#1041 - Biópsia hepática no diagnóstico da intoxicação por Senecio brasiliensis (Asteraceae) em bovinos, p.53-60

Laboratory tests of liver function are not always reliable indicators of subclinical affected animals. Liver biopsy could be useful to identify cattle with hepatic lesions but without clinical signs and would have also a prognostic value since it is generally believed that hepatic lesions will eventually cause liver failure and death. Such animals could be picked out by liver biopsy before clinical signs develop and be sent to slaughter, minimizing economic losses. This study was aimed to evaluate the liver biopsy as a diagnostic and prognostic tool in cases of Senecio spp. poisoning in cattle. An outbreak of Senecio brasiliensis was diagnosed in dairy calves which ingested hay contaminated by 5-10% of this Senecio species. Liver biopsy using a Menghini needle by right transthoracic approach was carried out in 135 calves that ingested the contaminated hay. Biopsed calves were followed up for 26 months after the biopsy. Seventeen biopsied calves had typical lesions of Senecio spp. poisoning (positive calves) and 118 had histologically normal livers (negative calves). Hepatic lesions of positive calves included fibrosis, hepatomeglocytosis, and biliary hyperplasia. Fifteen out of the 17 positive calves died with typical clinical signs of Senecio spp. poisoning within 17-149 days after the biopsy; 13 of those were necropsied and had typical gross and histopathological lesions of Senecio spp. poisoning. Two positive calves were clinically normal at the end of the post-biopsy observation period. The prognostic value (sensibility) of the test was considered high since 88.23% of the positive calves died. The specificity of the test was considered very high (99.16%) since only one of the 118 negative calves died in the observation period. In none of the biopsed calves a negative effect related to the biopsy technique was observed.

The isolates were grouped as BVDV-1 (11/19), BVDV-2 (6/19) or “atypical” pestivirus (2/19). Among the BVDV-1, eight isolates were classified as subgenotype BVDV-1a, whereas most (4 out of 6) BVDV-2 belonged to subgenotype 2b. #1043 - Tristeza parasitária bovina na região sul do Rio Grande do Sul: estudo retrospectivo de 1978-2005

E-mail: alschild@terra.com.br A retrospective study of tick fever was made, which occurred from 1978-2005 in southern Rio Grande do Sul in the influence area of the Regional Diagnostic Laboratory of the Federal University of Pelotas. From 4,884 cattle specimens, sent by practitioners or which were from necropsies performed at the Diagnostic Laboratory, 231 (4.7%) were diagnosed as tick fever. Data from 221 of those outbreaks were analyzed. Ninety one (41.1%) outbreaks were caused by Babesia bovis, 11 (4.9%) by Babesia bigemina, and 65 (29.41%) by Anaplasma marginale. In other 33 (14.93%) outbreaks of babesiosis there is no information if the disease was caused by B. bovis or B. bigemina, and 21 (9.5%) outbreaks were caused by mixed infection of A. marginale and B. bovis or B. bigemina. Mean morbidity, mortality, and letality rates in 149 outbreaks were 11.17%, 6.81%, and 70.04%, respectively. Most outbreaks occurred during summer (January-March) and autumn (April-June), mainly in 1 to 3-year-old cattle. Clinical signs were depression, weakness, fallen ears, fever, and weight loss. Low packed cell volume values were always found. Hemoglobinury was observed in babesiosis. Neurological signs characterized by gait alterations, muscular tremors, aggressiveness and falling down with tonic and clonic convulsions were observed in babesiosis by B. bovis. The main gross lesions were anemia, jaundice, splenomegaly, hepatomegaly, yellow liver and cardiac hemorrhages. Hemoglobinury was observed in babesiosis, and congestion of the cerebral cortex in babesiosis by B. bovis. It is concluded that B. bovis is the main agent causing thick fever in southern Rio Grande do Sul. In that region with a cattle population of 2,630,000 heads the annual losses due to tick fever can be estimated in 6,220 cattle or US$ 1,623,000.00. Preventive measures to diminish tick fever losses in the region are necessary.

E-mail franklin.riet@pesquisador.cnpq.br Spongy degeneration (status spongiosus) includes many histological alterations characterized by vacuoles in the neuropile in different regions of the central nervous system (CNS). The vacuolization can be caused by intramyelinic edema, as in maple syrup urine disease and some toxic diseases, or by astrocytic edema, as in citrullinemia. This paper reports a spongy degeneration in two Sindhi calves, a female and a male. Both calves were offspring from the same bull, and the herd had high degree of inbreeding. The female calf was born normal, but progressive nervous signs were observed since 2 months old. The male calf had progressive nervous signs since it was born. Both calves were euthanized with severe nervous signs when 4 months old. At necropsy the liver was pale in the male calf. Histological alterations were mild to accentuated vacuolization, mainly in the deep cerebral cortex, cerebellar white matter, grey matter of the spinal cord, internal capsule, and brain stem. Fatty degeneration was observed in the liver of both animals. On electron microscopy it was found that the status spongiosus was caused by astrocytic edema. It is suggested that the disease is caused by a metabolic hereditary error.

