and comparative measurements among the groups with respect to total wall thickness, muscle tunic, mucosa, and tunica mucosa were carried out. Caliciform cells were quantified. The other part of the intestinal segments was fixed in formol acetic acid and dissected having the tunica mucosa and the tela submucosa removed. Neurons were stained with Giemsa, counted, and measured. Chickens from the experimental group presented diarrhea and inflammatory infiltrates in the tunica mucosa, thickness reduction of all the parameters assessed in the intestinal wall, and an increase of the number of caliciform cells. There was a ~70% reduction regarding the intensity of myenteric neurons; and the remaining cells presented a reduction of ~2.4% of the perikarion and ~40.5% of the nucleus (p<0.05). Chronic infection induced by T. gondii oocysts resulted in intestinal wall atrophy, mucin secretion increase, death and atrophy of chicken myenteric plexus neurons. Death and atrophy of myenteric plexus neurons may be related with the causes of diarrhea observed in chickens with toxoplasmosis.

E-mail: debora@unipar.br O objetivo deste trabalho foi analisar os efeitos da infecção pelo Toxoplasma gondii sobre a parede intestinal e o plexo mientérico de Gallus gallus. Dez galinhas de 36 dias de idade separadas em dois grupos: controle e experimental inoculado com oocistos da cepa M7741 de T. gondii (genótipo III) pela via oral. Após 60 dias os animais foram submetidos à eutanásia e o duodeno coletado. Parte dos segmentos intestinais foi submetida à rotina histológica, coloração por HE e técnica histoquímica de PAS e Alcian Blue. Realizou-se uma avaliação qualitativa da parede intestinal e medidas comparativas entre os grupos da espessura da parede total, túnica muscular, muscular da mucosa e túnica mucosa. As células caliciformes foram quantificadas. Outra parte dos segmentos intestinais foi fixada em formol acético e dissecada retirando-se a túnica mucosa e a tela submucosa. Os neurônios foram corados pela técnica de Giemsa, contados e mensurados. Os animais do grupo experimental apresentaram diarréia e infiltrados inflamatórios na túnica mucosa, redução da espessura de todos os parâmetros avaliados da parede intestinal e aumento do número das células caliciformes. Houve uma redução de ~70% da densidade dos neurônios mientéricos e as células remanescentes sofreram redução de ~2,4% do pericário e ~40,5% do núcleo (p<0,05). A infecção crônica induzida por oocistos de T. gondii levou a atrofia da parede intestinal, aumento da secreção de mucinas, morte e atrofia dos neurônios do plexo mientérico de galinhas. A morte e atrofia dos neurônios do plexo mientérico podem estar envolvidas na causa da diarréia observada em galinhas com toxoplasmose.

E-mail: dayseabreu@predialnet.com.br The pathogenicity of Escherichia coli strains is partially related to the expression of virulence factors genes, present in genetic elements called plasmids. APEC strains responsible for diseases in birds may present the iss gene which increases the resistance of E. coli strains to the lityc effect of the host’s serum, besides resistance to several antimicrobials. This study was conduced in order to detect E. coli in tracheae of meat-type quails and to evaluate, by the presence of the iss gene and the profile of antimicrobial susceptibility, the pathogenic potential of the isolated samples for birds and humans. One hundred and eighty tracheae of quails were collected for detection of E. coli, antimicrobial sensitivity tests, and for polymerase chain reaction (PCR), for detection of iss gene. From the examined quails, 8.9 % (16/180) were positive for E. coli, from which 20 strains of this bacterium were obtained. Most of them were resistant to Tetracycline (16/20), followed by Ceftadizime (13/20) and Nalidixic-acid (12/20) and only one isolate was resistant to Amoxicillin. The detection of iss gene occurred in 55% (11/20) of the isolates, indicating that these strains had the potential to be pathogenic not only for quails, but also for other kinds of birds, other animals and even human beings that would be in contact with these E. coli isolates.

