#161 - Fatores de risco associados à ocorrência de linfadenite em suínos na fase de crescimento-terminação

E-mail: armando@cnpsa.embrapa.br An epidemiological study in 60 units of swine termination (UT), pertaining to producers integrated with the main Agribusiness of the States of Santa Catarina and Rio Grande do Sul. These units had been selected due to the occurrence of lymphadenitis in slaughtered pigs. The batch size studied in each UT varied from 22 to 960 pigs. The animals were considered positive or negative for lymphadenitis, following the evaluation routine of the Service of Federal Inspection (SIF) at slaughter. For data collection at the UTs, a questionnaire with 156 variables was applied at a single visit to the farms. In the 60 UT studied, the percentage of positive animals for lymphadenitis was 10.37%. Among the variables analyzed, nine risk factors were identified (explicative variables) and associated with the occurrence of lymphadenitis (objective variables). The explicative variables were: bad hygiene of the feeders and drinking fountains, untreated drinking water, bad conservation of the facilities, use of the same truck for ration and animal transportation, production of ration inside the farm, free access of other animals to the ingredients of ration, storage of ready usable ration in boxes or bags and handling of the production of animals in an continuous system.

#162 - Antimicrobial resistance and R-plasmid in Salmonella spp from swine and abattoir environments

E-mail: nslazaro@aol.com Salmonella serovars isolated from swine are of particular interest not only because of the pathogenic potential for this animal species, but also due to its relevance with regard to public health. On basis of the profile of resistance to antimicrobials, 13 Salmonella strains were selected which belonged to the serovars Muenster (7), Derby (4), Typhimurium (1), and Braenderup (1). They were isolated from healthy swine as well as from the abattoir environment in the state of Rio de Janeiro. All strains of Salmonella were subjected to bacterial conjugation, and the E. coli K12 Nalr Lac+ F standard strain was used as receptor, with the purpose to verify the ability to transfer the resistance marks. Gene transfer phenomenon was detected in seven strains, and except Salmonella Typhimurium which transconjugated to Sm, Tc and Su, the remaining ones were characterized by transferring mark Su only. By plasmidial analysis of strains used and their respective transconjugants, 63 Kb plasmid was found, which was probably related to S. Typhimurium resistance.

E-mail: nslazaro@aol.com Thirty-eight strains of Salmonella Muenster, isolated from swine and the abattoir environment, in the State of Rio de Janeiro, Brazil, from March 1991 to February 1992, were analyzed for the presence of plasmids. The strains were selected according to their profile regarding the antimicrobials: streptomycin, tetracycline, sulphonamide and sulfametoxazole-trimethoprim. Thirteen strains were resistant to one or several antimicrobials, 18 with intermediate degree and seven were sensitive. Plasmids varying in size from 1.2 Kb to 42 Kb were detected in 37 (97.36%) of the 38 samples, corresponding to 11 different profiles (P1- P11), varying from 1 to 6 plasmids per model. The number and plasmids diversity was greater than the resistance marks for strains. The plasmid of 2.85 Kb was the most frequent, being present in 83.78% of the 37 strains; only the one of 7.5 Kb was detected at the two slaughterhouses. There was no parallelism between resistance pattern and plasmidial profile, and a same antibiotype was found in several plasmidial profiles. The results of the present investigation, allowed us to conclude that the plasmid characterization is an useful and simple tool for the epidemiological typing of this sorovar.

Samples of feces and lymph nodes of 300 animals were collected in three different slaughterhouses, and submitted to bacteriological analysis. The prevalence of Salmonella carrier animals was 55.66%, being 17.6% of the animals Salmonella positive in lymph nodes, 18.3% in feces and 19.6% in both materials. Twenty-six different serovars were identified among 226 Salmonella isolates. The most prevalent serovars were: Typhimurium (24.3%), Agona (19.9%), Derby (13.2%) e Bredeney (12%). These results point out the need of control programs to reduce the prevalence of carrier pigs at slaughter.

E-mail: elporti@fcv.unlp.edu.ar Swine eperythrozoonosis is a haemotrophic disease caused by Eperythrozoon suis, actually called Mycoplasma suis, an extracellular bacterial organism that apparently adheres to pig erythrocyte membrane, inducing its deformation and damage. Since little is known about the ultrastructural and morphometrical aspects of this microorganism, the present work aimed to deal with these issues. The ultrastructural study revealed the presence of structures corresponding to tubules disseminated throughout the soma of M. suis. A variable separation between the microorganism membrane and that of the erythrocyte was also observed. The structural and positional attitude of M. suis could allow speculation about its mechanism of action.

