#1 - Herbaspirillum seropedicae as a degrading bacterium of monofluoroacetate: effects of its inoculation in goats by ingesting Amorimia septentrionalis and the concentrations of this compound in plants sprayed with the bacterium

Abstract in English:

Herbaspirillum seropedicae is a nitrogen-fixing bacterium capable of using toxic compounds as a source of carbon. Bacteria with this capacity can be used to make animals resistant to plant poisoning containing monofluoroacetate (MFA), such as Amorimia septentrionalis. The aim of this study was to evaluate if H. seropedicae is efficient in the degradation of MFA present in A. septentrionalis and if the inoculation of this bacterium in goats confers protection to A. septentrionalis intoxication. Two experiments were performed: in the first experiment 12 goats were divided into 2 groups. Goats in Group 1 were orally administered a solution containing the H. seropedicae bacterium for 10 days. From day 10 onwards, they received a daily dose of 5g/kg of A. septentrionalis with the bacteriauntil clinical signs of intoxication were observed. Group 2 goats received only the plant at the same dose, also until the observation of clinical signs of intoxication. The amount of MFA found in A. septentrionalis used in the experiment with goats was 1.6±0.058μg/mg. The total plant dose ingested by all goats in Group 1 was 80.83±12.81g/kg (129.33±20.50mg/kg MFA), which were significantly greater ​​(p<0.05) than those of Group 2 goats (39.16±19.08g/kg plant and 62.66±30.53mg/kg MFA). Group 1 goats took an average of 16.16±2.56 days to develop clinical signs of intoxication, significantly longer (p=0.0012) than Group 2 goats (7.83±3.81 days). Two Group 2 goats died on the same day that they developed clinical signs of intoxication. At necropsy of these two animals, no significant changes were observed. In the second experiment, samples of A. septentrionalis were sprayed with a solution containing H. seropedicae. Before and eight days after spraying, the samples were pressed and dried for quantitation of MFA. The amount of MFA present in samples of A. septentrionalis 8 days after spraying with H. seropedicae was significantly lower (p=0.017) than that found prior to spraying. It can be concluded that administration of H. seropedicae in goats is capable of causing greater resistance to A. septentrionalis intoxication, and spraying the plant with this bacterium significantly reduces the amount of MFA in the plant.

Herbaspirillum seropedicae é uma bactéria fixadora de nitrogênio, capaz de utilizar compostos tóxicos como fonte de carbono. Bactérias com essa capacidade podem ser utilizadas para tornar os animais resistentes à intoxicação por plantas que contém monofluoroacetato (MFA), como Amorimia septentrionalis. O objetivo do presente estudo é avaliar se H. seropedicae é eficiente na degradação do MFA presente em A. septentrionalis e se a inoculação dessa bactéria, em caprinos, confere proteção à intoxicação por A. septentrionalis. Foram realizados dois experimentos: no primeiro experimento foram utilizados 12 caprinos, divididos em dois grupos. Os caprinos do Grupo 1 receberam diariamente, oralmente, uma solução contendo a bactéria H. seropedicae durante 10 dias. A partir do décimo dia passaram a receber, diariamente, além da solução com a bactéria 5g/kg de A. septentrionalis até a observação de sinal clínico de intoxicação. Os caprinos do Grupo 2 receberam apenas a planta na mesma dose, também até que a observação de sinais clínicos de intoxicação. A quantidade de MFA encontrada em A. septentrionalis utilizada no experimento com caprinos foi de 1,6± 0,058µg/mg de planta em média. A dose total de planta ingerida por todos os caprinos do Grupo 1 foi de 80,83±12,81g/kg (129,33±20,50mg/kg de MFA), valores significativamente maiores (p<0,05) do que os dos caprinos do Grupo 2 (39,16±19,08g/kg de planta e 62,66± 30,53mg/Kg de MFA). Os caprinos do Grupo 1 demoraram em média 16,16 ±2,56 dias para desenvolver sinais clínicos da intoxicação, período significativamente maior (p=0,0012) que os caprinos do Grupo 2 (7,83±3,81dias). Dois caprinos do Grupo 2 morreram no mesmo dia que desenvolveram sinais clínicos da intoxicação. Na necropsia desses dois animais não foram observadas alterações significativas. No segundo experimento, amostras de A. septentrionalis foram pulverizadas com uma solução contendo a bactéria H. seropedicae. Antes e oito dias após a pulverização, as amostras foram prensadas e secas para posterior quantificação do MFA. A quantidade de MFA presente nas amostras de A. septentrionalis oito dias após a pulverização com H. seropedicae foi significativamente menor (p=0,017) do que a encontrada antes da pulverização. Pode-se concluir que a administração de H. seropedicae em caprinos é capaz de causar uma maior resistência à intoxicação por A. septentrionalis, e a pulverização da planta com esta bactéria reduz significativamente a quantidade de MFA na planta.