O patotipo APEC, responsável por diferentes tipos de doenças em aves, pode apresentar o gene iss que aumenta a resistência das cepas de E. coli aos efeitos líticos do soro, além da resistência a diversos antimicrobianos. Este estudo foi conduzido para detectar E. coli em traquéias de codornas destinadas ao abate e avaliar, pela presença do gene iss e o perfil de susceptibilidade antimicrobiana, o potencial patogênico para aves e humanos dos isolados obtidos. Foram coletadas 180 traquéias de codornas para detecção de E. coli, determinação do perfil de resistência a agentes antimicrobianos e posterior detecção, por reação em cadeia da polimerase (PCR), do gene iss. Das traquéias analisadas, 8,9 % (16/180) foram positivas para E. coli, sendo obtidos 20 isolados deste agente. A maioria dos isolados foi resistente à Tetraciclina (16/20), seguida pela Ceftazidima (13/20) e Ácido Nalidíxico (12/20), sendo apenas um resistente à Amoxicilina. A detecção do gene iss ocorreu em 55% (11/20) dos isolados. A presença do gene iss e a resistência a múltiplos antimicrobianos dos isolados obtidos neste estudo pode indicar um possível potencial patogênico das cepas de E. coli tanto para codornas quanto para outros tipos de aves e animais e mesmo para o ser humano que fique em contato com as mesmas.

E-mail: lucasbmoraes@gmail.com Fifty Bursa of Fabricius (BF) were examined by conventional optical microscopy and digital images were acquired and processed using Matlab® 6.5 software. The Artificial Neuronal Network (ANN) was generated using Neuroshell® Classifier software and the optical and digital data were compared. The ANN was able to make a comparable classification of digital and optical scores. The use of ANN was able to classify correctly the majority of the follicles, reaching sensibility and specificity of 89% and 96%, respectively. When the follicles were scored and grouped in a binary fashion the sensibility increased to 90% and obtained the maximum value for the specificity of 92%. These results demonstrate that the use of digital image analysis and ANN is a useful tool for the pathological classification of the BF lymphoid depletion. In addition it provides objective results that allow measuring the dimension of the error in the diagnosis and classification therefore making comparison between databases feasible.

Redes Neurais Artificiais (ANN) foram geradas com a utilização do software Neuroshell® Classifier, e os dados das análises óptica e digital foram comparados. A ANN classificou corretamente a maioria dos folículos, atingindo sensibilidade e especificidade de 89% e 96%, respectivamente. Quando os folículos foram agrupados de forma binária houve um aumento da sensibilidade para 90% e obteve-se um valor máximo para a especificidade de 92%. Estes resultados demonstram que o uso da análise digital de imagem associada à ANN é uma ferramenta bastante útil para a classificação patológica da depleção linfóide da BF. Além disso, fornece resultados objetivos que permitem medir a dimensão do erro classificatório, tornando possível a comparação entre distintos bancos de dados.

E-mail: olney@uenf.br Pythiosis is caused by Pythium insidiosum and the occurrence of disease in horses was described in the North and Northwest State of Rio de Janeiro, Brazil. The disease was described in cattle, sheep, humans, and horses in different states and regions across the country. This paper describes the development of IgY and IgG polyclonal antibodies, in chicken and rabbits, respectively against proteins extracted from kunkers and hyphae of P. insidiosum from affected horses. The proteins were recognized by chicken, rabbit and horse antibodies by immunodiffusion and Western blot against majority bands of 27 and 43 KDa, and titrated by ELISA. The antibodies IgY developed by the first time against Brazilian strains of P. insidiosum may represent a valuable tool in the detection of antigens of the pathogen and contribute to further studies aimed at immunotherapy and knowledge about this disease in endemic areas in Rio de Janeiro and in Brazil.