E-mail: josiseixas@ufrrj.br Clinic-pathological aspects and the pathogenesis of experimental abamectin poisoning were studied, after subcutaneous administration of different abamectin doses in 9 calves, as well as the clini-cal and pathological aspects of 74 cases of the iatrogenic poisoning with this drug in cattle, which occurred in the states of Rio Grande do Sul, Pará, Maranhão, Paraíba and Mato Grosso do Sul. From the 9 calves submitted to experimental administration of single doses, 5 calves died (4 calves received doses 6-10 times higher than recommended, and one received the therapeutic daily dose during 11 days). Abamectin poisoning induces neurological dysfunctions, characterized by an initial phase of hyperexcitability, followed by widespread muscular hypotony and progressive depression. No macroscopic or microscopic alterations were observed in the central nervous system or in any other organ. It is concluded that abamectin is an antihelmintic which should be used with restriction, because of the risks leading to death when used in young calves, even in therapeutic doses.

At necropsy of a horse that died with clinical signs of the disease, the main alterations were characterized by hemorrhagic areas of the mucosa of the large intestine with liquid contents, and congested and hemorrhagic areas alternating with normal areas as well as nodules with purulent secretion in the mucosa of the small intestine. The histological changes were characterized by moderate mononuclear infiltration of the mucosa, characterizing a lympho-histiocytic enteritis with predominance of macrophages. In the jejunum and ileum, related to the invagination of crypts and villi, a granulomatous enteritis was observed. To confirm the diagnosis of infection by Neorickettsia (Ehrlichia) risticii, blood samples of seven affected horses were taken and a polimerase chain reaction (PCR) was performed. Three tested blood samples were positive (42.8%) with observation of a band of 529pb characteristic of N. risticii.

At necropsy of a horse that died with clinical signs of the disease, the main alterations were characterized by hemorrhagic areas of the mucosa of the large intestine with liquid contents, and congested and hemorrhagic areas alternating with normal areas as well as nodules with purulent secretion in the mucosa of the small intestine. The histological changes were characterized by moderate mononuclear infiltration of the mucosa, characterizing a lympho-histiocytic enteritis with predominance of macrophages. In the jejunum and ileum, related to the invagination of crypts and villi, a granulomatous enteritis was observed. To confirm the diagnosis of infection by Neorickettsia (Ehrlichia) risticii, blood samples of seven affected horses were taken and a polimerase chain reaction (PCR) was performed. Three tested blood samples were positive (42.8%) with observation of a band of 529pb characteristic of N. risticii.

Deaths are very often the consequence of incorrect or late diagnosis when animals no longer respond to treatment. This study aimed standardization of the ELISA assay for the serodiagnostic of pythiosis in horses and rabbits, in order to minimize errors and delays in the diagnosis of the disease. Sera of 72 healthy and 44 of by pythiosis affected horses were used for development and evaluation of the test. The ELISA for equine diagnostic showed 97.72% sensitivity, 90.27% specificity, 86% positive predictive value, 98.4% negative predictive value, and 93.1% efficiency. The rabbit test was standardized with 48 sera of healthy rabbits and 24 sera of rabbits immunized with P. insidiosum antigens. The results were 91.66 % sensitivity, 95.83% specificity, 91.66% positive predictive value, 95.83% negative predictive value, and 94.44% efficiency. It can be concluded that ELISA is a reliable test for diagnostic and serological monitoring of pythiosis.

Serum antibody levels were determined by ELISA in cows before and after calving, and in goats at 3, 80, 120 and 160 days of age. No significant difference in serum antibody levels was observed between vaccinated and unvaccinated cows, or between the four groups of goats evaluated at 3 days of life. Groups 3 and 4 presented mean antibody titers of 0.6 and 1.1 IU/ml, respectively, 40 days after first vaccination. The vaccine response of Group 4 was 1.8 IU/ml 40 days after the booster dose and was higher than that observed for Group 3 (0.2 IU/ml). Thus, in the proposed regimen the use of heterologous colostrum did not induce passive immunization in goat kids. However, first vaccination and a booster dose after 40 days triggered satisfactory antibody levels.

Fifty symptomatic mixed breed dogs from a region of high incidence of visceral leishmaniasis in Brazil were examined. The results showed that in the positive animals, diagnosed by cytological examination, the ELISA using protein A assay system (mean optical density ± SD / 2.078 ± 0.631) detected more antibodies than the anti-IgG assay (mean optical density ± SD / 1.008 ± 0.437), while in the negative animals, the results by both systems were similar. These results suggest that the ELISA assay using protein A peroxidase conjugated could be useful to detect early infected animals in endemic areas, and thus help to control the spread of the infection.