E-mail: grasivet@hotmail.com Sporotrichosis is a subcutaneous mycosis caused by a dimorphic fungus from Sporothrix schenckii complex, which affects various species, especially feline, able to cause local, disseminated or systemic impairment. We aimed to report the occurrence of an outbreak of feline sporotrichosis in the Metropolitan Region of Recife. Swabs of skin ulcers in cats treated in the Veterinary Hospital of the Federal Rural University of Pernambuco and in particular veterinary establishments were collected. Diagnosis was made by cytological examination and fungal culture. From 115 suspicious cases, 59 cases were confirmed, unprecedented results in the state of Pernambuco. These registered cases of feline sporotrichosis have demonstrated clinical and epidemiological characteristics similar to those occurring in other regions of the country, characterized as a quick-spreading disease, of difficult treatment, which affects predominantly semi-domiciled young males in reproductive age.

E-mail: grasivet@hotmail.com A esporotricose é uma micose subcutânea, causada por um fungo dimórfico do complexo Sporothrix schenckii que acomete várias espécies, especialmente a felina, causando comprometimento local, disseminado ou sistêmico. Objetivou-se relatar a ocorrência de um surto de esporotricose felina na Região Metropolitana do Recife. Foram coletados suabes de úlceras cutâneas de felinos atendidos no Hospital Veterinário da Universidade Federal Rural de Pernambuco e em Clínicas Veterinárias particulares. O diagnóstico foi realizado por meio de exame citológico e cultura fúngica. Dos 115 casos suspeitos, 59 casos foram confirmados, número nunca registrado anteriormente no estado de Pernambuco. Os casos de esporotricose felina registrados têm demonstrado aspectos clínicos e epidemiológicos semelhantes aos ocorridos em outras regiões do país, caracterizando-se como uma doença predominantemente de machos jovens em idade reprodutiva e semi-domiciliados, de difícil tratamento e de rápida disseminação.

E-mail: danipessoavet@gmail.com In Brazil is estimated that poisoning of livestock by sodium monofluoroacetate (MFA) containing plants causes the death of about 500.000 cattle per year. The ruminal inoculation of bacteria that degrade MFA has been proposed as a way to prevent the poisoning. This study aimed to evaluate in goats resistance to the MFA-containing plant Amorimia septentrionalis induced by ruminal inoculation of the bacteria Pigmentiphaga kullae and Ancylobacter dichloromethanicus. Twelve goats, without previous contact with MFA-containing plants, were divided into two groups of six animals each. In group 1, 60ml of a mixture of the two bacteria was inoculated every day for 10 days into each goat. In group 2, the goats did not receive the bacteria. At the 10th day of inoculation, A. septentrionalis began to be administered daily at a dose of 5g/kg body weight to both groups. The administration was interrupted in each goat after first clinical signs of poisoning were observed.. The goats of group 1 showed clinical signs 5.83±2.56 days after the administration of the plant, what differed significantly (p=0.037) from goats of group 2, that showed clinical signs 2.67±0 52 days after the beginning of ingestion. The amount of A. septentrionalis ingested by inoculated goats (28.83±12.97g/kg) to cause clinical sings was significantly greater (p=0.025) than the amount ingested by the non-inoculated (12.03±3.65) goats to cause clinical signs and was also statistically different between the groups. We concluded that the intraruminal administration of Pigmentiphaga kullae and Ancylobacter dichloromethanicus increases the resistance to poisoning by MFA-containing plants.

A partir do 10º dia de inoculação, A. septentrionalis foi administrada, diariamente, na dose de 5g/kg de peso vivo, sendo interrompida em cada animal após a observação dos primeiros sinais clínicos da intoxicação. Os caprinos do grupo 1 apresentaram sinais clínicos 5,83±2,56 dias após a administração da planta o que diferiu significativamente (p=0,037) dos caprinos do grupo 2, que apresentaram sinais clínicos aos 2,67±0,52 dias. A quantidade de planta ingerida pelos caprinos inoculados (28,83±12,97g/kg) e os não inoculados (12,03±3,65g/kg) para desencadear os sinais clínicos foi, também, estatisticamente diferente entre os grupos (p=0,025). Conclui-se que a administração intraruminal de Pigmentiphaga kullae e Ancylobacter dichloromethanicus induz resistência à intoxicação por plantas que contêm MFA.