A caracterização antigênica consistiu na sorotipagem capsular e determinação de variabilidade do gene de proteína de membrana externa (ompH) pela reação em cadeia da polimerase (PCR) e digestão com cinco enzimas de restrição. Das 22 cepas, 21 foram compatíveis com sorotipo capsular A e uma com D. A caracterização do gene ompH agrupou os isolados em sete padrões distintos que apresentaram boa correlação com os testes bioquímicos.

#167 - Occurrence of Salmonella sp in finishing pigs in Rio Grande do Sul, Brazil, 22(3):104-108

[Occurrence of Salmonella sp in finishing pigs in Rio Grande do Sul, Brazil.] Ocorrência de Salmonella sp em suínos de terminação no Rio Grande do Sul. Pesquisa Veterinária Brasileira 22(3):104-108. Depto Medicina Veterinária Preventiva, Faculdade de Veterinária, UFRGS, Av. Bento Gonçalves 9090, Porto Alegre, RS 91540-000, Brazil. Pen feces samples were taken in ten finishing farms in Rio Grande do Sul (Brazil) and submitted to Salmonella isolation protocol. Two farms negative and one positive in the preliminary screening were chosen, and feces from 25 randomly selected pigs in each of them were individually collected. The sarne animals were later sampled (rectal swab, intestinal content and mesenteric lymph nodes) at the slaughterhouse. After the introduction of new animals into farm Nl and Pl, other 25 pigs on each farm were examined as described above. In the preliminary screening, Salmonella was isolated from pen feces samples of 3 of the 10 investigated farms. Two of these herds showed a high level contamination. Eight Salmonella serotypes (Agona, Bredeney, Lexington, London, Mbandaka, Panama, Schwartzengrund, Salmonella sp) were found, with serotypes Agona and Bredeney being the most frequent. When individual animals were sampled, Salmonella was isolated in all selected farms. Salmonella was isolated in 6.4% of feces collected on the farm, 5.3% of intestinal contents and 5.6% of lymph nodes. Antibiogram testing of the isolated strains showed 97.8% resistance to sulphonamides, 82.6% to streptomycin, 36.9% to tetracyclin and 15.2% to sulfazotrim.

Foram colhidas amostras de fezes de lote de suínos em 10 granjas terminadoras do Estado do Rio Grande do Sul para pesquisa de Salmonella sp. Duas granjas consideradas negativas (Nl e N2) e uma positiva (Pl) nesta primeira etapa foram escolhidas para colheita de amostras de fezes individuais de 25 animais escolhidos aleatoriamente. No abatedouro foram coletados swab retal, conteúdo intestinal e linfonodos mesentéricos dos mesmos animais amostrados na granja. Após a introdução de novos animais nas granjas Nl e Pl, outros 25 animais foram amostrados em cada granja, da mesma forma descrita acima. Três granjas tiveram amostras de fezes de lote positivas, sendo que em duas foi constatado um alto nível de contaminação. Foram encontrados 8 sorotipos de Salmonella (Agona, Bredeney, Lexington, London, Mbandaka, Panama, Schwartzengrund, Salmonella sp), sendo os sorotipos Agona e Bredeney os mais encontrados. Na colheita individual realizada, todas as granjas amostradas foram positivas. Em 6,4% das amostras de fezes colhidas na granja, 5,3% das amostras de conteúdo intestinal e 5,6% dos linfonodos mesentéricos foi possível isolar Salmonella. O antibiograma das linhagens de Salmonella isoladas demonstrou 97,8% de resistência à sulfonamida, 82,6% à estreptomicina, 36,9% à tetraciclina e 15,2% à sulfazotrim.