E-mail: ana6113@hotmail.com A retrospective study was conducted to identify the occurrence and types of ocular disorders in 57 Amazon parrots admitted to the Ophthalmology Service, Veterinary Teaching Hospital, School of Veterinary Medicine, University of São Paulo, Brazil from 1997 to 2006. The most frequent observed disorder was cataracts, present in 24 of the 114 examined eyes (57 parrots). Uveitis, ulcerative keratitis and keratoconjunctivitis were frequently diagnosed as well. The cornea was the most affected ocular structure, with 28 reported disorders. Uveal disorders also were commonly observed. Conjunctiva and eyelid disorders were diagnosed in lower frequency. Results suggest that cataracts are common and that cornea, lens and uvea are the most affected ocular structures in Amazon parrots.

E-mail: ana6113@hotmail.com Realizou-se estudo retrospectivo para identificar a ocorrência e os tipos de alterações oculares observadas em 57 papagaios atendidos no Serviço de Oftalmologia do Hospital Veterinário da Faculdade de Medicina Veterinária e Zootecnia da Universidade de São Paulo (USP), de 1997-2006. Catarata em diferentes estágios de evolução foi a alteração ocular mais frequentemente diagnosticada, sendo observada em 24 dos 114 olhos examinados. Uveíte, ceratite ulcerativa e ceratoconjuntivite foram também diagnosticadas. A córnea foi a estrutura ocular mais acometida (28 registros). Alterações uveais foram frequentemente observadas. Alterações das pálpebras e conjuntiva foram observadas em menor freqüência. Concluí-se que catarata foi a alteração mais frequentemente observada e que a córnea, lente e úvea são as estruturas oculares mais susceptíveis a alterações em papagaios.

Pesquisa Veterinária Brasileira 29(9):725-730, Universidade Estadual de Maringá, Av. Colombo 5790, Maringá, PR 87020-900, Brazil. E-mail: aemurakami@uem.br An experiment was carried out to evaluate the effect increasing levels of yeast wall and broiler breeders’ age levels on lymphoid organs weight, cellular immune response and hematological parameters in broilers. A total of 3,360 Cobb broilers were allotted, in a completely randomized design and a 2x5 factorial arrangement, and two controls, compound of two broiler breeders age (34 and 57 weeks of age) and five yeast wall levels (zero, one, two, three and four kg of yeast wall/ton of diet). Broiler breeders’ age affected all studied variables. The inclusion of 3kg of yeast wall/ton of meal increased, at 57 weeks age broiler breeders, more intense inflammatory reaction when compared to control diet; however the circulated heterophils and lymphocytes numbers were not increased. In conclusion, the use of yeast wall, associated or not to sorghum on broilers diet still needs complementary studies, for example, purified components of yeast wall (MOS and ß-glucano).

E-mail: aemurakami@uem.br Um experimento foi conduzido para avaliar o efeito de níveis crescentes de parede de levedura e idade das matrizes reprodutoras sobre o peso dos órgãos linfóides, a resposta imune celular e o perfil hematológico de frangos de corte. Foram utilizados 3.360 pintos de corte da linhagem Cobb, distribuídos em delineamento inteiramente casualisado, em esquema fatorial 2x5, mais dois controles, sendo duas idades de matrizes (34 e 57 semanas de idade) e cinco níveis de suplementação de parede de levedura (zero, um, dois, três e quatro kg de parede de levedura/tonelada de ração). A idade das matrizes influenciou a resposta de todas as variáveis. A inclusão de 3kg de parede de levedura/tonelada de ração promoveu, na progênie de reprodutoras de 57 semanas, reação inflamatória mais intensa quando comparada a dieta controle, no entanto não houve aumento significativo no número de heterófilos e linfócitos circulantes. Conclui-se que a utilização da parede de levedura associada ao sorgo ou não em rações de frangos de corte ainda necessita de estudos complementares, que incluam, por exemplo, os componentes purificados da parede de levedura (MOS e ß-glucano).