E-mail: mateus.costa@univasf.edu.br The objective of this study was to evaluate the effect of Ascophyllum nodosum brown seaweed meal (FAM) on the health of Nile tilapia submitted to inoculation with Aeromonas hydrophila. The experiment was conducted for a period of 40 days using 120 Nile tilapia fingerlings, with age of 40 days, distributed in 20 tanks. A diet including Ascophyllum nodosum seaweed meal at 20g.kg-1 and a control diet (without FAM) were provided which constituted the treatments. Thirty days after beginning the experiment, A. hydrophila was inoculated by bacterial inoculum diluted in sterile saline solution at a concentration of 106 CFU ml-1. Except for the width, which was greater for the treatment with the provision of FAM (P<0.05), there was no influence on the performance parameters of the fingerlings, but the occurrence of lesions in animals inoculated with A. hydrophila and fed with FAM was lower and they also exhibited a decline in the lesions in a shorter period of time than the control group. FAM prevents hepatopancreatic congestion in infected animals. Ascophyllum nodosum brown seaweed meal reduced the number of lesions in fish in a shorter time when compared to the control group.

Foi fornecida uma ração com inclusão da farinha da alga marinha Ascophyllum nodosum a 20g.kg ração-1 e uma ração testemunha (sem FAM), que constituiram os tratamentos. Após 30 dias experimentais foi realizada a inoculação de Aeromonas hydrophila por meio de um preparado de inóculo bacteriano com diluição em solução salina estéril a concentração de 106 UFC/ml. Exceto para a largura, que foi maior para o tratamento com o fornecimento da FAM (P<0,05), não houve influência sobre parâmetros de desempenho dos alevinos, mas a ocorrência de lesões nos animais inoculados com A. hydrophila e alimentados com FAM foi menor e, eles também exibiram um declínio das lesões em um menor período de tempo em relação ao grupo controle. FAM impediu a congestão hepatopancreática em animais infectados. A farinha de alga marinha marrom Ascophyllum nodosum reduziu o número de lesões nos peixes em menor tempo quando comparado ao controle.

Following replication in the nasal mucosa, viral invasion of the brain is thought to occur mainly by the olfactory pathway. To address the role of this pathway in the pathogenesis of neurological infection in a laboratory model, 30 days old rabbits had the main olfactory bulbs (MOBs) surgically removed and were subsequently inoculated intranasally (IN) or conjunctivally (IC) with a highly neurovirulent BoHV-5 strain (SV-507). Following IN inoculation, 10 out of 10 (100 %) control rabbits developed neurological disease. The clinical onset ranged from day 5 to 10 post-inoculation (pi, average 7.5 days); nine being euthanized in extremis and one recovering after a mild clinical course. In contrast, only one rabbit (9.1 %) of the group lacking the MOBs (n=11) developed neurological disease (onset at day 17 pi). Dexamethasone administration to the survivors (n=10) at day 50pi was followed by virus shedding in nasal and/or ocular secretions by 8 animals, demonstrating that the virus was able to reach the trigeminal ganglia (TG) during acute infection. These results demonstrate that the olfactory route provides the main, yet not the sole access to the brain of rabbits following IN inoculation. To address the role of a second pathway, groups of control (n=12) or MOB-lacking rabbits (n=12) were inoculated into the conjunctival sac (IC), following which the virus would be expected to use the ophtalmic branch of the trigeminal nerve to reach the brain. Ten control rabbits (83.3 %) developed neurological disease upon IC inoculation (onset 15.3 days [11 to 20]). Previous ablation of the MOBs did not affect the frequency and course of neurological disease: ten out of 12 rabbits (83.3 %) lacking the MOBs developed neurological disease (onset 9 to 15 dpi, average: 12.7 days) upon IC inoculation. These results demonstrate that both IN and IC routes may operate in the transport of BoHV-5 to the brain of experimentally infected rabbits, depending on the route of inoculation. IN inoculation results in a fast and efficient transport by the olfactory pathway, the trigeminal route providing an alternative, much slower and less efficient transport; IC inoculation results in efficient viral transport by the trigeminal route, yet with a delayed kinetics comparing to the transport provided by the olfactory pathway.