[Desenvolvimento e padronização de um ELISA indireto para o diagnóstico sorológico de peste suína clássica.] Pesquisa Veterinária Brasileira, 19(3/4):123-127. Equipe de Virologia, FEPAGRO- Centro de Pesquisas Veterinárias Desidério Finamor, Caixa Postal 2076, Porto Alegre, RS 90001-970, Brazil. An indirect enzyme linked immunoassay (ELISA-I) was developed and standardized for the serological diagnosis of classical swine fever (CSF). For the comparison, nine hundred and thirty-seven swine serum samples were tested by serum neutralization followed by immunoperoxidase staining (NPLA), considered ·as the standard. Of these, 223 were positive and 714 negative for neutralizing antibodies to classical swine fever virus (CSFV). In relation to the NPLA, the ELISA-I presented a 98.2% sensitivity; 92.86% specificity, 81.11% positive predictive value, 99.4% negative predictive value and a 94.1% precision. Statistical analysis showed a very strong correlation (r=0,94) between both tests. When compared to a commercially available ELISA kit, the performance of both, in relation to the NPLA, was simila1: It was concluded that the ELISA-I is suitable for large scale screening of antibodies to classical swine fever virus, although it does not distinguish antibodies to classical swine fever vírus from those induced by other pestiviruses.

Equipe de Virologia, FEPAGRO- Centro de Pesquisas Veterinárias Desidério Finamor, Caixa Postal 2076, Porto Alegre, RS 90001-970, Brazil. Um ensaio imunoenzimático do tipo ELISA indireto (ELISA-I) foi desenvolvido e padronizado para o diagnóstico sorológico de peste suína clássica. Na comparação foram utilizadas novecentas e trinta e sete amostras de soros suínos, as quais foram testadas pelo teste de soroneutralização seguido de revelação por imunoperoxidase (NPLA), tomado como padrão, resultando em 223 amostras positivas e 714 negativas. Em relação ao NPLA, o ELISA-I apresentou sensibilidade de 98,21%, especificidade de 92,86%, valor preditivo positivo de 81, 11%, valor preditivo negativo de 99,4% e precisão de 94, 1 %. A análise estatística dos resultados revelou uma correlação muito forte (r=0,94) entre os dois testes. Quando comparado com um "kit" de ELISA disponível comercialmente, a performance de ambos em relação ao NPLA foi similar. Concluiu-se que o ELISA-I é um teste apropriado para triagem em larga escala de soros para a detecção de anticorpos contra o Vírus da Peste Suína Clássica (VPSC), embora não seja capaz de diferenciar entre anticorpos induzidos pelo VPSC ou outros pestivírus.

The main limitation in the use of rock phosphate in swine diets is the fluorine contente that may be toxic when used for long periods. The objective of this study was to evaluate the effects of monocalcium phosphate (MP) and Tapira phosphate (TP) to sows and their offspring when fed during four reproductive cycles, as compared to the traditional use of dicalcium phosphate (DP). The study involved 13, 16 and 13 sows for DP, MP and TP phosphates, respectively. The litters were also fed with the sarne respective phosphate treatments until they reached 95 kg of body weight. The variables analyzed were: macroscopic tooth lesions score in sows, histological evaluation of bone tissue, the thyroid and parathyroid glands, and the evaluation of bone mineral contents and bone morphometrical characteristics. The results showed that TP is toxic for sows when used continuously during 4 reproductive cycles and causes lesions of chronic fluorosis in the teeth and bone tissue. TP also affected the litters with some minor lesions of fluorosis in bone tissue at the 4th reproductive cycle. MP induced some minor inespecific lesions only in bone tissue of the sows. No deleterious effects were observed in sows and litters fed DP.

A limitação principal do uso de fosfatos naturais na alimentação de suínos é a presença de flúor que pode ser tóxico quando ingerido por longos períodos. O objetivo deste trabalho foi avaliar os eventuais efeitos deletérios dos fosfatos Monocálcico (MP) e Tapira (TP), sobre porcas e suas proles alimentadas com esses fosfatos durante quatro ciclos reprodutivos consecutivos, comparativamente com o tradicional fosfato bicálcico (DP). Foram usadas 13 porcas alimentadas com FB (testemunha) 16 com MP e 13 com TP. As leitegadas oriundas de cada ciclo também foram alimentados com os respectivos fosfatos até a idade de abate. As seguintes variáveis foram avaliadas: lesões macroscópicas nos dentes das porcas, alterações histológicas no tecido ósseo e nas glândulas tireóide e paratireóide, conteúdo de minerais no tecido ósseo e características morfométricas do osso. Os resultados obtidos mostraram que o uso de TP por 4 ciclos reprodutivos, na alimentação de suínos, provocou lesões de fluorose crônica nos dentes e tecido ósseo das porcas. As leitegadas alimentadas com este fosfato também apresentaram discretas lesões de fluorose, mas somente evidentes no 4° ciclo reprodutivo. Somente algumas alterações inespecíficas ocorreram nos ossos das porcas que receberam o MP.O DP não provocou alterações patológicas no tecido ósseo, tanto nas porcas como nas suas leitegadas.