E-mail: danielcgarcia@hotmail.com “Inflow Occlusion” technique can be used in heart surgeries when heart is required to be opened just for few minutes, to allow quick repairs. However, circulatory arrest, event occasioned by this technique, may produce serious metabolic and neurological consequences to the patient. In this study, 12 mongrel dogs were used, divided into two groups, A and B, which were submitted to 7 and 8 minutes of total circulatory arrest, respectively, using “Inflow Occlusion” technique. Normothermia was tried during surgical procedures. Clinical and behavior evaluation were performed after surgery to both groups, and biochemical data were collected to compare pre and post-operatory moments. There were two transoperatory deaths in Group B. Transitory clinical problems were observed in group A until moment M7 (48 hours after surgery), and in Group B these problems were more intense and seen even after M7; and permanent blindness in one animal of Group B was present during all follow up period. Despite all alterations found during the study, it might be safe to use “Inflow Occlusion” technique for periods up to 7 minutes, however, it is contra-indicated for longer periods.

Transitory clinical problems were observed in group A until moment M7 (48 hours after surgery), and in Group B these problems were more intense and seen even after M7; and permanent blindness in one animal of Group B was present during all follow up period. Despite all alterations found during the study, it might be safe to use “Inflow Occlusion” technique for periods up to 7 minutes, however, it is contra-indicated for longer periods.

Within these attended birds and its respective orders, most morbid conditions occurred with the Psittaciforme order and were caused by traumatism, in 45 cases (14.32%). Of these, 17 animals (30.91%) had bone fractures; the most common were on radius-ulna (17.65%). Other conditions were presence of ectoparasites (12.50%) and endoparasites (10.68%), respiratory disease (10.42%), dermatological ailments (6.51%), neoplasms (4.95%), ophthalmic illnesses (4.43%), gastrointestinal diseases (3.91%), cachexia (3.39%), neurological diseases (2.86%), self-mutilation (2.86%), obesity (2.34%), aggression by other animals (1.56%), nutritional deficiencies (1.30%), egg retention (1.04%), avian poxvirus (0.78%) and uric gout (0.52%), as well as there was clinical routine measures to be taken (7.55%). Traumas and illnesses were observed as the highest prevalence on the casuistic, which could be prevented by correct husbandry practices.

A total of 36 Campolina and Mangalarga Marchador mares in reproductive age (3-23 years) were used, 11 of them were healthy mares (Control group, 7 embryo recipient and 4 embryo donors), and 25 repeat breeders (10 embryo recipient and 15 embryo donors), classified as based on their reproductive history. Endometrial and blood samples were collected for respectively histological and plasma progesterone concentration evaluation. The endometrial samples obtained after biopsy were fixed in Bouin’s fluid, processed, included in paraffin, and stained with Hematoxylin-Eosin (HE) for histopathological examination. Plasmatic progesterone concentrations were evaluated by enzyme immunoessay (ELISA). There was no correlation between progesterone concentration and fertility. But there was a positive correlation between age and fertility, as older mares had major tendency of subfertility than younger ones. There was also a correlation between biopsy categories and fertility, as more histological alterations were found, higher were the chances for the mares to be subfertile. However not all mares classified as Category I and II maintained pregnancy until parturition. Other factors could influence pregnancy maintenance. In the same way, not all mares in Category III were infertile. The endometrial biopsy was shown to be an easy and cheap diagnostic technique with minimal discomfort to the animals and, together with other data, to be a very important component in the investigation of mare fertility.

The corpuscles are composed of an outer core, inner core and central nerve terminal. The outer core presents numerous lamellae, while the inner core shows compact structure of cytoplasm sheets. The corpuscles are elongate or oval in shape and are surrounded by bundles of collagen fibers. Each lamella is composed of a dense network of thick fibrils. The terminal axons are located along the axis and form a bulb terminal. The fibers of external core stained by Picrosirius and examined by polarized light microscopy revealed to be green in color like type I collagen fibers, and at the periphery is a large amount of collagen type III. The corpuscles are surrounded by flat cells and dense collagen fibers at the periphery.