#6 - Ultrastructural observation of the airways of recovered and susceptible pigs after inoculation with Mycoplasma hyopneumoniae, 18(1):1-7

To determine the morphological differences in the epithelium of the airways of recovered and susceptible pigs after Mycoplasma hyopneumoniae challenge, twenty-four 4-week-old M. hyopneumoniae-free pigs were intratracheally inoculated with 107ccu/ml of a pure low-passaged culture of the P5722-3 strain of M. hyopneumoniae challenge material. Eight pigs (group I) were challenged at the beginning of the experiment and rechallenged 3 months late 1: Group II pigs were also challenged at the beginning of the experiment and necropsied 3 months later: Group III pigs were challenged at the sarne time as the rechallenge of group I pigs. Eight nonchallenged pigs served as contrais (group IV). Three days after the second challenge of group I and the first challenge of group III, and every 3 and 4 days thereafter, two pigs from each group were euthanatized by electrocution and necropsied. Samples of bronchi and lung tissue were examined using light and electron microscopy (SEM and TEM). Macroscopic lesions were observed in the lungs of all group III pigs (average = 4.74%) and were characterized by purple-red areas of discoloration and increased firmness affecting the cranioventral aspect of the lungs. Macroscopic lesions of pneumonia in groups I and II were minimal (less than 1%). There were no gross lesions of pneumonia in contrai (group IV) pigs. Microscopic lesions were characterized by hyperplasia of the peribronchial lymphoid tissue and mild neutrophilic infiltrates in alveoli. Electron microscopy showed patchy areas with loss of cilia and presence of leukocytes and mycoplasmas in bronchi of susceptible pigs (group III). The bronchial epithelium of rechallenged (group I), recovered (group II), and control (group IV) pigs was ultrastructurally similar indicating recovery of the former two groups. Although mycoplasmas were seen among cilia, a second challenge on pigs of group I did not produce another episode of the disease nor did it enhance morphological changes, suggesting that those pigs could become carriers of M. hyopneumoniae.

RESUMO.- Irigoyen L.F., Van Alstine W, Turek j. & Clark L.K. 1998. Ultrastructural observation of the airways of recovered and susceptible pigs after inoculation with Mycoplasma hyopneumoniae. [Observação ultra-estrutural das vias aéreas de suínos recuperados e suscetíveis após inoculação com Mycoplasma hyopneumoniae.] Pesquisa Veterinária Brasileira 18(1):1-7. Depto Patologia, Universidade Federal de Santa Maria, Campus Camobi, Santa Maria, RS 97119-900, Brazil. Para determinar as diferenças morfológicas no epitélio das vias aéreas de suínos suscetíveis e recuperados da infecção por Mycoplasma hyopneumoniae, 24 leitões com 4 semanas de idade, livres de infecção por M. hyopneumoniae, foram inoculados intratraquealmente com material contendo 107ccu/ml de cultura pura da cepa P5722- 3 de M. hyopneumoniae. Oito leitões (grupo I) foram inoculados no início do experimento e reinoculados 3 meses mais tarde, após apresentarem a doença e terem se recuperado. Os oito leitões do grupo II foram também inoculados no início do experimento e necropsiados 3 meses depois. Os leitões do grupo III foram inoculados ao mesmo tempo da reinoculação dos leitões do grupo I. Oito leitões não inoculados serviram como controles (grupo IV). Três dias após a reinoculação dos leitões do grupo I e a inoculação do grupo III e, a partir disso, a cada 2 dias, dois leitões de cada grupo foram sacrificados e necropsiados. Segmentos de brônquios e pulmão foram examinados em microscopia ótica e microscopia eletrônica de transmissão e de varredura. Lesões macroscópicas foram observadas nos pulmões de todos os leitões do grupo III (média: 4.74% da superfície) e eram caracterizadas por áreas firmes e avermelhadas nas porções cranioventrais. Lesões macroscópicas semelhantes foram mínimas em pulmões de leitões dos grupos I e II (menos de 1%) e ausentes.no grupo IV. As lesões microscópicas consistiam de hiperplasia do tecido linfóide peribronquial e infiltrado neutrofilico leve nos alvéolos. Na microscopia eletrônica havia lesões focais no epitélio ciliado, caracterizadas por perda de cílios e descamação de células, e presença de leucócitos e micoplasmas nos brônquios de leitões suscetíveis (grupo III). O epitélio bronquial de leitões reinoculados (grupo I), recuperados (grupo II) e controles (grupo IV) era ultraestruturalmente similar, indicando recuperação nos primeiros dois grupos. Embora micoplasmas tenham sido observados entre cilios, uma segunda inoculação em leitões do grupo I não causou outro episódio da doença nem aumentou as alterações morfológicas nos brônquios, sugerindo que esses leitões podem se tornar portadores de micoplasmas.