#170 - Ultrastructural observation of the airways of recovered and susceptible pigs after inoculation with Mycoplasma hyopneumoniae, 18(1):1-7

To determine the morphological differences in the epithelium of the airways of recovered and susceptible pigs after Mycoplasma hyopneumoniae challenge, twenty-four 4-week-old M. hyopneumoniae-free pigs were intratracheally inoculated with 107ccu/ml of a pure low-passaged culture of the P5722-3 strain of M. hyopneumoniae challenge material. Eight pigs (group I) were challenged at the beginning of the experiment and rechallenged 3 months late 1: Group II pigs were also challenged at the beginning of the experiment and necropsied 3 months later: Group III pigs were challenged at the sarne time as the rechallenge of group I pigs. Eight nonchallenged pigs served as contrais (group IV). Three days after the second challenge of group I and the first challenge of group III, and every 3 and 4 days thereafter, two pigs from each group were euthanatized by electrocution and necropsied. Samples of bronchi and lung tissue were examined using light and electron microscopy (SEM and TEM). Macroscopic lesions were observed in the lungs of all group III pigs (average = 4.74%) and were characterized by purple-red areas of discoloration and increased firmness affecting the cranioventral aspect of the lungs. Macroscopic lesions of pneumonia in groups I and II were minimal (less than 1%). There were no gross lesions of pneumonia in contrai (group IV) pigs. Microscopic lesions were characterized by hyperplasia of the peribronchial lymphoid tissue and mild neutrophilic infiltrates in alveoli. Electron microscopy showed patchy areas with loss of cilia and presence of leukocytes and mycoplasmas in bronchi of susceptible pigs (group III). The bronchial epithelium of rechallenged (group I), recovered (group II), and control (group IV) pigs was ultrastructurally similar indicating recovery of the former two groups. Although mycoplasmas were seen among cilia, a second challenge on pigs of group I did not produce another episode of the disease nor did it enhance morphological changes, suggesting that those pigs could become carriers of M. hyopneumoniae.

Para determinar as diferenças morfológicas no epitélio das vias aéreas de suínos suscetíveis e recuperados da infecção por Mycoplasma hyopneumoniae, 24 leitões com 4 semanas de idade, livres de infecção por M. hyopneumoniae, foram inoculados intratraquealmente com material contendo 107ccu/ml de cultura pura da cepa P5722- 3 de M. hyopneumoniae. Oito leitões (grupo I) foram inoculados no início do experimento e reinoculados 3 meses mais tarde, após apresentarem a doença e terem se recuperado. Os oito leitões do grupo II foram também inoculados no início do experimento e necropsiados 3 meses depois. Os leitões do grupo III foram inoculados ao mesmo tempo da reinoculação dos leitões do grupo I. Oito leitões não inoculados serviram como controles (grupo IV). Três dias após a reinoculação dos leitões do grupo I e a inoculação do grupo III e, a partir disso, a cada 2 dias, dois leitões de cada grupo foram sacrificados e necropsiados. Segmentos de brônquios e pulmão foram examinados em microscopia ótica e microscopia eletrônica de transmissão e de varredura. Lesões macroscópicas foram observadas nos pulmões de todos os leitões do grupo III (média: 4.74% da superfície) e eram caracterizadas por áreas firmes e avermelhadas nas porções cranioventrais. Lesões macroscópicas semelhantes foram mínimas em pulmões de leitões dos grupos I e II (menos de 1%) e ausentes.no grupo IV. As lesões microscópicas consistiam de hiperplasia do tecido linfóide peribronquial e infiltrado neutrofilico leve nos alvéolos. Na microscopia eletrônica havia lesões focais no epitélio ciliado, caracterizadas por perda de cílios e descamação de células, e presença de leucócitos e micoplasmas nos brônquios de leitões suscetíveis (grupo III). O epitélio bronquial de leitões reinoculados (grupo I), recuperados (grupo II) e controles (grupo IV) era ultraestruturalmente similar, indicando recuperação nos primeiros dois grupos. Embora micoplasmas tenham sido observados entre cilios, uma segunda inoculação em leitões do grupo I não causou outro episódio da doença nem aumentou as alterações morfológicas nos brônquios, sugerindo que esses leitões podem se tornar portadores de micoplasmas.
